The degree of development of rigor mortis in the carcases of slaughter pigs was assessed subjectively on a three-point scale 35 minutes after they were exsanguinated, and related to the levels of cortisol, lactate and creatine kinase in blood collected at exsanguination. Earlier rigor development was associated with higher concentrations of these stress indicators in the blood. This relationship suggests that the mean rigor score, and the frequency distribution of carcases that had or had not entered rigor, could be used as an index of the degree of stress to which the pigs had been subjected.

The loading and unloading of cattle for road transport is stressful but the stress is difficult to evaluate in terms of the welfare of the cattle. Over one year, 40 normal commercial journeys in northern Spain were analysed in terms of time limits and behavioural events in order to design an objective method for assessing the stresses imposed on the animals, and a scoring method was developed for assessing the welfare of the animals during loading and unloading. Several definitions of time intervals were assessed to calculate a time score per animal, and easily observable behavioural events were scored and combined with the time score to obtain a total loading/unloading score. More than half of the loadings and unloadings involved turns, slips and vocalisations. Mounts and bouts of fighting were infrequent and balks and falls were significantly more frequent during loading than unloading. The plasma concentrations of cortisol, glucose and lactate, the activity of creatine kinase and the pH of the meat 24 hours after the animals were slaughtered were also measured. The results indicated that loading was more stressful than unloading and that higher scores implied significantly higher levels of stress.

Over a period of 10 days, 17 dogs became weak and developed neurological deficits of different degrees of severity. About 12 hours before these clinical signs appeared they had all eaten a particular brand of commercial dog food from a recently opened bag. They were all quadriparetic and hyporeflexic, and some of them also showed additional systemic or neurological signs, including dyspnoea, a high body temperature, tongue laxity, hyperaesthesia and anisochoria. Serum biochemical abnormalities included high activities of creatine kinase, lactate dehydrogenase and aspartate aminotransferase. Analysis of the suspect food revealed high concentrations of the ionophore lasalocid. Fifteen of the dogs were given supportive treatment at home and two were hospitalised. Five of the dogs died, but the others improved gradually and had fully recovered by one to four days after the appearance of the clinical signs.

Blood samples were taken from the ventral coccygeal vein of 15 El Hierro giant lizards (Gallotia simonyi) (seven females and eight males), six La Gomera giant lizards (Gallotia bravoana) (four males and two females) and four Tenerife giant lizards (Gallotia intermedia) (two males and two females), and 31 blood parameters were measured. Among the haematological parameters there were significant differences between the three species in heterophils, azurophils and lymphocytes, but no significant differences in red blood cell count, white blood cell count, haemoglobin, packed-cell volume, monocytes, eosinophils and basophils. In terms of blood chemistry there were significant differences between the three species in cholesterol, triglycerides, glucose, sodium, chloride, urea, uric acid, total proteins, prealbumin, albumin and gamma globulins, but no significant differences in calcium, potassium, aspartate aminotransferase, alanine aminotransferase, creatine kinase, bile acids, alpha-1 and alpha-2 globulins and beta globulins.

Continuous muscle fibre activity was observed in a crossbred dog, a Yorkshire terrier, a border collie and three Jack Russell terriers. The clinical signs consisted of episodes of generalised myokymia which developed into muscle stiffness and delayed muscle relaxation and generally led to the dogs collapsing into lateral recumbency. These episodes were preceded by intense facial rubbing in three of the dogs, and were associated with severe hyperthermia in five of them. All three Jack Russell terriers showed continuous ataxia. The dogs had above normal activities of aspartate aminotransferase, alanine aminotransferase and creatine kinase, but their cerebrospinal fluid was normal. Myokymic discharges were observed by electromyography in two of the dogs. Two of them were treated with membrane-stabilising agents, with variable results.

Blood from 31 healthy, free-ranging golden jackals held in captivity for seven days was collected while they were anaesthetised. Haematological and serum biochemical measurements were analysed and the 95 per cent confidence interval for each variable was compared with the reference value for domestic dogs. The measurements of their red blood cells were within the reference interval for dogs, but the jackals had higher white blood cell counts and eosinophil counts than dogs. The male jackals had a higher haematocrit, red blood cell count, mean corpuscular volume and mean corpuscular haemoglobin concentration, and a lower red blood cell distribution width than the female jackals. High activities of muscle enzymes were detected in many of the jackals, in several of which the activity of creatine kinase exceeded 5000 U/l; these were considered abnormal.

Measurements on 28 pens of pigs containing 384 animals to be slaughtered at a commercial abattoir showed that the mean ear temperatures of the pigs in each pen, measured with a thermal imaging camera, were significantly correlated (r=0.71, P<0.001) with the mean temperature of the blood the pigs lost at exsanguination. In measurements on 220 pigs in 16 of the 28 pens, the mean activity of serum creatine kinase was positively correlated with the mean ear temperature (r=0.55, P<0.05) and the mean concentration of serum cortisol was positively correlated with the mean blood temperature (r=0.50, P<0.05), suggesting that the hotter pigs were suffering from a higher level of stress.

A total of 417 pigs with an estimated average liveweight of 91 kg were transported to slaughter in three journeys, carried out in late June, each consisting of a three-hour drive followed by a one-hour holding period at the slaughter plant before they were unloaded. The pigs were accommodated on the lower two decks of a three-deck articulated lorry, each deck having five equal-sized pens; half were kept in naturally ventilated pens and half in pens with fan-assisted ventilation. The pigs' body temperature was recorded as the temperature of the blood lost at exsanguination, and as the temperature of the inner surface of the ear measured by thermal imaging. The pigs carried on the upper deck were hotter than those carried on the lower deck, and the pigs carried at the front of the vehicle were hotter than those at the back. There were small differences between the temperatures of the pigs in the different pens, but overall the temperatures of the pigs kept in pens with fan-assisted ventilation were no lower than those of the pigs kept in pens with natural ventilation. The higher temperature of the pigs transported in the front of the vehicle was associated with significantly higher serum concentrations of cortisol and activities of creatine kinase, and a tendency to higher albumin concentrations and osmolality, suggesting that they may have been exposed to more physical and psychological stress, and were possibly slightly more dehydrated, than the pigs in the rearmost pens. The type of ventilation had no effect on the blood composition of the pigs.

The clinical findings and treatment of 94 cattle with listeriosis are described. The general behaviour and condition of the animals were mostly moderately to severely disturbed. A common abnormality in posture was an exaggerated forward or sideward stance, and 11 of the animals were recumbent. More than half of the animals were ataxic and 22 circled. The most frequent cranial neurological signs observed were facial nerve paralysis, salivation, strabismus, reduced or absent pupillary light reflex, reduced or absent tongue movement and head tilt. The haematological and biochemical findings did not contribute to the diagnosis of listeriosis, but they were useful indicators of dehydration and the acid-base status of the animal. Forty-four of 57 of the animals had high leucocyte counts in the cerebrospinal fluid (CSF), mostly mononuclear cells. Eighty-seven of the animals were treated with various antibiotics (penicillin G, oxytetracycline, amoxicillin, and amoxicillin and gentamicin combined), but there was no significant difference in the success rate of the different treatments. Only two of the nine recumbent animals that were treated survived. Univariable analysis suggested that animals that were recumbent, excited, with an absent or weak menace reflex, nystagmus, high numbers of leucocytes in the CSF, high serum concentrations of urea and calcium and high serum activities of aspartate aminotransferase and creatine kinase, and an acid-base deficit, had a smaller chance of surviving. When a logistic regression model was constructed, only recumbency, excitement and a weak or absent menace reflex remained significant factors affecting the likelihood of survival.

The aim of the study was to obtain basic data on individual biological variation, the required number of specimens to define the homeostatic setpoint (the aspired value of a variable adjusted to the individuals homeostasis) and critical differences of selected chemistry parameters in budgerigars (Melopsittacus undulatus). Blood from 99 healthy budgerigars was sampled 12 times at four-week intervals. Aspartate aminotransferase (ast), calcium, creatine kinase (ck), glutamate dehydrogenase (gldh), glucose, lactate dehydrogenase (ldh), total protein and uric acid were investigated. The indices of individuality obtained in the present study were relatively low (total protein 0.93, ast 1.02, gldh 1.04, ck 1.12, ldh 1.24, uric acid 1.26, glucose 1.39, calcium 1.61) and suggest that population-based reference limits might be useful. Comparison of data showed that the application of intraindividual reference values identified much greater variation in the reference values than using conventional population-based reference limits. Otherwise, the moderate to low degrees of individuality may allow the use of reference values of one individual as a standard for another individual of the same species. The critical differences that define the change needed between two serial results to indicate a biological change was highest in gldh with 120 per cent or 1.7 U/l. In calcium, a change of 30.5 per cent or 0.5 mmol/l would be significant. In the other parameters critical differences varied between 38 per cent in glucose, up to 93 per cent in uric acid.
