Pharmacological and ultrastructural methods were used to demonstrate alpha-adrenergic regulation of secretory granule content of acinar cells of Bowman's glands and to localize and identify adrenergic and cholinergic axonal varicosities and terminals in the olfactory mucosa of the tiger salamander. The alpha-adrenergic agonist phenylephrine caused secretory granule depletion from Bowman's glands; the alpha-adrenergic antagonist phentolamine partially blocked this effect. These observations were quantified using light microscopic computer-assisted morphometric techniques. Both drugs caused morphological signs of electrolye/water transport. Adrenergic axonal varicosities were identified by the presence of small granular vesicles (SGVs, 45-60 nm in diameter) containing electron-dense material that was enhanced by 5-hydroxydopamine loading and chromaffin reaction fixation techniques. Throughout the lamina propria, small fascicles with axons containing SGVs as well as varicosities and terminals with SGVs were located adjacent to blood vessels, Bowman's gland acini, and melanocytes. Mean vesicle diameters at these sites were 54 +/- 7 nm, 50 +/- 9 nm, and 56 +/- 8 nm, respectively; varicosities were located approximately 0.1-1.0 microns from their presumed cellular targets. Axonal varicosities containing small agranular vesicles (AGVs, 65 +/- 8 nm in diameter), identified as cholinergic by their size and by the absence of electron-dense material after 5-hydroxydopamine loading and chromaffin reaction fixation, were located between adjacent acinar cells. In addition, adrenergic varicosities containing SGVs (56 +/- 6 nm in diameter) were found within 1 micron of blood vessels associated with Bowman's gland ducts and sustentacular cells near the base of the olfactory epithelium. These results characterize the ultrastructural basis for adrenergic and cholinergic regulation of vasomotor tone and secretion within the olfactory mucosa.

The enamel organ of growing rat incisors was perfusion-fixed with a mixture of formaldehyde and glutaraldehyde and processed for ultracytochemical demonstration of ouabain-resistant, K+-stimulated p-nitrophenylphosphatase representing the second dephosphorylative step of H-K-ATPase by use of the one-step lead method. Throughout the stages of amelogenesis, the enzymatic activity was found in the plasma membranes, mitochondrial membranes, and lysosomal structures of the cells of stratum intermedium, papillary layer, and ameloblast layer. Gap junctions and desmosomes between these cells were, however, free of reaction product or showed slight precipitates of reaction. The stellate reticulum and the outer enamel epithelium at the stage of enamel secretion were usually negative for reaction. Although secretory, transition, and ruffle-ended maturation ameloblasts showed enzymatic activity at their basolateral cell surfaces, their distal cell surfaces facing the enamel were always free of reaction product. On the other hand, the smooth-ended maturation ameloblasts seldom showed a positive reaction, except in lysosomes and along their basal cell surfaces. An energy-dispersive X-ray microanalysis of reaction products of H-K-ATPase in unosmicated tissue sections demonstrated that they were composed of lead and phosphorus, which had been released during the dephosphorylation of substrate. In cytochemical controls, the enzymatic activity was completely dependent on substrate and potassium ion, resistant to ouabain and levamisole, and inhibited by nolinium bromide, a specific inhibitor of H-K-ATPase. In addition, inorganic trimetaphosphatase as enzymatic marker of lysosome was localized in dark and pale lysosomes, phagosomes, multivesicular bodies, and ferritin-containing vesicles of the ameloblasts and the cells of stratum intermedium and papillary layer. These membrane-bound structures were also positive for H-K-ATPase reaction. These results suggest that: 1) H-K-ATPase functions to maintain an acidic internal pH of lysosomes in the enamel organ cells; and 2) H-K-ATPase localization in the plasma membranes of enamel organ cells is concerned with efflux of protons derived from cytoplasmic water.

Uncertainty concerning the use, efficacy and possible adverse effects of clostridial vaccination in goats prompted a study of the injection site reactions and antibody responses in 40 goats and 40 sheep. The vaccines used were Covexin 8, Heptavac and Tasvax 8. In all the animals swellings averaging 2.5 cm in diameter were present at the injection site seven days after vaccination and were still apparent 28 days after vaccination. The injection site reactions could not be attributed to faulty vaccination technique because they did not occur in a control group injected with sterile water. By 14 days the reactions were significantly larger in sheep than in goats and by 28 days the reactions to Covexin 8 were larger than those to the other vaccines in sheep and goats. Serum antibody was present in all groups before vaccination and, with the exception of the goats vaccinated with Heptavac, increased 14 days after vaccination. The increase was greater in sheep than in goats. By 28 days antibody levels had declined in all but the sheep vaccinated with Heptavac in which a further increase occurred. At that time, the antibody levels in vaccinated sheep were still higher than in the unvaccinated sheep whereas the antibody levels in vaccinated goats were no longer different from those in the control goats. These results suggest that there is a difference between the vaccines used and between the responses of the two species and support the clinical observation that the protection afforded to goats by multivalent clostridial vaccines is poorer than that afforded to sheep.

The cardiac atria are known to play a role in blood volume homeostasis, secreting a peptide that induces a potent natriuresis and diuresis. This peptide is atrial natriuretic factor (ANF), and its primary site of storage is within atria-specific granules found in atrial cardiocytes. Since salt loading results in an increase in circulating levels of ANF, our aim was to determine if the atria-specific granule population in the cardiocytes of Dahl rats would decrease accordingly. To this end, the fractional volume of the atria-specific granules was determined by ultrastructural morphometric analysis in the Dahl salt model of hypertension. This analysis was performed on the right atria of Dahl Salt-resistant (DR) and salt-sensitive (DS) rats fed either a low-salt (0.4%) or high-salt (8%) diet for 12 weeks prior to sacrifice. DR and DS rats fed a low-salt diet had significantly reduced plasma sodium levels and osmolalities, and a significantly lower mean arterial blood pressure than did rats fed a high-salt diet. The fractional volume of atria-specific granules was significantly lower in salt-loaded DR (P less than 0.01) and DS (P less than 0.025) rats than in their respective low-salt controls. This significant decrease in atrial granules corresponds to the reported decrease in the storage of atrial ANF in salt-loaded rats, and provides a morphological verification of the biochemical studies. Moreover, these results, in combination with a growing body of physiological data, lend support to the hypothesized role of ANF in the regulation of water-electrolyte balance, which may play an important role in cardiovascular pathophysiological states related to hypertension.

Lungworm-infected seeder calves were used on four 1.41 ha paddocks to ensure that groups of 11 calves would be exposed to a heavy challenge with Dictyocaulus viviparus. By the 39th day after turnout there was a serious episode of respiratory disease and a diagnosis of parasitic bronchitis was confirmed by post mortem and faecal examination. One group of trial calves was treated with netobimin administered in the drinking water at 2.8 mg/kg/day for seven consecutive days; another group received the same treatment supplemented with flunixin meglumine at 2.2 mg/kg/day for three days; a third group was given a single oral dose of 7.5 mg netobimin/kg; only emergency treatments were given to calves in the control group. The clinical response to the drinking water treatments was highly satisfactory and better than the response to the single oral treatment.

Pregnant ewes were injected intramuscularly with 300,000 iu of vitamin D3 in a water miscible vehicle either 10, seven or four weeks before the expected lambing date and the effects on plasma concentrations of 25-hydroxyvitamin D3 were monitored. The concentrations increased quickly and remained high at parturition but at no time were they outside the normal physiological range. The concentrations in the plasma of the newborn lambs were higher than in uninjected controls and were well correlated with the concentrations in their mothers. Dosing pregnant ewes with 300,000 iu of vitamin D3 in a rapidly available form, approximately two months before lambing, provided a safe means of increasing the vitamin D status of the ewe and the newborn lamb by preventing the seasonally low concentrations of 25-hydroxyvitamin D3.

Counterimmunoelectrophoresis, direct immunofluorescence and immunodiffusion procedures were used to serotype 15 strains of Haemophilus pleuropneumoniae isolated from the respiratory tract of pigs in southern Brazil. Antigens were prepared by extracting cultures with a saline solution or by the phenol-water method. Antisera were prepared in rabbits against serotypes 1, 2, 3 and 5. Thirteen of the isolates were type 5 and two were type 3. No differences were observed between the results obtained in serotyping with counter immunoelectrophoresis and direct immunodiffusion, but both procedures were significantly better than immunodiffusion except with the saline extracted antigen. Counterimmunoelectrophoresis was quicker, more sensitive and more easily performed than the other techniques.

Under experimental conditions, fenbendazole given at doses of 0.4 and 1.0 mg/kg body weight suppressed calves' faecal output of Ostertagia and Cooperia species eggs and Dictyocaulus viviparus larvae. Both dose levels were given in the form of small daily drenches and the higher level showed greater efficacy. In a grazing experiment, medication with fenbendazole at 1.0 mg/kg/day administered intermittently to calves using an automatic dose dispenser almost completely suppressed the output of trichostrongylid eggs. As a result, infection on the pasture and in the calves remained at a low level throughout the grazing season. By contrast, control pasture and control calves showed rather heavy infection from mid-August onwards with significantly lower weight gains and widespread signs of parasitic gastroenteritis. At post mortem examination of representative calves from each group in November, the medicated animals had 99 per cent less Ostertagia species, whether adults or larvae arrested at the early fourth stage, and 95 per cent less Cooperia species compared with controls. Medication in the drinking water suppressed the faecal output of D viviparus larvae for most of the grazing season by comparison with the controls but the medicated calves became infected with this parasite towards the end of the season. Until this problem is overcome, precautions against parasitic bronchitis are advised when this system of medication is adopted.

The cause of the increased egg weight in hens with lathyrism was investigated by feeding laying birds with diets containing beta-aminopropionitrile, the toxic principle of certain legume seeds in the genus Lathyrus. There was a significant increase (P less than 0.01) in the amount of albumen in the egg owing to an increased formation of thin white. The water permeability of the shell membranes demonstrated an increased porosity and a loss of tensile strength which contributed not only to a high water uptake but also to abnormalities of the shape of eggs during 'plumping'. It was concluded that a defect in the shell membrane protein system alters the membrane's general properties and that the increased egg weight in lathyrism is a result of the defect.

We had examined auditory brainstem responses (ABRs) of boxers in order to study human cerebral concussion. ABRs were not prolonged after bouts or knock-out but were unexpectedly shortened after any exertion with or without impact to the head. It was supposed that hyperthermia induced by exertion shortened brainstem conduction velocity as it occurred in peripheral nerve. We investigated correlation of ABR latency and tympanic membrane temperature, which indicated hypothalamic temperature or central nervous temperature, during exertion and body heating. I. Exertion group Six young males aged from 21 to 24 years old were examined (the two were examined twice). T ty (tympanic membrane temperature) and T rec (rectal temperature) were monitored. ABRs and carotid blood flow were examined (QFM). Then subjects were covered with PVC clothes and exercised on bicycle ergometer for 12 approximately 38 min. After T ty reached approximately one degree higher (degrees C) than base line, ABRs and carotid blood flow were recorded in the course of cooling down. T ty was elevated by 0.82 +/- 0.27 degrees C T rec, 0.60 +/- 0.26 degrees C after exertion. ABR I-V IPL was shortened from 4.24 +/- 0.60 to 4.10 +/- 0.11 msec (p less than 0.01) and it was correlated with T ty change rather than T rec or carotid blood flow. ABR IPL returned to base line as T ty came down. II. Heating group Six young males were examined.
