New approaches in radical carbonylation chemistry are described. We have successfully integrated tin mediated radical carbonylation chemistry into modern fluorous applications and separation techniques. We revealed that radical carbonylation reactions can be performed using fluorous tin mediators, such as fluorous tin hydride and fluorous allyltin reagents. Fine tuning of the reaction conditions resulted in a good efficiency equivalent to conventional tin mediators. The tedious procedure of removing organotin byproducts can be circumvented through the use of fluorous/organic liquid-liquid extraction or fluorous liquid-solid phase extraction with fluorous reverse phase silica (FRPS). Also described are newly developed tandem carbonylation reactions that are based on species hybridization approaches. Using a radical/anionic hybrid system based on zinc-induced one-electron reduction, we achieved a three-component coupling reaction consisting of 4-alkenyl iodides, carbon monoxide, and electron-deficient alkenes. We observed two types of annulations processes, namely [4 + 1](radical)/[3 + 2](anionic) and [5 + 1](radical)/[3 + 2](anionic), which lead to the production of bicyclo[3.3.0]octanols and bicyclo[3.2.1]octanols, respectively. We found a radical/palladium hybrid system to be useful in the construction of new cyclic systems that incorporate two or three molecules of carbon monoxide.

In order to clarify the role of antioxidant enzymes in the male rat submandibular gland against short-term normobaric oxygenation, we performed immunocytochemical staining of manganese-containing superoxide dismutase (Mn-SOD), copper- and zinc-containing SOD (Cu/Zn-SOD), catalase (CAT), glutathione peroxidase, and glutathione S-transferases (GST alpha, GST mu, and GST pi) between days 1 and 7 after normobaric oxygenation. Ultrastructural alterations and immunoreactivities for malondialdehyde (MDA), a lipid peroxidation-related molecule, of the acinar and ductal cells after the oxygenation were also investigated. Immunoreactivity for MDA was exhibited in the acinar cells throughout the experiment. On the other hand, immunoreactivity for the SODs, CAT, and GSTs was not altered, when compared to that of controls, but was significantly elevated in the granular, striated, and excretory ductal cells. Since an increase of lipid peroxidation as indicated by enhanced immunoreactivity for MDA was detected in the acinar and intercalated ductal cells, the results indicate that the enhanced antioxidant enzymes in the granular, striated, and excretory ductal cells play a crucial role in the self-defense system of the male rat submandibular gland against normobaric oxygenation.

The Ag(I)-promoted oxidative meso-meso coupling reaction of 5,15-diaryl Zn(II)-porphyrin was serendipitously found in the course of our synthetic approaches towards photosynthetic reaction centers. Based on this reaction, a variety of directly linked and fused porphyrin arrays have been synthesized, including linear meso-meso-linked porphyrin arrays, windmill- and grid-shaped porphyrin arrays, meso-beta singly linked diporphyrins, beta-beta linked diporphyrins, meso-beta doubly linked (fused) diporphyrins and oligoporphyrins, meso-meso beta-beta doubly linked (fused) diporphyrins, and meso-meso beta-beta-beta-beta triply linked (fused) diporphyrins. The meso-meso coupling reaction of 5,15-diaryl Zn(II)-porphyrins is advantageous considering its high regioselectivity as well as its ease of extension to large porphyrin arrays as is demonstrated by the synthesis of a discrete meso-meso-linked 128-mer and poly(5,15-porphyrinylene). Finally, the oxidation of end-phenyl capped meso-meso-linked zinc porphyrins with DDQ-Sc(OTf)(3) gave pi-conjugated flat porphyrin tapes. To the best of our knowledge, the meso-meso linked 128-mer is the longest man-made discrete molecule, and the porphyrin tape 12-mer is the most extensively conjugated porphyrin array, as evinced by the lowest electronic band peak at 3500 cm(-1).

The development of the complex network of specialized cells that form the atrioventricular conduction system (AVCS) during cardiac morphogenesis occurs by progressive recruitment within a multipotent cardiomyogenic lineage. Understanding the molecular control of this developmental process has been the focus of recent research. Transcription factors representative of multiple subfamilies have been identified and include members of zinc-finger subfamilies (GATA4, GATA6 HF-1b), skeletal muscle transcription factors (MyoD), T-box genes (Tbx5), and also homeodomain transcription factors (Msx2 and Nkx2.5). Mutations in some of these transcription factors cause congenital heart disease and are associated with cardiac abnormalities, including deficits within the AVCS. Mouse models that closely phenocopy known human heart disease provide powerful tools for the study of molecular effectors of AVCS development. Indeed, investigations of the Nkx2.5 haploinsufficient mouse have shown that peripheral Purkinje fibers are significantly underrepresented. This piece of data corroborates our previous work showing in chick, mouse, and humans that Nkx2.5 is elevated in the differentiating AVCS relative to adjacent working ventricular myocardial tissues. Using the chick embryo as a model, we show that this elevation of Nkx2.5 is transient in the network of conduction cells comprising the peripheral Purkinje fiber system. Functional studies using defective adenoviral constructs, which disrupt the normal variation in level of this gene, result in perturbations of Purkinje fiber phenotype. Thus, the precise spatiotemporal regulation of Nkx2.5 levels during development may be required for the progressive emergence of gene expression patterns specific to differentiated Purkinje fiber cells.

The photocurrent at the polarized water/1,2-dichloroethane (DCE) interface was successfully observed in the presence of a lipophilic sensitizer, 5,10,15,20-tetraphenylporphyrinato zinc (ZnTPP), in the organic phase. The photocurrent transient responses were apparently affected by the employed organic supporting electrolyte: tetrapenthylammonium tetraphenylborate (TPnATPB) or tris(tetraoctylammonium)tungstophosphate ((TOcA)3PW12O40). The photocurrent measured in the TPnATPB system exhibited rather slow responses associated with the ion transfer of photoproducts. On the other hand, the photoinduced heterogeneous electron transfer could be observed in the use of (TOcA)3PW12O40. The photocurrent intensity in the (TOcA)3PW12O40 system exhibited an apparent pH dependence and the photoreduction of hydrogen ions probably took place at the water/DCE interface. By analyzing the real and imaginary components of the photocurrent depending on the photoexcitation frequency, we roughly estimated the phenomenological rate constants of the product separation (k(ps)) and recombination (k(rec)) processes as log(k(ps)/s(-1)) = 1.5 +/- 0.2 and log(k(rec)/s(-1)) = 1.8 +/- 0.1, respectively.

The effect of factors including the horses' farm environment, their sex and age and whether they suffered from summer seasonal recurrent dermatitis (sweet itch) on the concentrations of zinc in the plasma, whole blood and blood cells of 104 Icelandic horses was investigated. Its concentration in plasma varied significantly between farms (P<0.01), but its concentration in blood and blood cells was not influenced by any of the variables. The concentration of zinc in the blood cells was 10.5 times greater than in plasma, but its concentration in plasma was not correlated with its concentration in whole blood or blood cells owing to the variability in the proportion of whole blood zinc present in plasma (relative plasma zinc), which ranged between 9 and 24 per cent. This variability was significantly influenced by a three-way interaction between farm, sex and sweet itch (P<0.05). Relative plasma zinc was positively correlated with absolute plasma zinc (r=0.78, P<0.001) and negatively correlated with whole blood and blood cellular zinc (r=-0.58, r=-0.71, P<0.001).

Outbreaks of pithomycotoxicosis (facial eczema), a hepatogenous photosensitisation caused by the mycotoxin sporidesmin, have affected ruminants in the Azores Islands of Portugal after warm, humid periods during late summer and autumn. Twenty-two outbreaks were recorded in cattle between 1999 and 2001, affecting 11.4 per cent of the animals in the affected herds, and in 2000 there was an outbreak in one sheep flock in which more than 20 per cent of the sheep died. The clinical signs included decreases in milk production, weight loss, photosensitisation and its sequelae, including death. The animals had high activities of gamma glutamyltransferase in their serum, and icterus and severe liver disease, including biliary hyperplasia and fibrosis, were found postmortem. The characteristic spores of the toxigenic saprophytic fungus Pithomyces chartarum were found on grass; all 381 isolates of the fungus were toxigenic for sporidesmin by elisa, and the results were confirmed by high-performance liquid chromatography analysis. Cattle from farms at greatest risk of pithomycotoxicosis were protected by supplementing their concentrate feed with zinc oxide, or using a slow-release intraruminal zinc bolus.

Only few of the human endogenous retrovirus (HERV) sequences in the human genome can produce proteins. We have previously reported that (i) patients with germ cell tumors often make antibodies against proteins encoded by HERV-K elements, (ii) expression of the HERV-K rec gene in transgenic mice can interfere with germ cell development and induce carcinoma in situ, and (iii) HERV-K np9 transcript is overproduced in many tumors including breast cancers. Here we document that both Np9 and Rec physically and functionally interact with the promyelocytic leukemia zinc finger (PLZF) tumor suppressor, a transcriptional repressor and chromatin remodeler implicated in cancer and the self-renewal of spermatogonial stem cells. Interaction is mediated via two different central and C-terminal domains of Np9 and Rec and the C-terminal zinc fingers of PLZF. One major target of PLZF is the c-myc proto-oncogene. Coexpression of Np9 and Rec with PLZF abrogates the transcriptional repression of the c-myc gene promoter by PLZF and results in c-Myc overproduction, altered expression of c-Myc-regulated genes, and corresponding effects on cell proliferation and survival. Thus, the human endogenous retrovirus proteins Np9 and Rec may act oncogenically by derepressing c-myc through the inhibition of PLZF.

Alk-2-enyl(trialkyl)stannanes with heteroatom substituents at the 4-, 5-, and 6-positions are transmetallated stereoselectively using tin(IV) halides to generate allyltin trihalides, which react with aldehydes to give (3Z)-homoallylic alcohols with efficient 1,5-, 1,6-, and 1,7-stereocontrol. This chemistry has been used to develop new strategies for natural product synthesis. Because of the toxicity of organotin reagents and the problems in removing organotin residues from reaction products, alternative procedures that avoid the use of organotin reagents have been investigated. To date, alk-2-enylgermanium reagents have been shown to deliver effective 1,5- and 1,6-stereocontrol, which is analogous to that observed for the organotin compounds. Organobismuth intermediates, which can be generated from allyl bromides and zinc-bismuth(III)iodide, react with aldehydes with efficient 1,5-stereocontrol which is complementary to that observed with the organo-stannanes or -germanes in that (3E)-homoallylic alcohols are obtained.

Clinical measurements, including a subjective clinical score and water intake, and biochemical measurements, including blood glucose, fructosamine, beta-hydroxybutyrate, cholesterol, triglycerides, triglycerides corrected for free glycerol, glycerol and urine glucose were compared for monitoring diabetic cats treated with porcine insulin zinc suspension. The data were grouped by subjective clinical score and the sensitivity of each measurement in differentiating the grouped data was assessed. None of the measurements was able to differentiate between the ranked clinical score groups, but two-hourly measurements of blood glucose over 24 hours, water intake, urine glucose and fructosamine were useful in differentiating cats that subjectively had the water and food consumption and general appearance of a normal cat from cats in which the signs of diabetes were less well controlled. Measurements of plasma lipids were not well correlated with the other measurements. The measurements that were most closely correlated with apparently perfect clinical control were the J index, water intake and maximum and mean blood glucose concentrations. In practice, water intake, maximum blood glucose concentration, mean blood glucose concentration and urine glucose would be the most useful indicators of clinical control in diabetic cats treated with porcine insulin zinc suspension.
