Blood samples were taken before and after a cross country race over the marathon distance of 42 km. There was a rise in blood glucose and plasma free fatty acids and glycerol associated with a rise in plasma cortisol and glucagon but the fall in insulin was not significant (P > 0.05). Plasma

and albumin concentrations increased, calcium decreased and there was no change in sodium or bicarbonate concentrations. There was an increase in plasma urea, creatinine, uric acid, bilirubin and isocitrate dehydrogenase but no change in alkaline phosphatase. There was a rise in plasma creatine kinase. These results of a competitive race are compared with those of the 80 km non-competitive Golden Horseshoe Ride.

Forty-two mineral balances were measured in 50 kg pigs fed various diets, including barley, maize or potato, and used to examine the presumption that the current recommended dietary requirements for the major minerals, especially phosphorus, are essential to the well-being of the pig. All the diets contained similar levels of all the mineral elements, except

0.22 or 0.16. The low efficiencies of utilisation of the digested minerals, especially phosphorus (0.53), suggested that a reduction in dietary phosphorus levels may be justified in terms of reducing the pollution of the environment with phosphorus caused by the application of pig slurry.

The results of analyses of plasma samples from healthy and sick adult birds and mammals of a wide range of species were used to calculate reference ranges and clinical ranges for 11 biochemical variables (calcium, sodium, chloride, total protein, albumin, globulin, inorganic phosphate,

, bicarbonate, urea and creatinine concentrations) in six mammalian Orders (Primata, Carnivora, Pinnipedia, Proboscidea, Perissodactyla and Artiodactyla) and six avian Orders (Ciconiiformes, Gruiformes, Pelecaniformes, Psittaciformes, Falconiformes and Strigiformes). The patterns of variation between Orders for the different biochemical variables were then used to provide guidelines for the interpretation of plasma biochemistry results in the species in these Orders for which there are no reference data. The observed clinical ranges provide an indication of the potential diagnostic value of each variable.

The history and results of a clinical examination were recorded for 32 spring-born suckler calves which were hospitalised for intravenous fluid therapy. Blood samples were taken before treatment, during treatment and before discharge and analysed for colostral status, total carbon dioxide as an indication of acid-base status, and haematocrit. All the calves were given intravenously 5 to 10 litres of electrolyte solution containing 144 mmol/litre sodium, 4 mmol/litre
potassium
, 113 mmol/litre chloride and 35 mmol/litre bicarbonate, supplemented, in 24 calves, with up to 450 ml of 1M sodium bicarbonate. Nearly all the calves were recumbent but less than half were dehydrated on admission. The signs of dehydration were well correlated with each other and with the haematocrit. Neither the history nor the clinical signs were useful predictors of acidosis. There was no relationship between the severity of acidosis and the degree of dehydration. Acidosis was more prevalent in older calves (P < 0.01). For the severely acidotic calves, supplementary intravenous fluid with sodium bicarbonate significantly (P < 0.05) improved the total blood carbon dioxide at discharge. All 32 calves recovered. It is possible to treat acidotic calves with intravenous fluid therapy effectively, economically and according to their individual needs. The Harleco apparatus is a simple, useful, cost-effective adjunct to the diagnosis and treatment of this life-threatening condition.

A combination of light and electron microscopy techniques was applied to characterize a luteinizing hormone (LH)-gonadotroph-like cell in the anterior pituitary gland of the adult domestic chicken. This cell type (mean +/- sem, 91 +/- 8 microns2) was larger than typical LH-gonadotrophs (71 +/- 5 microns2, p < 0.01) and seen in pituitary glands from adult males but not those from adult females of the same age. The ultrastructural features of these cells were similar to typical LH-gonadotrophs in the same section, except for the presence of large (1,126 +/- 77 nm diameter), round, or polymorphic electron-dense inclusions (14 +/- 3 per cell) in the cisternae of the endoplasmic reticulum. When resin sections of anterior pituitary gland were stained with the vapours from the fixative osmium tetroxide and a solution of
potassium
ferricyanide, inclusions were found that appeared to be lipid in nature and were circumscribed by the cisternal membranes of the endoplasmic reticulum. These lipid inclusions were associated occasionally with acid phosphatase and lysosome-like bodies. The cellular pigment, lipofuscin, considered to be a marker of aging, was absent from the anterior pituitary glands from these adult chickens. These observations suggest that the appearance of lipid in LH-gonadotrophs is unlikely to be age-related and an alternative explanation is discussed in relation to a sex difference in the LH response of adult chickens to gonadotrophin-releasing hormone (GnRH).

Reference values for some haematological and plasma biochemical constituents were established in Spanish ibex (Capra pyrenaica hispanica) restrained either physically or chemically with tiletamine-zolazepam. The following variables were studied: haematocrit, haemoglobin concentration, total erythrocyte and leucocyte counts, haematological indices, erythrocyte dimensions, differential count of leucocytes, glucose, urea, uric acid, cholesterol, triglycerides, creatinine, aspartate aminotransferase, alanine aminotransferase, alkaline phosphatase, creatinine phosphokinase, lactic dehydrogenase, gamma-glutamyl transpeptidase, total plasma protein, albumin, globulins, albumin-globulin ratio, sodium,
potassium
, calcium, magnesium, total phosphorus, chloride and osmolality. No haematological data have been published before but the values observed were in the general range of other artiodactyls, with the exception of the number and size of the erythrocytes which were respectively larger and smaller than in most other ruminants. Significant differences were found for a number of the variables between the values recorded in physically restrained animals and the values recorded in anaesthetised animals; they included the number of erythrocytes and related parameters, the plasma proteins and some inorganic ions.

A method for creating tough, flexible, bronchoalveolar casts from formalin-fixed canine lung is described. A lung was washed using simple methods and fixed in 10% neutral-buffered formalin. While still wet with formalin, an intact lobe was injected with silicone sealant, Silastic 734 RTV (Room Temperature Vulcanizing), using a caulk gun. Following digestion with protease and corrosion with
potassium
hydroxide, a bronchoalveolar cast was recovered giving detail as shown using scanning electron microscopy or conveniently seen by stereo light microscopy. This method should be useful for micro-anatomy studies of normal and diseased lungs.

Lactating mammary glands fixed by perfusion with 5% glutaraldehyde subsequently were postfixed with

ferrocyanide reduced osmium or were treated with tannic acid. Stained thin sections were examined with the electron microscope and stereopairs were prepared. The distribution of casein submicelles was analyzed in the various components of the Golgi apparatus. The Golgi stacks were composed of five or six elements, all of which contained casein submicelles 20 nm in diameter. The cis-tubular network or cis-element, as well as the underlying three or four midsaccules, showed these casein submicelles either attached to their membrane or free in the lumen. The trans-most element of the stacks formed distended prosecretory granules in which both isolated or clustered casein submicelles were suspended in an electron-lucent fluid. These micellar aggregates increased in size and became progressively more compact to form spherical dense bodies or casein micelles, in which the individual 20 nm particles could easily be resolved. Casein micelles were seen in secretory granules in addition to a wispy material of low density. The numerous small spherical vesicles (80 nm or larger) seen on the cis, lateral, or trans aspects of the stacks did not appear to contain free casein submicelles. This raises questions regarding the role of these vesicles in the transport of casein macromolecules through the Golgi stacks. It was noticeable that in this Golgi apparatus a trans-Golgi network was limited to a few small residual tubules free from casein submicelles. It thus appears that the greater part of the trans-most Golgi element gives rise to the large prosecretory granules. After leaving the Golgi region and prior to exocytosis, the secretory granules often fuse to form larger granules before exocytosis.

A study was made of the effects of exogenous adrenocorticotropin (ACTH) on the levels of blood components in 109 dairy replacement calves and the statistical correlations between these effects and the growth rates of the calves from birth to six months. Blood samples were taken from a jugular vein before ACTH was injected and then at two, four, six and eight hours afterwards, and analysed for plasma cortisol concentration, total white cell counts, packed cell volume, haemoglobin, plasma glucose, sodium,
potassium
, magnesium and inorganic phosphorus, erythrocyte sodium,
potassium
and magnesium, serum ionised calcium and total protein and total plasma calcium concentration. The injection of 1.1 +/- 0.02 iu/kg of ACTH intramuscularly resulted in a peak plasma cortisol concentration after two hours which had not returned to normal after eight hours. It also resulted in leucocytosis, lymphopenia, neutrophilia, eosinopenia and hypophosphataemia; the mean changes were repeatable (P < 0.05) in 49 of the calves tested two months later. The weight gains to six months of age could be predicted from the degree of the changes in several blood constituents. Significant partial regression coefficients were found for the change in glucose concentration (0 to four hours), absolute neutrophil count (0 to two hours), absolute lymphocyte count (0 to four hours) and loge absolute eosinophil count (0 to two hours). The multiple regression sum of squares was highly significant (P < 0.0001), and the multiple coefficient of determination was 0.305. It was concluded that the changes in these blood components after an injection of ACTH might be used to predict the weight gains of dairy replacement calves.

The present study investigated the relationship between plasma

ion concentration ([K+]) and skeletal muscle torque during three different 15-min recovery periods after fatigue induced by four 30-s sprints. Four males and one female completed the multiple sprint exercise on three separate days; recovery was passive, i.e. no cycling exercise (PRec), active cycling at 30% peak oxygen consumption. VO2peak (30%

) and active cycling at 60% VO2peak (60%

). Plasma [K+] was measured from blood sampled from an antecubital vein of subjects at rest and at 0, 3, 5, 10 and 15 min into each recovery. Isokinetic leg strength was measured at rest and at 1, 6, 11 and 16 min during each recovery. Following the exhaustive sprints, [K+] increased significantly from an average mean (SEM) resting value of 3.81 (0.07) mmol.l-1 to 4.48 (0.19) mmol.l-1 (P < 0.01). In all recovery conditions, plasma [K+] returned to resting levels within 3 min following the fourth sprint. However, in the two active recovery conditions plasma [K+] increased over the remainder of the recovery periods to 4.36 (0.12) mmol.l-1 in the 30%
