Two dogs with metabolic epidermal necrosis had hyperkeratosis of the footpads accompanied by erythematous, erosive and crusting lesions affecting the muzzle, external genitalia, perineum and periocular regions. Histopathological examination of skin biopsies revealed a superficial hydropic dermatitis with marked parakeratosis. Both dogs had high plasma activities of alkaline phosphatase and alanine aminotransferase and high concentrations of
glucose
, and also a marked hypoaminoacidaemia. Despite these similarities, the cutaneous eruptions were associated with different underlying diseases. One dog had a pancreatic carcinoma which had metastasised widely; the primary tumour and the metastases showed glucagon immunoreactivity on immunocytochemical staining, and the dog's plasma glucagon concentration was markedly greater than that of control dogs. The other dog had diffuse hepatic disease; its plasma glucagon concentration was similar to that of control samples and cirrhosis was identified post mortem. Metabolic epidermal necrosis in dogs is a distinct cutaneous reaction pattern which may be associated with different underlying systemic diseases; however, the pathogenesis of the skin lesions remains unclear.

Twin and single pregnancies were induced in two groups of oestrus-synchronised beef cows by using a combination of artificial insemination and the transfer of in vitro fertilised (IVF) embryos. Single IVF embryos transferred non-surgically to the uterine horn contralateral to the corpus luteum of 43 previously inseminated cows resulted in a calving rate of 72 per cent with a twinning rate of 38.7 per cent. In 45 cows, two IVF embryos were transferred non-surgically to one uterine horn resulting in a calving rate of 51.1 per cent with a twinning rate of 39.1 per cent. The median gestation length for cows bearing twins was 10 days shorter than that of cows bearing single calves (P < 0.001). The proportions of cows that received assistance at calving were similar for twin and single births (57 per cent vs 45 per cent, P > 0.05), but the incidence of retained fetal membranes was much higher after the birth of twins (62 per cent vs 3 per cent, P < 0.001). Nineteen per cent of twin calves were stillborn compared with 6 per cent of single calves (P > 0.05). The median birthweight of the twin calves was 32 kg (68 per cent of the median weight of single calves). , non-esterified fatty acid and beta-hydroxybutyrate concentrations. Serum immunoglobulin concentrations were similar in single and twin calves suggesting that the passive transfer of antibody was not compromised in the twin calves.

Ten horses underwent a standardised strenuous treadmill exercise test, before, during and after which measurements were made of plasma beta-endorphin and cortisol concentrations, blood lactate,
glucose
, haemoglobin and pH, the activities of creatine kinase, lactate dehydrogenase and aspartate amino-transferase, and heart rate, oxygen uptake and expired minute volume. The correlations between the exercise-induced response of beta-endorphin and the changes observed in the other physiological measurements were examined. There was a large variation in the beta-endorphin response of the horses to exercise. The increase in beta-endorphin was correlated significantly and inversely with the treadmill velocity at which maximal oxygen uptake was reached. It was also significantly and directly correlated with the heart rate during recovery, the increase in plasma lactate concentration and the change in blood pH, indicating that the exercise-induced increase in beta-endorphin concentration was smaller in horses with a higher aerobic capacity.

The records of 21 horses with an adenoma of the pars intermedia of the pituitary gland were reviewed. The animals comprised nine Dutch warmblood horses and 12 ponies. They ranged in age from 12 to 30 years (mean +/- sem 21 +/- 1.3 years) for 20 of the horses, and comprised 13 geldings and eight mares. All the animals showed hirsutism. In 1991, nine horses were diagnosed as having the adenoma, equivalent to 0.5 per cent of the horses examined in 1991. The mean +/- sem survival time of four of the horses (from discharge to the development of complications which required euthanasia) was 192 +/- 59 days (range 120 to 368 days). Routine biochemical measurements were within their normal ranges except for a high alkaline phosphatase activity (768 +/- 487 iu/litre) and a high plasma
glucose
concentration (7.8 +/- 0.9 mmol/litre) in 16 of the horses; only six had a
glucose
concentration above 9.0 mmol/litre. The basal plasma concentration of adrenocorticotrophic hormone (ACTH) in the diseased horses was at least 191 pg/ml. Eight of the horses had mean plasma cortisol concentrations before and after the administration of 25 iu of synthetic ACTH1-24 of 168 +/- 21 (range 74 to 248) and 478 +/- 58 nmol/litre (range 248 to 771), respectively. Analysis of the urine of five of the horses revealed a mean specific gravity of 1.034 +/- 0.0034 (range 1.025 to 1.045).(ABSTRACT TRUNCATED AT 250 WORDS)

When 24 cows were classified according to whether they had a better or worse energy status (defined as a plasma beta-hydroxybutyrate level below or above 15 mg/100 ml and a plasma
glucose
level above or below 55 mg/100 ml) the median interval between calving and the onset of cyclicity was shorter and conception rates were better for the cows with a better energy status at seven and 14 days after calving. This was not repeated when the cows' energy status was assessed as better at either 21 days after calving or at the time of service. In the same group of cows there was a positive correlation between the loss of condition score in early lactation and beta-hydroxybutyrate levels. In a further trial 15 cows fed a silage-based diet during the dry period had better conception rates and greater luteinising hormone responses to buserelin than 14 cows fed a straw-based diet. The straw-fed cows lost more weight before calving and had significantly higher non-esterified fatty acid levels. After calving both groups of cows were fed the same diet but the cows fed straw in the dry period produced less milk and lost less weight than the cows fed silage. Dry cow nutrition affected subsequent fertility and production. The non-esterified fatty acid level in the last week or two of pregnancy is suggested as a practical indication of the level of dry cow nutrition and its probable effect on productivity. No relationships were observed between plasma urea concentrations and the fertility of the cows.

Recombinant bovine somatotrophin was used in addition to conventional therapy to treat a sample of post-calving cows in a herd suffering a high incidence of fat cow syndrome. Somatotrophin was also given to cows before they calved. There were no significant differences between the treated and untreated groups in the plasma concentrations of
glucose
, beta-hydroxybutyrate, albumin, total protein or calcium. Significant decreases in mean plasma urea concentration were observed in the pre-calving cows seven and 10 days after treatment with somatotrophin, and a significant decrease in mean plasma urea concentration was also recorded in post-calving cows with the fat cow syndrome. There was no evidence that somatotrophin, with its many potent actions in cows with acetonemia, exacerbated clinical ketosis. The authors' subjective clinical impression was that somatotrophin was helpful in the treatment of cows with the fat cow syndrome.

The three-dimensional configuration of the Golgi apparatus has been examined with the electron microscope in thick Golgi sections of Saccharomyces cerevisiae prepared from a wild-type strain and from sec7 mutants maintained for various periods of time at the nonpermissive temperature of 37 degrees C and then returned to the permissive temperature of 24 degrees C. Reduced osmium postfixation of glutaraldehyde fixed specimens stained intensely the content of Golgi elements and thus facilitated their three-dimensional characterization. In wild-type S. cerevisiae, the Golgi elements usually appeared as isolated networks of membranous tubules dispersed throughout the cytoplasm. Along such networks, distensions filled with stained material were similar in size to nearby secretory granules, suggesting that the latter formed by fragmentation of the Golgi elements. In sec7 mutants maintained at 37 degrees C in low (0.1%)
glucose
medium, secretion granules progressively decreased in number and soon disappeared. Concomitantly the networks of Golgi tubules increased in size and complexity, lost their distensions, and then transformed into flattened saccules forming stacks of up to seven or eight saccules that were similar to the Golgi stacks seen in mammalian cells. However in contrast to the latter, connections between the saccules were evident and Golgi-associated small vesicles were generally absent. Following return to the permissive temperature (24 degrees C), secretion granules reappeared, the Golgi stacks progressively decreased in size, and resumed their initial state of separated small tubular networks. Laboratories engaged in secretory studies of rat pancreatic islets often encounter high baseline insulin secretion with poor secretory response to secretagogues, such as
glucose
. The specific morphologic abnormalities that accompany this unregulated release have not been described. We isolated islets comparing two approaches. Both used stationary digestion with collagenase. In method I, we distended the biliary duct extracorporeally with collagenase and minced the pancreas after a 28 min digestion (37 degrees C). In method II, we distended the pancreas intracorporeally and digested for 40 min without mincing. Both methods utilized a similar collagenase concentration (2 micrograms/ml in Hank's balanced salt solution (HBSS). Both methods yielded over 300 islets/rat. Islets from both methods appeared intact, when viewed under the dissecting microscope. We found that adequate secretion from incubated islets was evoked with method I, i.e., low basal insulin levels at low
glucose
(3.3 mM), tripling at 11.0 mM
glucose
, and nearly quadrupling in response to higher
glucose
(16.7 mM). In contrast, method II was characterized by high basal levels without response to higher
glucose
. Ultramicroscopic examination of islet B cells in method I revealed normal cytological features, while B cells in method II showed marked degranulation, profiles of swollen endoplasmic reticulum, and swollen mitochondria. Morphometric analysis of B cells confirmed quantitatively a decrease in secretory granule density and mitochondrial enlargement in method II compared to method I. Anatomic changes, largely confined to the B cells of islets may account for functional alterations of responses. Defects cannot be predicted from gross appearance of islets.

Reference values for some haematological and plasma biochemical constituents were established in Spanish ibex (Capra pyrenaica hispanica) restrained either physically or chemically with tiletamine-zolazepam. The following variables were studied: haematocrit, haemoglobin concentration, total erythrocyte and leucocyte counts, haematological indices, erythrocyte dimensions, differential count of leucocytes,

, urea, uric acid, cholesterol, triglycerides, creatinine, aspartate aminotransferase, alanine aminotransferase, alkaline phosphatase, creatinine phosphokinase, lactic dehydrogenase, gamma-glutamyl transpeptidase, total plasma protein, albumin, globulins, albumin-globulin ratio, sodium, potassium, calcium, magnesium, total phosphorus, chloride and osmolality. No haematological data have been published before but the values observed were in the general range of other artiodactyls, with the exception of the number and size of the erythrocytes which were respectively larger and smaller than in most other ruminants. Significant differences were found for a number of the variables between the values recorded in physically restrained animals and the values recorded in anaesthetised animals; they included the number of erythrocytes and related parameters, the plasma proteins and some inorganic ions.

A study was made of the effects of exogenous adrenocorticotropin (ACTH) on the levels of blood components in 109 dairy replacement calves and the statistical correlations between these effects and the growth rates of the calves from birth to six months. Blood samples were taken from a jugular vein before ACTH was injected and then at two, four, six and eight hours afterwards, and analysed for plasma cortisol concentration, total white cell counts, packed cell volume, haemoglobin, plasma
glucose
, sodium, potassium, magnesium and inorganic phosphorus, erythrocyte sodium, potassium and magnesium, serum ionised calcium and total protein and total plasma calcium concentration. The injection of 1.1 +/- 0.02 iu/kg of ACTH intramuscularly resulted in a peak plasma cortisol concentration after two hours which had not returned to normal after eight hours. It also resulted in leucocytosis, lymphopenia, neutrophilia, eosinopenia and hypophosphataemia; the mean changes were repeatable (P < 0.05) in 49 of the calves tested two months later. The weight gains to six months of age could be predicted from the degree of the changes in several blood constituents. Significant partial regression coefficients were found for the change in
glucose
concentration (0 to four hours), absolute neutrophil count (0 to two hours), absolute lymphocyte count (0 to four hours) and loge absolute eosinophil count (0 to two hours). The multiple regression sum of squares was highly significant (P < 0.0001), and the multiple coefficient of determination was 0.305. It was concluded that the changes in these blood components after an injection of ACTH might be used to predict the weight gains of dairy replacement calves.
