Cat intrafusal muscle fibers were examined histochemically in serial transverse sections of tenuissimus muscle spindles. The "myofibrillar" adenosine triphosphatase staining reaction was used to recognize the nuclear bag and the nuclear chain fibers in 309 spindle poles. Poles of 40 nuclear chain fibers extended for 1,000 micrometer or more beyond the termination of the spindle capsule. These
long chain
fibers stained less intensely for nicotinamide adenine dinucleotide tetrazolium reductase (NADH-TR) than the typical chain fibers of shorter polar length. In sections stained for cholinesterases (ChE), the extracapsular regions of most
long chain
fibers displayed one or two short, dense "plate"-type ChE deposits, which may represent the terminals of skeleto-fusimotor axons. In addition, about one-third of the
long chain
fibers displayed one or more thinner and smaller areas of ChE activity, possibly corresponding to the endings of fusimotor axons. The overall ChE staining pattern of the typical chain fibers was unlike that of the long chains. However, some of the shorter nuclear chain fibers resembled
long chain
fibers with the NADH-TR reaction, even though their ChE "plates" were located intracapsularly. It is concluded that nuclear chain fibers in the cat spindle form a class of intrafusal fibers with heterogeneous histochemical properties, and that the
long chain
fibers represent one fiber subtype.

The development of solvent-free low-dimensional polymer electrolytes intended for use in solvent-free lithium batteries operating at ambient or sub-ambient temperatures is described. The synthetic routes to the amphiphilic polymers I having 5-alkoxy-3,4-phenylene units connected with oligoethoxy segments via polyester-ether or pure polyether links (abbrev. CmOn, m = 12, 16, 18, n = 1-5) and to the copolymers CmO1-CmOn are described. The structures, thermal properties and SAXS long spacings of their complexes with lithium salts (type A) and with
long chain
n-alkane or alkyl side chain intercalation (type B) are discussed. However, high ambient conductivities (10(-4)-10(-3) S cm(-1)) are observed in type C systems when a second copolymer based on polytetramethylene oxide segments (II) is incorporated as a microphase between the lamellae of I and serving as an ion bridge or "glue". DC polarization between Li electrodes also gives ambient conductivities >/=ca.10(-3) S cm(-1). In type D systems the I/II interface is stabilized by including a copolymer III, promoting high reproducibility in performance. Copolymers I of CmO1-CmO5 having CmO1 in excess give optimum conductivities with low temperature-dependence. This, together with molecular modeling, suggests uncoupled ion mobilities by hopping between small aggregates in the interlamellar spaces.

The post-translational addition of C-16
long chain
fatty acids to protein cysteine residues is catalysed by palmitoyl-S-acyl-transferases (PAT) and affects the affinity of a modified protein for membranes and therefore its subcellular localisation. In apicomplexan parasites this reversible protein modification regulates numerous biological processes and specifically affects cell motility, and invasion of host cells by Plasmodium falciparum merozoites and Toxoplasma gondii tachyzoites. Using inhibitor studies we show here that palmitoylation is key to transformation of zygotes into ookinetes during initial mosquito infection with P. berghei. We identify

as a unique PAT mediating ookinete formation and morphogenesis. Essential for life cycle progression in asexual blood stage parasites and thus refractory to gene deletion analyses, we used promoter swap (ps) methodology to maintain dhhc2 expression in asexual blood stages but down regulate expression in sexual stage parasites and during post-fertilization development of the zygote. The ps mutant showed normal gamete formation, fertilisation and DNA replication to tetraploid cells, but was characterised by a complete block in post-fertilisation development and ookinete formation. Our report highlights the crucial nature of the
DHHC2
palmitoyl-S-acyltransferase for transmission of the malaria parasite to the mosquito vector through its essential role for ookinete morphogenesis.

Reversible lysine deacetylation is exerted by both zinc and NAD

-dependent deacetylases. It is an important factor in epigenetic regulation and more generally in the posttranslational regulation of protein stability, association and activity. Some of these enzymes can also cleave off fatty acids or dicarboxylic acids from lysines in proteins. The NAD
+
-dependent deacetylases are termed Sirtuins and are implicated in the pathogenesis of different diseases. For the isotype Sirt2 highly selective inhibitors have been identified in the last few years. Many of those Sirt2 selective compounds, like the Sirtuin rearranging ligands (SirReals) discovered in our group, have been shown or are postulated to bind to the so-called selectivity pocket. This binding site is not observed in crystal structures of the apo-enzyme but can be opened up by
long chain
fatty acid substrates respectively suitable inhibitors. Recently, this unique feature of Sirt2 was exploited to provide highly potent and selective tools for the chemical biology of Sirtuins. Here, we shortly review Sirtuin biology, present inhibitors that have either been confirmed or postulated to bind to the selectivity pocket, their applications and an outlook regarding mechanistic investigations.
