Pigs from four farms (two producing 'easy' and two 'difficult to handle' pigs) were slaughtered at two abattoirs, each with two slaughter handling systems, so that 25 pigs of each behavioural type were slaughtered by all four handling systems on each day; there were 16 replicates (3200 pigs) in total. The pigs at abattoir X were electrically stunned, either in a floor pen holding five pigs or in a race-restrainer. The following measurements were made: hot carcase weight and backfat thickness at P2, degree of rigor mortis 35 minutes post mortem, skin blemish, pH and muscle reflectance in the m longissimus dorsi at 60 minutes and 18 hours post mortem, and pH and muscle reflectance in the m adductor at 18 hours post mortem. At abattoir X, the pigs slaughtered in the race-restrainer had heavier carcases (74.0 kg vs 73.0 kg, P < 0.05), developed rigor mortis more rapidly (8.1 mm vs 7.3 mm, P < 0.01), had more skin blemish (2.8 vs 2.7, P < 0.01), paler m longissimus dorsi muscles after one hour (15.7 vs 13.9, P < 0.01) and 18 hours (27.8 vs 26.6, P < 0.05), and paler m adductor muscles (24.5 vs 22.7, P < 0.001) after 18 hours. PMID:Pig welfare and carcase quality: a comparison of the influence of slaughter handling systems at two abattoirs. 784 33
Hypoxaemia commonly develops during general anaesthesia and in the recovery period in horses. The Hudson demand valve has been used to increase arterial PO2, but it has been found to increase airway resistance considerably when used during spontaneous ventilation. This paper evaluates a modification of the valve designed to reduce this resistance. The effects of the valve and its modification on arterial oxygen (PaO2), and
carbon dioxide
(PaCO2) tensions were evaluated in four ponies anaesthetised by a total intravenous technique. The valve increased PaO2 from 8.3 +/- 1.1 to 32.7 +/- 7.6 kPa during spontaneous ventilation and to 44.2 +/- 7.4 kPa during intermittent positive pressure ventilation. With the modification, the PaCO2 was increased to 9.0 +/- 2.5 kPa during spontaneous ventilation PaO2 was unchanged by the valve (7.2 +/- 0.4 kPa to 7.1 +/- 0.7 kPa) but it was reduced to 6.4 +/- 0.9 kPa with the modification. The valve was also evaluated in 20 clinical cases during their recovery from halothane anaesthesia. It increased PaO2 from 7.4 +/- 2.1 kPa to 17 +/- 18.3 kPa during spontaneous ventilation and from 8.0 +/- 1.8 kPa to 23.4 +/- 22.2 kPa during positive pressure ventilation. With the modification, PaO2 was increased from 7.8 +/- 1.4 kPa to 10.4 +/- 3.8 kPa during spontaneous ventilation and from 7.6 +/- 1.5 kPa to 14.8 +/- 8.4 kPa during positive pressure ventilation. During spontaneous ventilation PaCO2 was increased from 5.9 +/- 0.4 kPa to 6.2 +/- 0.6 kPa with the unmodified valve and from 6.3 +/- 0.5 kPa to 6.6 +/- 0.5 kPa with the modification.(ABSTRACT TRUNCATED AT 250 WORDS)

PMID:Evaluation of a modification of the Hudson demand valve in ventilated and spontaneously breathing horses. 788 96
The vascular interrelationship of the well-established porcine placenta has previously been described from vascular casts and histology, but not its developmental stages. This study was performed using the same methods on 17 sows of well-known stages of gestation ranging from 9 1/2 to 43 days post coitum (p.c.). At the precontact stage, days 9 1/2 to 12 1/2 p.c., the subepithelial capillaries formed a wide open meshwork of variable diameter, 3-14 microns, without any difference between meso- and antimesometrial side. At the early contact and adhesion stages (days 13 to 18 p.c.), the first increase in vasculature was seen at the mesometrial side close to the embryonic disc of the very long blastocyst at day 15 p.c., 2 days after the first contact between trophoblast and maternal epithelium was seen. At day 18 p.c., the areas with dense capillaries increased markedly at the mesometrial side with the same parallel organization as seen at day 15 p.c., whereas the antimesometrial side still had a relative loose appearance comparable to the previous stages. At the early placental stages (days 20 1/2 to 23 p.c.), the capillary bed formed smooth folds, which in some areas at day 20 1/2 days developed into smaller folds or prerugae. Here the capillaries changed to convoluted forms with slightly bulbous dilations measuring about 30-35 microns in diameter. This developmental progress became more elaborate at day 23: capillaries of the low ridges of prerugae formed irregular dilations up to 50 microns in some areas. At this stage the parallel arrangement of the capillary meshwork characteristic of the previous stage was not longer discernable. By days 32-43 p.c., an increase in microscopic folding was present, and the maternal arterioles could be traced to the top of the ridges, creating the characteristic vascular architecture needed for an efficient exchange of oxygen,

Eighteen Belgian white and blue double-muscled calves suffering from the acute respiratory distress syndrome were studied. Fifteen of the calves inhaled ipratropium bromide (0.6 mg) four times a day for three to four days whereas the other three control calves inhaled sterile 0.9 per cent saline. All the animals were injected with ceftiofur sodium (1 mg/kg/day) for five days, the first injection being given one hour after the first inhalation of ipratropium bromide or saline. Arterial oxygen tension, alveolar arterial oxygen difference,

tension and arterial pH, respiratory and heart rates, oscillatory resistance and phase angle, measured by the mono-frequency forced oscillation technique, were recorded both before and one hour and 168 hours after the first inhalation. The measurement of oscillatory resistance and phase angle made it possible to resolve the impedance of the respiratory system into its real and imaginary components. The oscillatory compliance (Cosc) was determined from the imaginary component (Im). By one hour after the first inhalation of ipratropium bromide the oscillatory resistance was already significantly reduced and Im and Cosc had significantly increased, but the other parameters showed no significant improvement. However, between one hour and 168 hours after the first inhalation all the parameters reached physiological values. The control calves did not show any change. It was concluded that the pulmonary dysfunction associated with the acute respiratory distress syndrome in these calves was at least partly due to a severe bronchoconstriction.(ABSTRACT TRUNCATED AT 250 WORDS)

To establish the mode of fertilization in a marsupial, a morphological investigation was made of the gametes of the South American grey short-tailed opossum. Monodelphis domestica, at the time of fertilization in vivo and in vitro. Oestrus was induced in females by the introduction of an unfamiliar male. To obtain oocytes recently fertilized in vivo, females were killed 18-24 hours after the first mating and the region of the oviduct containing eggs excised and fixed. Unfertilized mature oocytes were recovered from ovarian follicles 15-18 hours after first mating and fertilized in vitro with cauda epididymal spermatozoa in a modified MEM medium supplemented with bovine serum albumin at 37 degrees C in 5%
CO2
in air. Following sperm-egg binding and fertilization, oocytes were fixed and prepared for light and electron microscopy. Spermatozoa unpaired prior to fertilization in vivo and in vitro and single spermatozoa bound to the zona surface by their plasmalemma overlying the acrosome on the dorsal face of the sperm head. The acrosome reaction was only observed at the zona surface (suggesting that it may be induced by zona components) and involved a vesiculation of sperm plasma and acrosomal membranes over the main body of the acrosome but not over the narrow, marginal region which persisted after the acrosome reaction was complete. Sperm penetration of the zona pellucida caused a large breach in the zona and the dispersal of perivitelline material. The fusion of the spermatozoon with the oolemma occurred first over the marginal acrosomal region and was accompanied by a fertilization cone which protruded through the zona penetration hole. Activation of the egg was characterized by the release of material from vesicles in the peripheral cytoplasm and extrusion of the second polar body. The mode of fertilization in Monodelphis was compared with what is known in other marsupials (New World and Australian) and eutherian (placental) mammals. It was concluded that the general features of the acrosome reaction and sperm-egg fusion may be essentially similar in both groups and that an evolutionary schism did not occur following the development of the eutherian mode of fertilization.

and 60 per cent argon in air (2 per cent residual oxygen and 8 per cent residual nitrogen); (B) 90 per cent argon in air (2 per cent residual oxygen and 8 per cent residual nitrogen); (C) 65 per cent
carbon dioxide
in air (7 per cent residual oxygen and 28 per cent residual nitrogen). The time to the loss of SEPs, EEG suppression and the onset of an isoelectric EEG, respectively, were 22, 16 and 35 seconds in mixture A, 44, 41 and 101 seconds in mixture B, and 15, 15 and 67 seconds in mixture C. Stunning turkeys with mixture A or B would be suitable under commercial conditions. Mixture C, containing 65 per cent

The history and results of a clinical examination were recorded for 32 spring-born suckler calves which were hospitalised for intravenous fluid therapy. Blood samples were taken before treatment, during treatment and before discharge and analysed for colostral status, total
carbon dioxide
as an indication of acid-base status, and haematocrit. All the calves were given intravenously 5 to 10 litres of electrolyte solution containing 144 mmol/litre sodium, 4 mmol/litre potassium, 113 mmol/litre chloride and 35 mmol/litre bicarbonate, supplemented, in 24 calves, with up to 450 ml of 1M sodium bicarbonate. Nearly all the calves were recumbent but less than half were dehydrated on admission. The signs of dehydration were well correlated with each other and with the haematocrit. Neither the history nor the clinical signs were useful predictors of acidosis. There was no relationship between the severity of acidosis and the degree of dehydration. Acidosis was more prevalent in older calves (P < 0.01). For the severely acidotic calves, supplementary intravenous fluid with sodium bicarbonate significantly (P < 0.05) improved the total blood
carbon dioxide
at discharge. All 32 calves recovered. It is possible to treat acidotic calves with intravenous fluid therapy effectively, economically and according to their individual needs. The Harleco apparatus is a simple, useful, cost-effective adjunct to the diagnosis and treatment of this life-threatening condition.

stunner. At exsanguination, 96 blood samples (24/farm) were collected from pigs slaughtered through each slaughter handling system (384 samples in total). Serum samples were analysed for cortisol, creatine phosphokinase (CPK) lactate dehydrogenase (LDH) and LDH-isoenzymes. There were no significant differences between the systems at abattoir X. At abattoir Y, pigs slaughtered through the floor pen system had higher levels of LDH-5 (39.84 +/- 1.39 per cent of total LDH vs 34.76 +/- 1.21 per cent, P < 0.05) indicating greater skeletal muscle damage in the floor pen. Pigs at abattoir Y had higher mean serum levels of CPK than pigs at abattoir X (3.63 log10 U/litre vs 3.41 log10 U/litre, 0.03 sed, P < 0.001), higher total LDH activities (1269.5 U/litre vs 922.8 U/litre, P < 0.001) and higher cortisol concentrations (1.70 log10 ng/ml vs 1.51 log10 ng/ml, P < 0.001). These differences suggested that the levels of physical and psychological stress were higher at abattoir Y.

via benzoate and 4-chlorobiphenyl to 4-chlorobenzoate. In curing and conjugation experiments, the A5 endogenous 51-kb IncP1 plasmid pSS50 was found to be dispensable for biphenyl and 4-chlorobiphenyl catabolism. Transfer of the biphenyl- and 4-chlorobiphenyl-degrading phenotype by means of pSS50 was observed at a frequency of 10(-5) per transferred plasmid in matings of A5 with other A. eutrophus strains. Transconjugants harbor enlarged pSS50 derivatives which contain additional genetic information governing the oxidation of biphenyl and 4-chlorobiphenyl to benzoate and 4-chlorobenzoate and originating from the chromosome of strain A5. The following observations indicate that the catabolic genes reside on a 59-kb large transposon (Tn4371) for which a restriction map is presented. (i) Tn4371 transposes between different replicons and at different locations of the same replicon. (ii) Transposition was observed in a

Brucella organisms which differed from the recognised species of the genus, were isolated from nine seals, eight cetaceans and one otter. A method is described for the isolation of Brucella species from sea mammals and the first isolations of Brucella species are recorded from an Atlantic white-sided dolphin (Lagenorhynchus acutus), two striped dolphins (Stenella coeuleoalba), a hooded seal (Cystophora cristata), a grey seal (Halichoerus grypus) and a European otter (Lutra lutra).
