Over a period of seven years, 262 recumbent dairy cows were investigated, and serum samples were analysed for creatine phosphokinase (CPK),
lactate dehydrogenase
(
LDH
) and aspartate aminotransferase (AST) activities. The results from cows for which all three measurements were available, were analysed statistically by the non-parametric receiver operating characteristic (ROC) method, to evaluate the value of the serum enzyme activities for predicting a failure to recover. The sensitivity and specificity of the activity levels were calculated over all cut-off points, and ROC curves were created by plotting sensitivity as a function of 1--specificity at each cut-off point for samples grouped by the interval after the cows became recumbent. The predictive values of the tests were compared by calculating the areas under the curves, and the positive and negative predictive values of the tests were calculated and plotted against the prevalence of a failure to recover. The results from each of the enzymes were significantly better than from a random test with no discriminatory ability. The optimal cut-off points maximising the sensitivity and specificity of the tests were 2330, 2225 and 171 U/litre for CPK,
LDH
and AST, respectively. The predictive value of AST was significantly better than that of CPK or
LDH
, and measures made on the second and third day of recumbency were significantly better with optimal cut-off points of 128 and 189 U/litre, respectively.

PMID:Value of measurements of the serum activities of creatine phosphokinase, aspartate aminotransferase and lactate dehydrogenase for predicting whether recumbent dairy cows will recover. 1284 88
Over a period of 10 days, 17 dogs became weak and developed neurological deficits of different degrees of severity. About 12 hours before these clinical signs appeared they had all eaten a particular brand of commercial dog food from a recently opened bag. They were all quadriparetic and hyporeflexic, and some of them also showed additional systemic or neurological signs, including dyspnoea, a high body temperature, tongue laxity, hyperaesthesia and anisochoria. Serum biochemical abnormalities included high activities of creatine kinase,
lactate dehydrogenase
and aspartate aminotransferase. Analysis of the suspect food revealed high concentrations of the ionophore lasalocid. Fifteen of the dogs were given supportive treatment at home and two were hospitalised. Five of the dogs died, but the others improved gradually and had fully recovered by one to four days after the appearance of the clinical signs.

The aim of the study was to obtain basic data on individual biological variation, the required number of specimens to define the homeostatic setpoint (the aspired value of a variable adjusted to the individuals homeostasis) and critical differences of selected chemistry parameters in budgerigars (Melopsittacus undulatus). Blood from 99 healthy budgerigars was sampled 12 times at four-week intervals. Aspartate aminotransferase (ast), calcium, creatine kinase (ck), glutamate dehydrogenase (gldh), glucose,

(ldh), total protein and uric acid were investigated. The indices of individuality obtained in the present study were relatively low (total protein 0.93, ast 1.02, gldh 1.04, ck 1.12, ldh 1.24, uric acid 1.26, glucose 1.39, calcium 1.61) and suggest that population-based reference limits might be useful. Comparison of data showed that the application of intraindividual reference values identified much greater variation in the reference values than using conventional population-based reference limits. Otherwise, the moderate to low degrees of individuality may allow the use of reference values of one individual as a standard for another individual of the same species. The critical differences that define the change needed between two serial results to indicate a biological change was highest in gldh with 120 per cent or 1.7 U/l. In calcium, a change of 30.5 per cent or 0.5 mmol/l would be significant. In the other parameters critical differences varied between 38 per cent in glucose, up to 93 per cent in uric acid.

The differences between the capture stress responses of captive and free-ranging roe deer (Capreolus capreolus) and their modulation by acepromazine, during a period of three hours' physical restraint after capture in drive-nets, were examined in 16 free-ranging and 16 captive roe deer. Eight of the free-ranging and eight of the captive animals received acepromazine intramuscularly, and the other eight free-ranging and eight captive deer received the same volume of saline. Heart rate, body temperature and haematological and serum biochemical parameters were analysed. In the groups treated with acepromazine, the heart rate stabilised sooner, and the red blood cell (RBC) count, haemoglobin concentration, packed-cell volume, the serum activities of creatine kinase (CK), aspartate aminotransferase (AST), alanine aminotransferase (ALT) and
lactate dehydrogenase
(
LDH
) and the concentrations of creatinine and lactate were significantly lower, and serum glucose started to decrease earlier, than in the untreated groups. Serum potassium levels decreased over time only in the untreated groups. The body temperature stabilised earlier, and the RBC count, haemoglobin concentration, serum CK, AST, ALT and
LDH
activities, and serum creatinine, lactate, cholesterol and glucose concentrations were significantly lower in the free-ranging roe deer than in the captive deer.

PMID:Effect of acepromazine on the signs of capture stress in captive and free-ranging roe deer (Capreolus capreolus). 1752 95
Haematological and biochemical data were collected over a period of six years from grey seal (Halichoerus grypus) pups undergoing rehabilitation. Pups bled during the first three days were allocated retrospectively to one of seven clinical groups and statistical analyses were carried out on six of these groups (130 pups). Compared with reference ranges, all the groups had lower mean packed-cell volumes (pcv), red blood cell (rbc) counts haemoglobin and albumin levels, pups with severe trauma had higher total white blood cell (wbc) and neutrophil counts, pups with severe trauma and malnourished pups had higher total bilirubin levels, and pups in all the clinical groups except those with severe trauma had lower sodium levels. There were significant differences (P<0.05) between some clinical groups for mean pcv, rbc counts, all wbc counts except monocytes, haemoglobin, total bilirubin, creatine kinase, amylase and potassium levels. Forty-nine pups were bled more than once during the first 10 days of rehabilitation for haematology, and 11 were bled more than once for biochemistry. There were significant decreases (P<0.05) during this period in pcv, and in the activities of alkaline phosphatase, alanine aminotransferase, aspartate aminotransferase, creatine kinase and
lactate dehydrogenase
, and significant increases in all wbc counts, total protein, globulin, amylase and calcium levels.

This study was designed to investigate whether kits to measure circulating cardiac troponin-I (cTn-I) and cardiac troponin-T (cTn-T) can be used to determine myocardial cell damage in cattle with traumatic reticuloperitonitis (trp). Twenty cattle with trp were compared with 10 clinically healthy cattle. cTn-I and cTn-T were determined qualitatively and cTn-I was determined quantitatively; biochemical analyses were also performed on both groups. The mean serum concentrations of total protein, globulin, glucose and calcium, and the mean activities of creatine kinase mb, aspartate aminotransferase,
lactate dehydrogenase
and gamma-glutamyl transferase were higher in the cattle with trp than in the control group. The cTn-I and cTn-T kits both gave positive results in three of the cattle with trp and the quantitative measurement of cTn-I was positive in 11 of the trp cases. Both tests were negative in the healthy cattle.

The fasting and postprandial serum concentrations of bile acids and other blood constituents were measured in a group of 10 clinically healthy, female, six-year-old captive red-eared terrapins (Trachemys scripta elegans). The terrapins were housed in a temperate room and maintained in four aquaria in which the water temperature ranged from 24 to 27 degrees C and the temperature above the basking site ranged from 27 to 30 degrees C. The serum concentrations of bile acids were measured four times in a period of five months, and at the second sampling the fasting and two postprandial (after 24 and 48 hours) serum concentrations of total protein, albumin, glucose, uric acid, cholesterol, triglycerides, alkaline phosphatase, alanine aminotransferase, aspartate aminotransferase,
lactate dehydrogenase
, and bile acids were determined. Coelioscopy revealed vitellogenic and previtellogenic follicles on the ovaries of all the terrapins, and eggs with calcified shells were detected in two of them. The livers were mostly pink to deep yellow in colour, with sharp edges, a smooth serosal surface, distinct large superficial vessels, and multifocal melanin deposits. Liver biopsies revealed fine, more or less oil red O-positive lipid droplets in all the hepatocytes, but in none of the cases was it considered to be pathological lipidosis. The mean (sd) bile acid concentrations ranged from 7.35 (4.52) to 10.04 (7.40) micromol/l. The fasting and postprandial concentrations were 3.1 (2.3), 4.5 (5.4) (24 hours) and 2.2 (1.5) (48 hours) micromol/l. High concentrations between 27.6 and 66.6 micromol/l were associated with lipaemia. There were no significant differences between the biochemical profiles of the fasting and postprandial serum samples.

Samples of peritoneal fluid and jugular venous blood were taken simultaneously from 95 clinically healthy Holstein-Friesian cows. The concentrations of total protein, albumin, glucose, cholesterol, fibrinogen, L-lactate and D-dimer, the activities of
lactate dehydrogenase
(
LDH
), alkaline phosphatase and creatine kinase, and the white blood cell count were determined in the samples. Light's criteria, the serum-ascites albumin gradient (SAAG) and the ratios of the concentration of each parameter in peritoneal fluid to its concentration in blood were calculated. The mean concentrations of total protein, albumin and D-dimer, the activity of
LDH
and the SAAG were different from the reference values for monogastric animals and human beings.

Regeneration gene protein 2 (Reg-2) is a small secreted protein expressed in motor and sensory neurons of spinal cord during developmental stages and following injury of peripheral nerves. Reg-2 appears to act as a neurotrophic factor and protects injured neurons from death during regeneration. To illustrate these potential protective effects in vitro, we investigated the blocking effects of Reg-2 antibodies on the survival of primary cultured spinal cord neurons and astrocytes, as well as on neurite outgrowth. In addition, the effects of Reg-2 in neuron injury models induced by peroxide and mitochondrial poisoning were assessed. Our results showed that Reg-2 antibody markedly reduced survival and neurite outgrowth from neurons, whereas astrocyte survival was unaffected. Addition of Reg-2 into the culture medium had no effect on neuron survival or neurite outgrowth. However, the addition of the Reg-2 into culture media after peroxide treatment or cellular hypoxia insult induced by mitochondrial poisoning can reduce
lactate dehydrogenase
release levels and cell death. Thus, the data suggests that Reg-2 is essential for the survival and neurite outgrowth of developing spinal cord neurons but not the survival of glial cells, and that Reg-2 plays protective effects on spinal cord neurons against injury in vitro.
