Repeated ultrasonographic examinations and collections of blood samples and uterine lumenal swabs between seven and 28 days after calving were used to examine the relative effects of bacterial contamination and involution of the uterus on the concentrations of acute phase proteins in the blood of 26 dairy cows. The severity of bacterial contamination, as determined by the total bacterial growth score, was a significant variable for the concentrations of the acute phase proteins alpha1-acid glycoprotein (P < 0.0001), haptoglobin (P < 0.05) and ceruloplasmin (P < 0.0001). In addition, the concentrations of alpha1-acid glycoprotein and ceruloplasmin were increased in the cows from which Escherichia coli (P < 0.0001) and Arcanobacterium pyogenes (P < 0.05), respectively, were isolated from the uterine lumen. Uterine involution, as determined by the decreasing diameter of the previously gravid uterine horn, was associated with a decrease in the concentrations of alpha1-acid glycoprotein (P < 0.005), haptoglobin (P < 0.05) and ceruloplasmin (P < 0.01). However, the response of the acute phase proteins to bacterial contamination was independent of the day on which the samples were collected, indicating that their concentrations were increased by bacterial contamination in addition to the changes associated with uterine involution.

This study assessed the effects of suckled sucrose and parenteral carprofen treatment on behavioural measurements of discomfort and serum haptoglobin concentration in lambs following rubber ring castration and/or tail docking. Twenty-eight male and 20 ewe lambs were allocated to either a male or ewe lamb control group (n=4) or to one of three male and two ewe lamb treatment groups (n=8). Male lambs in one treatment group received carprofen subcutaneously (0.5 mg/kg) 30 minutes before castration and docking. Lambs in all other treatment groups suckled sucrose or colostrum immediately before castration and/or tail docking. Behavioural measurements of discomfort were made following castration or tail docking in treatment groups and also in control animals which were not castrated or tail docked. Blood sampling of animals in treatment groups for analysis of serum haptoglobin, an acute phase protein used as an indicator of an acute inflammatory response, was performed before castration or docking and at 24 and 48 hours after castration or docking. Control lambs were blood sampled at 0, 24 and 48 hours following behavioural assessment Neither suckled sucrose nor carprofen treatment altered discomfort behaviour associated with castration or tail docking. Haptoglobin levels following castration or tail docking remained close to the detection limits of the assay and were similar to those recorded in control animals.

The concentrations of haptoglobin, C-reactive protein and ceruloplasmin were measured in symptomatic and asymptomatic dogs naturally infected by Leishmania infantum, and in healthy uninfected dogs to determine the potential value of these proteins for the diagnosis and prognosis of leishmaniasis. The concentrations of the acute phase proteins were significantly higher in the dogs with leishmaniasis than in the control dogs, and the concentration of C-reactive protein was significantly higher in the symptomatic dogs than in the asymptomatic dogs. There were no correlations between the acute phase proteins and the gamma globulins, the albumin/globulin ratio or the titre of anti-leishmanial antibodies.

A case-control study of 340 finishing pigs aged 10 to 25 weeks in 15 commercial Danish pig herds was carried out to investigate serum haptoglobin concentration as an objective marker of clinical signs of disease. Pigs with different clinical signs were matched to control pigs without clinical signs with respect to herd, pen, estimated weight and gender, and each pig was subjected to a standard clinical examination. In 86 of the case-control pairs, the rectal temperature was also recorded. There was a significantly higher mean haptoglobin concentration in the serum of lame pigs (P<0.0001), pigs with respiratory disease (P=0.0004), pigs with tail or ear bites (P=0.0004) and pigs with diarrhoea (P=0.02). Similarly, a higher mean rectal temperature was recorded in lame pigs (P<0.0001), pigs with respiratory disease (P=0.002) and pigs with tail or ear bites (P=0.0003). There was a significant but low correlation between rectal temperature and haptoglobin concentration in serum (P=0.003, r=0.20). The area under the receiver-operating-characteristic curve was between 0.67 and 0.78 for the different clinical signs. The maximum simultaneous levels of sensitivity (0.61 to 0.71) and specificity (0.61 to 0.77) of serum haptoglobin for the different clinical signs were obtained at a cut-off value of 1.1 mg/ml. At a cut-off value of 1.8 mg/ml, the sensitivity decreased to 0.31 to 0.60, and the specificity increased to 0.82 to 0.86. It was not possible to define a cut-off value which classified individual pigs according to their clinical signs.

The concentrations of the two acute phase proteins, serum amyloid A and haptoglobin, in serum and milk were compared in 10 cows with clinical mastitis, 11 cows with extramammary inflammatory conditions and 10 clinically healthy control cows. The concentrations of both acute phase proteins were higher in the serum and milk of the cows with mastitis than in the cows in the other two groups. Four of the cows with extramammary inflammatory conditions had serum amyloid A concentrations in serum above 100 microg/ml, but negligible concentrations in milk, indicating that a pathogen must be present in the mammary gland for serum amyloid A to accumulate in milk. The acute phase protein concentrations in milk increased significantly with increasing somatic cell count, suggesting that they may be indicators of the severity of an infection.

As part of a study of the development of hoof horn haemorrhages in first-lactation heifers, measurements were made of acute phase reactants to investigate the link between the acute phase response and the development of the haemorrhages. Over a period of two years, blood samples were taken from two separate groups of heifers, weekly in the three weeks before they calved and then twice weekly until eight weeks after calving. Plasma total protein, albumin, fibrinogen, haptoglobin, seromucoid and serum iron and caeruloplasmin were measured and the relationships between the peak concentration (or activity) or the area under the curve of each acute phase reactant and the peak scores for sole or white line haemorrhages were assessed by linear regression. The results suggested that the development of the hoof horn haemorrhages observed in the study was not accompanied by an acute phase response, and the haemorrhages were therefore not primarily caused by endotoxicosis. The diets and husbandry systems used were typical of dairy farms in the UK and the results therefore suggest that a significant proportion of hoof horn haemorrhages observed in UK dairy cows may not be caused by endotoxicosis.

The serum concentrations of haptoglobin, caeruloplasmin, C-reactive protein and serum amyloid A were measured in three groups of seven healthy dogs. Group 1 received a single dose of 1.1 mg/kg methylprednisolone acetate, administered subcutaneously; group 2 received 1 mg/kg per day of prednisone administered orally for three weeks; and group 3 received 2.2 mg/kg per day of prednisone administered orally for seven days. Before the administration of the glucocorticoids the serum concentrations of all the acute phase proteins were within the authors' laboratory reference ranges. After the administration of the drugs there were significant increases in the concentration of haptoglobin in all three groups, the increases being larger in groups 2 and 3. In contrast, the concentrations of C-reactive protein, caeruloplasmin and serum amyloid A were not affected.

The serum concentrations of serum amyloid A, haptoglobin and fibrinogen were measured in a group of horses before and at intervals after elective and non-elective surgery, and in a control group of normal horses. There was a significant, rapid and repeatable increase in the concentration of serum amyloid A in response to both elective and non-elective surgery. In the control horses its serum concentration was within the normal range, from 0 to 0.2 microg/ml. Twenty-four hours after elective surgery its mean peak concentration was 16.4 microg/ml, and after non-elective surgery it was 27.3 microg/ml. In contrast, the serum concentrations of haptoglobin and fibrinogen increased more slowly after surgery and had not decreased by 72 hours after surgery.

The prevalence of lameness among 646 sows and gilts in 21 selected herds was determined; 8.8 per cent of the animals were lame and the most common clinical diagnoses were osteochondrosis, infected skin lesions and claw lesions. The lame animals had higher serum concentrations of haptoglobin and C-reactive protein than the sound animals. Animals housed on slatted floors had twice the odds of being lame and 3.7 times the odds of being severely lame than animals housed on solid floors. Yorkshire pigs had 2.7 times the odds of being lame than Landrace or crossbred animals. Higher parity and the use of roughage decreased the odds of the sows not becoming pregnant; however, lameness was not a risk factor for non-pregnancy.

Six healthy bitches underwent ovariohysterectomy, performed by medial laparotomy according to routine methods. Blood samples were taken by venepuncture just before and 24, 48 and 72 hours and seven days after surgery. The serum C-reactive protein (CRP) (P<0.001), haptoglobin (P<0.001) and caeruloplasmin (P<0.05) levels were all significantly higher in the postoperative period than in the samples taken before surgery. The CRP levels increased rapidly and dramatically, while the levels of haptoglobin and caeruloplasmin rose more gradually.
