We fused spleen lymphocytes obtained from mice immunized against a human melanoma cell line and melanoma-mouse hybrid cells with the P3 X 63 Ag8 mouse myeloma in order to produce hybrids secreting antibodies against a human melanoma. Antibodies secreted by individual hybrids were tested for their reaction with a panel of human melanoma, colorectal carcinoma, and normal cells in an indirect radioimmunoassay, and they displayed different specificities and crossreactivities. Some reacted only with melanomas, whereas others crossreacted with normal human or human colorectal carcinoma cells. By analysis of competitive binding of mixtures of monoclonal antibodies, it was possible to delineate different epitopes on melanomas. Hybrids growing in nude mice and producing antimelanoma antibody suppressed growth of melanoma tumors.

When human epithelial FL cells were fused with lymphoid cells harboring Epstein-Barr virus (EBV) genome, EBV-determined nuclear antigen (EBNA) immunofluorescence became evident in high frequency in FL nuclei of heterokaryons several days after fusion. The implications of the findings are discussed in relation to the presence of EBV genome in nasopharyngeal carcinoma cells.

Hybridization of Ehrlich ascitic carcinoma cells and chicken fibroblasts using UV-inactivated Sendai virus produced the following variants of homo- and heterokaryons: (a) uninfected Ehrlich cells, (b) fowl plague virus-infected chicken fibroblasts, (c) uninfected Ehrlich cells and uninfected chicken fibroblasts; (d) infected Ehrlich cells and infected chicken fibroblasts, (e) infected Ehrlich cells and uninfected chicken fibroblasts. The analysis of the material produced by heterokaryons gave the following results: when Ehrlich ascitic carcinoma cells infected with classical fowl plaque virus and labeled with 3H-uridine fused with uninfected unlabeled chicken fibroblasts, the heterokaryons, in addition to structures with the buoyant density of 1.29 g/ml typical for infected cells, also produced virus structures with a density of 1.25 g/ml and had a higher infectious activity (5.2 X 10(7) PFU/ml) than the structures produced by infected Ehrlich cells (5.8 X 10(6) PFU/ml); after fusion of Ehrlich ascitic carcinoma cells infected with fowl plague virus and labelled with 3H-uridine with unlabelled chicken fibroblasts infected with fowl plague virus heterokaryons produced virus structures with densities of 1.29 g/ml and 1.22 g/ml which had even higher infectious activity (8.6 X 10(8) PFU/ml) than the structures produced by infected Ehlrich ascitic cells.

Four independently fused hybrid clones derived from a cross between the mouse mammary carcinoma TA3Ha and the human Burkitt lymphoma line Daudi were tested for the EBV-determined nuclear antigen (EBNA), EBV-DNA and the presence of human chromosomes, in the course of serial propagation in vitro. EBNA and EBV-DNA were lost in parallel with the loss of human chromosomes. It seems that the persistence of EB-viral genetic information does not require the presence of a specific human chromosome(s) in this particular hybrid combination.

When horse-radish peroxidase (HPO) was administered iv to Vx2 carcinoma-bearing rabbits, an HPO reaction product was detected in the lumina of blood vessels and the extracellular spaces between tumor cells in the first few minutes after administration. HPO was also seen in vesicles in tumor cells. Fifteen minutes to 1 hour after administration, the HPO reaction product was found mainly in the large membrane-bound vacuoles. Within 6--12 hours, the HPO activity gradually diminished in large membrane-bound vacuoles (lysosomes). In conclusion, exogenous HPO was rapidly incorporated into Vx2 carcinoma cells by pinocytosis, and then pinocytotic vesicles were fused with lysosomes.

Mouse NS-1 myeloma cells were fused with spleen cells from mice that had been immunized with cells from a human melanoma, M1804. Hybrid cells were grown in selective medium and tested for production of antibody to surface antigens of M1804 cells. Three hybrids that produced antibodies that bound to the melanoma cells but not to autologous skin fibroblasts were cloned. Antibodies produced by two of the clones were cytotoxic to M1804 cells in the presence of rabbit complement. Extensive specificity tests showed that the antibodies produced by the clones bound strongly only to M1804 cells; significant, although weaker, binding occurred with 2 of 11 allogeneic melanomas. Apart from weak binding of the antibody produced by one of the clones to a breast carcinoma, binding assays of five carcinomas, one sarcoma, and fibroblasts from 17 individuals were negative, as were cytotoxic tests of 10 lymphoblastoid cell lines and peripheral blood lymphocytes from 68 normal donors and 12 chronic lymphocytic leukemia patients. This suggests that we have identified one or more determinants of a melanoma-associated antigen(s), whose expression is limited to a small proportion of melanomas.

A man with crossed, non-fused renal ectopia had carcinoma of the distal ureter, leading to hydro-ureter, hydronephrosis and urinary infection. Crossed ectopic kidneys are more disease-prone than are normal kidneys. Complicating disorders are discussed, the value of thorough urologic investigation is stressed and the incidence of tumour of the upper urinary tract in crossed renal ectopia without fusion is reviewed.

A hitherto undescribed type of the rare "collision" tumours is reported: necropsy in a 77-year-old male patient revealed liposarcoma of the right seminal vesicle in association with anaplastic carcinoma of the prostate. The carcinoma was fused with the liposarcoma as a result of its invasive local growth and had formed deposits in the lymph nodes, bones and lungs. The left seminal vesicle showed localized amyloidosis. A review of the published cases of mesenchymal tumours of the seminal vesicles is presented.

An 8-year-old male Irish Terrior dog and a male St. Bernard dog each had a thyroid medullary carcinoma. The first dog was hypocalcemic before removal and with recurrence of the neoplasm. In both dogs the neoplasms were bilateral and microscopic metastases were seen only in the regional lymph nodes. Both neoplasms were characterized by a solid alveolar pattern separated by thin vascular stroma or a linear festoon-like arrangement with thick stroma. The neoplastic cells varied little and were polygonal or round with abundant pale eosinophilic and finely granular cytoplasm. There was little mitotic activity. Both carcinomas contained amyloid. Electron microscopically, the neoplastic cells had moderate nuclear pleomorphism, well developed Golgi complexes, and characteristic membrane-bound electron-dense secretory granules. In some sections, the secretory granule membranes were fused to the cytoplasmic membrane and the granules were in the extracellular space. In one carcinoma there were linear fibrillar structures with an average width of 7.8 nanometers similar to that of amyloid fibrils.

Malignant tumors induce angiogenesis and modulation of microvasculature. Based on histologic and immunohistochemical analysis of human surgical material, we describe here the occurrence of glomeruloid structures in gastrointestinal carcinomas, and compare them with the microvasculature in inflammatory granulation tissue. The glomeruloid structures were composed of clusters of mutually fused capillaries with prominent swelling of endothelial cells and pericytes. They were thought to be specific for glioblastoma of the brain. The glomeruloid structures were observed juxtaposed to carcinoma nests in one-third of gastric carcinoma of intestinal type and colorectal carcinoma in the area of invasive growth beyond the muscularis mucosae. They were not observed in gastric carcinoma of diffuse type, intramucosal carcinoma, or inflammatory granulation tissue. The glomeruloid structures can be regarded as an extreme example of endothelial hyperplastic changes observed in cancer stroma. Our results suggested that glomeruloid structures can occur in carcinomas as vascular reaction, a mechanism different from that in inflammatory granulation tissues.
