Female rats were injected subcutaneously with ethionine, and enzymic activities of liver membranes (Na+-k+-stimulated

, glucose-6-phosphatase, NADPH: cytochrome c oxido-

and NAD-nucleosidase) examined at proper intervals, during the intraperitoneal treatment of an egg phospholipid preparation (EPL). It is shown that EPL is unable to overcome the enzymic changes due to severe ethionine treatment, but is able to facilitate the recovery times after drug withdrawal for all the enzymic activities, except for NAD-nucleosidase. At lower dosage of the drug, the ethionine treatment is able to prevent the observed change of the glucose-6-phosphatase activity but not that of the Mg2+-ATPase. It is suggested that the EPL treatment may modify the chemical composition ahd/or architecture of liver membranes, altered by the ethionine injection, thus acting, at least partially, on the enzymic changes.

Cellular membranes were prepared from the non-extending part of dark grown hypocotyls of Phaseolus aureus. The relative effectiveness of continuous and discontinuous sucrose gradient centrifugation for the separation of membranes was investigated. Characteristic densities of membranes were determined by the localization of enzyme activities on continuous sucrose gradients: NADH-cytochrome c-

for endoplasmic reticulum, beta-1-3-glucan synthetase for plasma-membrane and IDPase for dictyosomes. The difficulties involved in the application of

and IDPase as specific membrane markers are discussed. Negative staining of isolated fractions indicated that intact dictyosomes could be prepared from this tissue without the use of chemical fixatives in the homogenization medium. Extraction of isolated membranes showed that carbohydrate-binding proteins (lectins) were present both in an easily removable and in a more strongly bound form. In vivo incorporation of D-[U-14C]glucose and subsequent isolation and solubilization of the different membranes showed that sugar-containing polymers could be released without hydrolytic techniques and were present in the equivalent extracts that exhibited lectin activity. The possibility of lectin-polysaccharide complexes in endoplasmic reticulum and dictyosomes and their involvement in the synthesis and transport of secretory substances by the membranes is discussed.

The crude protein levels as well as the activities of various enzymes were studied in certain tissues of fetuses (80th through 114th days of development), piglets of different age groups, and pigs for slaughter. In most of the tissues tested the postnatal activities of Na-K-

The present work is a continuation of our studies on mitochondrial functions and enzyme activities after acute exhaustive swimming in liver and myocardium. In rat heart mitochondria the activities of SDH, cytochrome oxidase and

(DNP-stimulated) increase after swimming and remain at that level until the end of the 22-hour rest period studied. The enzyme complexes--rotenone-sensitive NAD. H-cytochrome c-

and succinate-cytochrome c-
reductase
--decrease their activities in both experimental groups. The reduced activity of these two enzymes is determined by changes in this part of the respiratory chain which occur after the incorporation of DCPIP in the oxidation-reduction processes. --reveals unchanged activity after swimming and a 22-hour period of rest. The different changes in the activities of enzymes with different localization and organization in heart mitochondria are explained by disorganization of the inner membranes after exhaustive swimming, which could induce both activation of some enzymes and inhibition of others. The effect of certain factors during muscle exercise which could cause the established structural and functional changes in the mitochondria is discussed.

A sporadic case of central core disease in a 5 1/2-year-old girl is reported. Clinically, a retarded motor development existed, furthermore, a muscle weakness and hypotonia of the extremities and trunk, contractures of the hip- and knee-joint,and luxation of both hip-joints. Biopsy specimens are taken from both Mm. gastrocnemii. Muscle fibres show, by morphologic examination, 95 per cent cores, which are characteristic for this myopathy.
