Muscle samples were obtained from the gastrocnemius of 17 female and 23 male track athletes, 10 untrained women, and 11 untrained men. Portions of the specimen were analyzed for total phosphorylase, lactic dehydrogenase (LDH), and succinate dehydrogenase (SDH) activities. Sections of the muscle were stained for myosin adenosine triphosphatase, NADH2 tetrazolium reductase, and alpha-glycerophosphate dehydrogenase. Maximal oxygen uptake (VO2max) was measured on a treadmill for 23 of the volunteers (6 female athletes, 11 male athletes, 10 untrained women, and 6 untrained men). These measurements confirm earlier reports which suggest that the athlete's preference for strength, speed, and/or endurance events is in part a matter of genetic endowment. Aside from differences in fiber composition and enzymes among middle-distance runners, the only distinction between the sexes was the larger fiber areas of the male athletes. SDH activity was found to correlate 0.79 with VO2max, while muscle LDH appeared to be a function of muscle fiber composition. While sprint- and endurance-trained athletes are characterized by distinct fiber compositions and enzyme activities, participants in strength events (e.g., shot-put) have relatively low muscle enzyme activities and a variety of fiber compositions.

The experiments were carried out in mice which were divided into 4 experimental groups and a control group. In the course of the experiment SD, NADH2-tetrazolium reductase; ATP-ase, G-6-P-ase and AP were observed. It was found that acute benzene intoxication causes the disturbances in the enzymatic activities of the cells of the main segment of the nephron. The impairment of tissue respiration and oxygen phosphorylation and of active transport is due to benzene intoxication. Benzene leads to injury of the endoplasmatic reticulum in the cells of the kidney.

Enzyme activity of adenosinetriphosphatase, NADH2-tetrazolium reductase and lactate dehydrogenase was studied in the rat perineurium 10, 20, 30, and 90 days post partum. The effect of under-nutrition was shown by a lack of enzyme activity in rats 30 days post partum in contrast to control animals, 30 days also corresponds to the time when, in normal rats, the perineurial diffusion barrier to exogenous macromolecules becomes established. In undernourished rats this diffusion barrier does not appear. At 90 days post partum raised enzyme activities indicate a high metabolic turnover in the perineurium even in animals with undernutrition.

Male guinea pigs were exposed to nitrogen dioxide (2 mg/m3) during 180 days (8 hours a day). Long-term exposure induced thickening of the corneal layer of the epidermis as well as inflammatory infiltrations in the proper skin. The following enzymes were estimated histochemically in skin samples of experimental and control animals: succinic dehydrogenase, NADH2-tetrazolium reductase, lactate dehydrogenase; alkaline phosphatase, acid phosphatase and adenosine triphosphatase. Chronic exposrue stimulated a decrease of NADH2-tetrazolium reductase in the epidermis and connective tissue components of proper skin and marked positive reaction of lactate dehydrogenase in epidermal cells and hair follicles. Increase of a diffuse reaction on adenosine triphosphatase in smooth muscles of the skin was found also in exposed animals.

The experiments were carried out on dogs. Experimental animals were subjected to the trauma of the thorax during operation. The localization and activity of succinic dehydrogenase, NADH2-tetrazole reductase, adenosine triphosphatase, alkaline and acid phosphates in the kidneys were examined. An increase of the activity of all the investigated enzymes takes place under the influence of the stress. On the basis of the investigations it can be supposed that the processes of oxygen phosphorylation and active transport are intensified. An increase of the activity of acid phosphates gives evidence of the intensity of phagocytosis and pinocytosis processes in the kidney.

1. Reduction of 4-nitrobenzoic acid by the nematode Ascaris lumbricoides var suum occurred in the intestinal brush border cells. Cuticle, mesenchyme fluid and reproductive tissue showed no activity. 2. 4-Nitrobenzoic acid reductase was in the supernatant fraction after centrifugation of intestinal homogenates at 75 000 g for 2 h. 3. The enzyme required as cofactors NADH2, a low molecular weight thiol such as glutathione or cysteine and a divalent metal ion such as Mn++ or Cu++. The reduction was not catalysed by NADPH2. 4. Reduction was not inhibited by O2 or CO. Optimum pH of the reaction was about 6.5.

By means of histochemical methods, folic acid, dihydrofolate reductase and NADH2-cytochrome-C-reductase were studied in the bovine superior cervical ganglion, in parallel with quantitative estimations of dihydrofolate reductase activity and in connection with the process of ageing. Various levels of folate metabolism were present in nerve cells and glial cells, as well as in pre or postganglionic nerves. In the process of ageing the activity of dihydrofolate reductase gradually decreased and the folic acid concentration in the nerve cells increased. Thus the enzyme --- substrate ratio appeared to favour the enzyme in young animals but the substrate in old animals.

The effect of ubiquinones with different length of their chain (CoQ0, CoQ1, CoQ2, CoQ6, CoQ9) and their synthetic analogues (analogues of ubiquinone-1, hexahydroubiquinone-4, monophytylquinone, diphytylquinone, triphytylquinone) on the activity of ubiquinone dependent enzyme systems was studied in mitochondrial fractions from the yeast Candida guilliermondii. All of the ubiquinone homologues studied activated these systems. The synthetic analogues of ubiquinone nonspecifically inhibited the activity of NADH2-oxidase system. The inhibition was reversible when CoQ0 and CoQ1, but not CoQ6 and CoQ9, were added to the system. In the succinate-CoQ-reductase system, the inhibition caused by the analogues of ubiquinone was eliminated when all of the tested homologues were added to the system. In contrast to other analogues of ubiquinone, hexahydroubiquinone-4 was an inhibitor for the NADH2-oxidase system and an activator for the succinate-CoQ-reductase system, and eliminated the inhibiting action of other ubiquinone analogues in this system. Similar action of ubiquinone homologues was shown in the elimination of the inhibition of ubiquinone dependent systems caused by the specific inhibitors of electron transport, viz. rotenone and antimycin A.

Histochemical and biochemical studies on the folate metabolism (folic acid an its principal enzyme-dihydrofolate-reductase) in bovine cortex - gyrus marginallis in the process of ageing were performed, in parallel with NADH2-cytocrom-C-reductase (diaphorase). Folic acid and folate enzyme, weak positive in neurons in young age, increased in old age in nerve cells and especially in their processes and in capillaries. The diaphorase strongly increased in all cells, glia and vessels, in old age.

In bovine caudate nucleus of young animal, a folic acid-positive reaction was found in the perikarya and in the neuroglia in parallel with a high dihydrofolate reductase activity in the nerve cells. In old animals, folic acid increased in neurons, neuroglia and some nerve cell processes; the folate enzyme was markedly decreased in neurons and increased neuroglia NADH2-cytochrom-C-reductase activity was strongly positive in nerve cells in young and old animals.
