Abnormal activity of the cardiac Na+/Ca2+ exchanger (NCX1) can affect intracellular Ca2+ homeostasis and cause arrhythmias. The n-3 polyunsaturated fatty acids (PUFAs), however, may prevent arrhythmias. To test the effect of PUFAs on the cardiac NCX1 current (I(NCX1)), the canine NCX1 cDNA was expressed in human embryonic kidney (HEK293t) cells. The average density of I(NCX1) was 10.9+/-2.6 pA/pF (n=44) in NCX1-transfected cells and eicosapentaenoic acid (
EPA
, C20:5n-3) significantly inhibited I(NCX1) The suppression of I(NCX1) by
EPA
was concentration-dependent with an IC50 of 0.82+/-0.27 microM.

had a similar effect on outward or inward I(NCX1). Docosahexaenoic acid (DHA, C22:6n-3) and arachidonic acid (AA, C20:4n-6) also significantly inhibited I(NCX1), whereas the saturated fatty acid, stearic acid (SA,

:0), did not. Our data demonstrate that the n-3 PUFAs significantly suppress cardiac I(NCX1), which is probably one of their protective effects against lethal arrhythmias.

The aim of this study was to characterize and to evaluate the lipidic composition of mexican marine fishes with special emphasis in n-3 fatty acids as nutraceuticals. The edible portion of 25 species: humidity (H), crude protein (CP), total lipids (TL) and fatty acids (FA). The average content (g/100g edible portion) of H was 75.20, PC was 18.40, TL was 3.60. Four n-3 FA were identified in all the samples and they were found in the next abundance order (mg/100g edible portion): C22:6n-3 (DHA)(229.60), C20:5 n-3 (

)(52.10),
C18
:3 n-3 (ALA)(11.80) and C20:3 n-3 (2.25). By their origin and climate there were no difference. By their biologycal classification, n-3 FA content was higher in bony fishes than cartilaginous fishes. It was detected a proportional relation with the n-3 FA concentration and total lipid content. According to their ecotic distribution there were numerical differences in DHA content (mg/100g edible portion) between pelagics (420.70), benthopelagics (125.30) and demersals fishes (225.40). Fatty fishes had higher content of
EPA
and DHA (mg/100g edible portion) (109.27 and 552.72) than semifatty fishes (56.12 and 226.29) and leanness (15.95 and 96.52), respectively. Bony, fatty and pelagic fishes had a higher content of EPA+DHA. According with the international recommendation values (200 to 600 mg EPA+DHA/day) the 44% of the analyzed species could be considered as functional foods due to their high content of

To estimate in vivo alpha-linolenic acid (ALA;

:3n-3) conversion, 29 healthy subjects consumed for 28 days a diet providing 7% of energy from linoleic acid (
C18
:2n-6) and 0.4% from ALA. On day 19, subjects received a single bolus of 30 mg of uniformly labeled [(13)C]ALA and for the next 8 days 10 mg twice daily. Fasting plasma phospholipid concentrations of (12)C- and (13)C-labeled ALA, eicosapentaenoic acid (
EPA
; C20:5n-3), docosapentaenoic acid (DPA; C22:5n-3), and docosahexaenoic acid (DHA; C22:6n-3) were determined on days 19, 21, 23, 26, 27, and 28. To estimate hepatic conversion of n-3 fatty acids, a tracer model was developed based on the averaged (13)C data of the participants. A similar tracee model was solved using the averaged (12)C values, the kinetic parameters derived from the tracer model, and mean ALA consumption. ALA incorporation into plasma phospholipids was estimated by solving both models simultaneously. It was found that nearly 7% of dietary ALA was incorporated into plasma phospholipids. From this pool, 99.8% was converted into
EPA
and 1% was converted into DPA and subsequently into DHA. The limited incorporation of dietary ALA into the hepatic phospholipid pool contributes to the low hepatic conversion of ALA into
EPA
. A low conversion of ALA-derived
EPA
into DPA might be an additional obstacle for DHA synthesis.

Cutaneous mast cells are considered as key immune effectors in the pathogenesis of canine atopic dermatitis (CAD). These cells release immediate-phase and late-phase mediators of inflammation. Dietary fatty acids are incorporated in cellular membranes and seem to influence mediator production and release. A dietary intervention with n6- and n3-fatty acids is thought to alleviate clinical symptoms in atopic dogs. The purpose of this study was to examine the effects of n6- and n3-fatty acids on the fatty acid composition of canine mastocytoma cells (C2) as a possible model for CAD. However, Delta5-desaturated products were not measurable. These results let us assume that C2 has no measurable activity of the Delta5-desaturase. In case the low activity of Delta5-desaturase is one of the mechanisms underlying the pathogenesis of CAD, C2 seems to be an adequate model for investigations in CAD.

-modified carbon-paste electrode has been used to determine voltammetrically endosulfan, despite its unfavourable electrochemical properties and behaviour. The methodology proposed is based on the decrease experienced by the peak intensity corresponding to voltammetric signals of Cu(II) when successive and constant additions of endosulfan are carried out. This decrease is directly proportional to the concentration of endosulfan what allows to perform an indirect quantification of the pesticide. The method uses sonication, by means of an ultrasonic probe, as a new tool for assisted extraction, coupled with reversed-phase liquid chromatography (RP-LC) with fluorescence detection (FL) for determination of 16 US

priority PAH. Separation and detection of the 16 PAH were complete in 45 min by RP-LC with a

column and acetonitrile-water gradient elution. Multivariate optimisation of the variables affecting extraction (ultrasound radiation amplitude, sonication time, and temperature of the water-bath in which the extraction cell was placed) was conducted. The accuracy of the method was determined by analysis of a certified reference material and comparison of the results obtained with those from another method (microwave-assisted extraction and GC-MS). The new technique avoids the main problems encountered in the determination of PAH in complex matrices such as marine biota, and no clean-up step is necessary. Diets rich in monounsaturated fatty acids (MUFA) are recommended for individuals with type 2 diabetes mellitus (T2DM). The American Heart Association recommends increasing intakes of n-3 polyunsaturated fatty acids (PUFA) to reduce the risk of vascular disease in high-risk individuals; however, the long-term effects of these bioactive fatty acids on glucose metabolism in insulin resistance are controversial. The present studies were conducted to evaluate the effects of diets rich in both MUFA and alpha linolenic acid (
C18
:3n-3, ALA), eicosapentaenoic acid (C20:5n-3,
EPA
), or docosahexaenoic acid (C22:6n-3, DHA), on glycemic control and other parameters related to vascular health in a mouse model of T2DM and insulin resistance. Male ob/ob mice (n = 15 per treatment) were fed 1 of 4 lipid-modified formula diets (LFDs) for 4 weeks: (1) MUFA control, (2) ALA blend, (3)
EPA
blend, and (4) DHA blend. A portion of a MUFA-rich lipid blend in the control LFD was replaced with 11% to 14% energy as n-3 PUFA. After 4 weeks, plasma glucose response to a standard meal (1.5 g carbohydrate/kg body weight) and insulin challenge (2 U/kg body weight, IP) was assessed, and samples were collected for analysis of glucose, insulin, and lipids. Vascular reactivity of isolated aortic rings was assessed in an identical follow-up study. The results showed that insulin-resistant mice fed an LFD with
EPA
and/or DHA blends had significantly (P < .05) lower triglycerides and free fatty acids, but insulin sensitivity and fasting plasma glucose were not improved. However, mice fed with the ALA blend had significantly improved insulin sensitivity when compared to those fed with other LFD (P < .05). Animals fed an LFD with n-3 PUFA from marine or plant sources showed significantly improved vascular responses as compared with the MUFA-rich LFD (E(max), P < .05) and ob/ob reference mice consuming chow (E(max) and pEC(50), P < .05). In summary, long-term consumption of LFD with n-3 PUFAs improved blood lipids and vascular function in an animal model of insulin resistance and T2DM; however, only MUFA-rich LFD with ALA also improved both insulin sensitivity and glycemic responses. Further studies of MUFA-rich LFD with ALA with individuals who have T2DM are warranted.

, C20:5]; and omega-9 series: oleic acid [OLE,

:1]), or two saturated fatty acids (stearic acid [STA,

:0] and arachidic acid [AD, C20:0]) were used to examine the effects on calcium response and degranulation from RBL-2H3 cells. Calcium response was monitored using the fluorescent calcium indicator fura-2, while degranulation was monitored by measuring histamine release from the cells. Three omega-6 PUFAs (AA, alpha-LN and LA) dose-dependently increased the cytosolic free-calcium concentration and histamine release from RBL-2H3 cells. This phenomenon was specific to the omega-6 PUFAs, the omega-3 PUFAs (alpha-LA and
EPA
), omega-9 PUFA (OLE) and the saturated fatty acids (STA and AD) had no effect. The increase in the cytosolic free-calcium concentration caused by the omega-6 PUFAs depended on the existence of external calcium, cell viability and the cellular IP(3) levels remained unchanged throughout the experiment. These results suggest that omega-6 PUFAs work as direct mediators of calcium signaling pathways in RBL-2H3 cells.
