We have recently shown that
CD99
(MIC2) is differentially expressed during normal early B-cell development in the bone marrow (BM). Since immature B-cell precursors (BCP) are assumed to correspond to some extent to acute lymphoblastic leukaemia (ALL) and
non-Hodgkin's lymphoma
(
NHL
) cells with respect to patterns of phenotypic differentiation, we wondered whether the particular maturation-associated expression patterns of
CD99
in the normal BCP stages were conserved also in malignant cells. Therefore we compared malignant and physiological B cells from paediatric ALL/
NHL
and normal BM samples with respect to
CD99
expression using selective gating and semi-quantitative flow cytometry. Common-ALLs (n = 45) were similar to their corresponding, very immature BCPs (stage 1) in expressing very high levels of
CD99
. Most pre-B ALLs (n = 16) were also CD99hi and thus differed from the patterns found in normal cytoplasmic mu-chain+ (cmu+) pre-B cells (stage 2, CD99lo). In particular, we found that those pre-B-ALL cases which were CD34+ also showed higher
CD99
expression than the CD34- cases. This prompted us to investigate the levels of
CD99
in those rare normal BCPs which also coexpress CD34 and cmu; these cells, which are transitory from stage 1 to stage 2, were found also CD99hi, thus precisely reflecting the patterns of CD34+ pre-B ALLs. The blasts of Burkitt-type B-cell ALL/
NHL
samples (n = 13) expressed considerably less
CD99
, similarly to the more differentiated BCP stages 2 (cmu+) and 3 (surface mu-chain+). In summary, we found that paediatric B-lineage malignancies display remarkable synchrony regarding the levels of
CD99
expression compared to their putative normal counterparts.

To verify the spectrum of
CD99
-expressing lymphoid malignancy, an immunohistochemical study for
CD99
was carried out in 182 cases of
non-Hodgkin's lymphoma
, including 21 lymphoblastic lymphomas, 11 small lymphocytic lymphomas, 9 mantle cell lymphomas, 12 follicular lymphomas, 37 diffuse large B cell lymphomas, 18 Burkitt's lymphomas, 28 NK/T-cell lymphomas, 8 angioimmunoblastic T-cell lymphomas, 23 peripheral T-cell lymphomas, unspecified, and 15 systemic anaplastic large cell lymphomas.
CD99
was positive in all T-lymphoblastic lymphomas and in 60% of B-lymphoblastic lymphomas. Majority of T and NK cell lymphomas were negative for
CD99
, except anaplastic large cell lymphomas (ALCLs). Eight of 15 cases (54%) of ALCLs reacted with anti
CD99
antibody. Seven of 10 (70%) ALK positive ALCLs expressed
CD99
, whereas only 1 of 5 (20%) ALK negative ALCLs were positive. Of the mature B-cell lymphomas, 5.4% (2/37) of diffuse large B cell lymphomas and 11.1% (2/18) of Burkitt's lymphomas expressed
CD99
. In conclusion,
CD99
is infrequently expressed in mature B and T cell lymphomas, except ALK-positive ALCL. High expression of
CD99
in ALK-positive ALCL is unexpected finding and its biologic and clinical significances have yet to be clarified.

non-Hodgkin's lymphoma

) in Shanghai. A retrospective analysis was conducted in 516 patients with
NHL
. Patient clinical data, including age, sex, diagnosis, immunophenotypes, and karyotypes, were collected. The median age was 58 years. There was a male predominance in all
NHL
, except extranodal marginal zone lymphoma of mucosa-associated lymphoid tissue. Patients with B cell
NHL
(1.5%) expressed CD3. T cell
NHL
patients (11.5%) expressed CD20. Epstein-Barr virus latent integral membrane protein 1, BCL6, CD10, Bcl-2, CD68, myeloperoxidase,

, CD30, CD15, and CD43 were present in various types of
NHL
. Complex karyotypes accounted for 92.3% of the 73.7% patients with abnormal karyotypes. Immunoglobin heavy chain gene translocation was present in 60.3% of B cell and 23.7% of T/NK cell neoplasms. Understanding the complex clinicopathological and molecular features of
NHL
may help with prognosis and serve as targets for treatments.

, neuroblastoma, hepatoblastoma, Wilm's tumor, desmoplastic small round cell tumor, and other morphologically similar entities. Differential diagnosis of Ewing sarcoma/primitive neuroectodermal tumor (Ewing/PNET sarcomas or EPS) is difficult. In addition to morphology and immunohistochemistry (IHC), differential diagnosis of these tumors is based on molecular analysis of the
EWSR1
gene rearrangement using fluorescent in situ hybridization (FISH) technique. We investigated the diagnostic value of combined
CD99
immunostaining and
EWSR1
t(22q12) alteration using a dual-color, break-apart rearrangement probe in forty-one formalin-fixed paraffin-embedded (FFPE) tissue samples from pediatric and adult patients diagnosed with EPS. IHC was performed in all cases using the
CD99
antibody and showed a positivity of 92.7% in the enrolled cases (38/41) followed by FISH analysis where 48.8% of the cases (20/41) were rearranged. Sensitivity and specificity for IHC assays were 88% and 58%, respectively. Notably, FISH had a sensitivity of 100% and a specificity of 87%. In addition,
CD99
positivity was found to correlate with
EWSR1
rearrangement (
p
< 0.05). This report shows that FISH has better sensitivity and specificity than IHC in the Moroccan population, and supports its combination with
CD99
immunostaining as diagnostic biomarkers for this rare malignant entity."
