To find cellular binding counterparts for the nucleocapsid protein (N) of Tula hantavirus (TULV), two cDNA libraries were screened using yeast two-hybrid systems based on LexA and Gal4 transcription factors. Five cDNA clones encoding SUMO-1 (Small
Ubiquitin
-related MOdifier, also known as
sentrin
) were selected in the LexA system. Confocal microscopy revealed that, in infected cells, TULV N protein and SUMO-1 colocalize at the perinuclear area providing further evidence for interaction between the two proteins. Neither endogenous nor transiently expressed SUMO-1 was found to be covalently linked to the N protein. Additional evidence that the interaction is non-covalent was obtained in immunoprecipitation experiments: N protein-specific antibodies precipitated SUMO-1 from TULV-infected Vero E6 cell lysate. By using a pepscan assay, two basic amino acid stretches in the N-terminal part of SUMO-1 were shown to be involved in the interaction.

-specific proteases (Ulp/SENPs) mediate both processing and deconjugation of small ubiquitin-like modifier proteins (SUMOs). Here, we show that Ulp/SENP family members SENP3 and SENP5 localize within the granular component of the nucleolus, a subnucleolar compartment that contains B23/nucleophosmin. B23/nucleophosmin is an abundant shuttling phosphoprotein, which plays important roles in ribosome biogenesis and which has been strongly implicated in hematopoietic malignancies. Moreover, we found that B23/nucleophosmin binds SENP3 and SENP5 in Xenopus laevis egg extracts and that it is essential for stable accumulation of SENP3 and SENP5 in mammalian tissue culture cells. After either codepletion of SENP3 and SENP5 or depletion of B23/nucleophosmin, we observed accumulation of SUMO proteins within nucleoli. Finally, depletion of these Ulp/SENPs causes defects in ribosome biogenesis reminiscent of phenotypes observed in the absence of B23/nucleophosmin. Together, these results suggest that regulation of SUMO deconjugation may be a major facet of B23/nucleophosmin function in vivo.

and ubiquitin-like proteins are conjugated to a wide variety of target proteins that play roles in all biological processes. Target proteins are conjugated to ubiquitin monomers or to ubiquitin polymers that form via all seven internal lysine residues of ubiquitin. SUMO (small ubiquitin-related modifier) shares the ability of ubiquitin to form chains via internal SUMOylation sites. SUMO chain-binding proteins including ZIP1, SLX5/8 (synthetic lethal of unknown function 5/8), RNF4 (RING finger protein 4) and CENP-E (centromere-associated protein E) have been identified that interact non-covalently with SUMO chains, thereby regulating target proteins that are conjugated to SUMO multimers. SUMO chains play roles in replication, in the turnover of SUMO targets by the proteasome and during mitosis and meiosis. Thus signalling via polymers is an exciting feature of the SUMO family.
