An effect of somatostatin on arterial blood pressure, isolated heart, and its interaction with adrenergic system have been studied in rats. It was found, that somatostatin does not exert any significant effect on blood circulation. It modifies however catecholamines action. Somatostatin decreases the activity of alpha-adrenergic receptors agonist (norepinephrine), and potentiates the activity of beta-adrenergic receptors agonist (isoprenaline).

Effect of thyroliberin, somatostatin, vasopressin and luliberin on RNA synthesis in isolated calf pituitary, brain cortex, rat liver and rat brain nuclei was assayed. None of the examined neuropeptides affected RNA synthesis.

Plasma somatostatin-like activity (SLI) was measured in obese women with type 2 diabetes in fasting condition and after a test meal (714 kcal: 27% carbohydrates, 8% protein, 73% fat). The tests were done twice: first, after a week of dietetic treatment and then 7 days of metformin administration, 1.5 g a day. Blood glucose and insulin concentration (IRI) were also followed throughout the test. The plasma SLI on fasting and after the test meal, before and after treatment with metformin did not show any significant differences. Blood glucose was lower in the fasting state significantly and serum insulin showed a tendency to decrease at the end of the test. These findings contradict the hypothesis of a possible role of somatostatin in peripheral blood in the enteric effect of metformin. The local (paracrine?) action of SLI secreted after metformin administration cannot be excluded.

The effect of somatostatin-14 on gastric emptying, as well as on serum concentrations of gastrin, insulin, glucose, calcium, and phosphorus after ingestion of a mixed solid-liquid meal was examined in seven healthy men in a double-blind placebo-controlled study. An i.v. bolus injection of 61 nmol somatostatin was followed by 244 nmol infused at a constant rate over 90 minutes. Gastric emptying of a radiolabelled meal was surveyed with the use of a gamma camera. A weak delaying effect of somatostatin on gastric emptying of a solid meal did not prove to be statistically significant. Somatostatin decreased significantly the postprandial gastrin release: the area under the gastrin curve, AUC0-90, 9954 +/- 2287 ng.l-1 min (placebo) vs 5327 +/- 718 ng.l-1 min (somatostatin), p less than 0.05. At the same time suppression of the postprandial insulin release by somatostatin was observed--area under the insulin curve, AUC0-90, 1450 +/- 161 mU.l-1 min (placebo) vs 501 +/- 60 mU.l-1 min (somatostatin), p less than 0.002. The postprandial increase in serum glucose concentration was initially attenuated, and shifted towards the end of somatostatin infusion, when referred to the placebo situation. Somatostatin did not change significantly the serum calcium or phosphorus concentrations. The results obtained indicate that somatostatin's effect on gastric evacuation is less pronounced in contrast to the significant inhibitory influence of somatostatin on release of gastrointestinal hormones.

Low concentrations of somatostatin inhibited, while high ones of dopamine concentrations stimulated the activity of kinase.

Short-lasting hypothermia during thiobutabarbital general anaesthesia causes no decrease of the absolute ATP level in the blood and liver of rats. The adenylate energy charge in the tissues is relatively high - 0.86 in the liver and 0.85 in the muscles, which might be an evidence of a significant "energy sparing" during moderate hypothermia (26 +/- 1 degree C). Somatostatin in a dose of 20 micrograms/kg of body weight given to the rats during hypothermia decreased the ATP level, the ATP/ADP ratio and the adenylate energy charge in the studied tissues, especially in the liver, evidencing increased intensity of catabolic processes caused by the inhibitory action of somatostatin on the release of insulin and glucagon, among other hormones, and on the change of the insulin/glucagon ratio.

This paper presents a patient with diarrhoeic syndrome, who was finally diagnosed to have carcinoid tumor of the heart. Symptomatic remission has been achieved with Somatostatin therapy. It seems to be treatment of choice in non-operable carcinoid tumours.

The paper describes a cytological pattern of carcinoid of the appendix metastasizing to the liver and lymph nodes, clinically manifested by increased insulin level, in whose cells immunohistochemical studies revealed also the presence of gastrin and somatostatin. The case was confirmed by autopsy examination.

Somatostatin (S), a naturally occurring peptide, was first discovered more than 20 years ago. This peptide was later found in many tissues, including human cardiac tissue. High concentration of S was found in the sinus and atrioventricular (AV) nodes. Negative chronotropic activity of S and depressive effects of this substance on AF junction have also been described. The purpose of the present study was to determine the effect of S on the cardiac conduction system in man during antegrade and retrograde conduction. The study group consisted of 12 pts (5 women and 7 men, mean age 43 years). They were subjected to invasive electrophysiologic testing because of sudden unexplained heart palpitations. After measuring all electrophysiologic variables in the control state, each patient was given S (preparation Stilamin, Serono) intravenously with a syringe pump at a dose of 8.5 micrograms/min, for about 30 min. All the electrophysiologic measurements were repeated in the same order, beginning from the 6th min after the start of S injection. S prolonged mean sinus cycle length by 13%, sinoatrial, intraatrial and AV nodal conduction times by 15, 9 and 10%, respectively. Effective refractory periods of the atrium, AV node and right ventricle were prolonged by 7, 8 and 8%, respectively (all p values less than 0.01). Antegrade AV nodal Wenckebach cycle length, as well as retrograde, increased significantly (by 9 and 10% respectively). There were no significant changes of the mean HV interval, corrected QT interval, ventriculoatrial conduction time and corrected sinus node recovery time during S infusion.(ABSTRACT TRUNCATED AT 250 WORDS)

The authors report a case of acute pancreatitis, complicated by pancreatic-peritoneal fistula and ascites in 26 year old man, with a history of excessive alcoholic intake. Biochemical investigation of ascitic and pleural fluids, which revealed markedly elevated activity of pancreatic enzymes and protein indicated their pancreatic origin. After 25 days of treatment with somatostatin and parenteral nutrition, the condition of the patient improved and he was dismissed home.
