Human blood polymorphonuclear neutrophils (PMN) are thought to be involved in the pathogenesis of asthma through their recruitment into the bronchoalveolar lumen and the lung by local release of chemotactic factors. Therefore chemotactic activities of several mediators (
PAF
, histamine and three neuropeptides substance P, VIP and a
somatostatin
analog) were compared on blood PMN from both healthy subjects (HS) and asthmatic patients (AP). The maximal response to
PAF
was significantly different (P less than 0.05) with cells from both groups. Moreover activity for the HS peaked at 10(-6) M, whereas the AP showed peak chemotactic activity at 10(-8) M. Histamine had no chemoattractant effect on PMN. Substance P did not induce PMN locomotion, whereas VIP induced a chemotactic response in a dose-dependent manner, particularly with cells from HS as compared to those from AP. BIM 23014 (a
somatostatin
analog) exhibited chemotactic activity which was also more pronounced with PMN from HS as compared to those from AP. Our findings showed that blood PMN could be involved in asthma through their heightened locomotor reactions to mediators which are known to be released locally by activated cells in bronchoalveolar lumen.

The factors responsible for in vivo eosinophil recruitment are poorly defined, although T-lymphocytes appear to be involved in the etiology of eosinophilia. In order to clarify this relationship, we studied the modulation of eosinophil mobilization in the rat after immune challenge, by chronic treatment with the
PAF
-antagonist, BN 52021, the
somatostatin
analog, BIM 23014 and with Cyclosporin A (CsA). In rats made hypereosinophilic by pretreatment with cyclophosphamide or sephadex, a significant increase of the eosinophil count in blood and peritoneal fluid was induced by anaphylactic reaction. CsA totally abolished both hypereosinophilia and peritoneal eosinophil infiltration. BIM 23014 also, significantly reduced the circulating eosinophils (-68%, p less than 0.001) and cell infiltration (-86%, p less than 0.05). In contrast, BN 52021 decreased peritoneal eosinophil recruitment, while having relatively little effect on circulating cells. CsA and
somatostatin
are known to affect T-cell proliferation, and as T-cells are involved in the differentiation of hematopoietic cells into eosinophils, these drugs could decrease eosinophil availability for recruitment. In contrast, the
PAF
antagonist may act by inhibiting
PAF
-induced eosinophil chemotaxis, providing a more specific inhibition of this process than that exerted by CsA, BIM 23014 and other immunosuppressive agents.

Platelet-activating factor and
somatostatin
receptors, two G protein-coupled receptors expressed in the rat hippocampus, were analyzed for the downstream signaling pathways in Chinese hamster ovary cells stably expressing each receptor. Ligand stimulation to each CHO cell line induced (1) inhibition of forskolin-induced accumulation of cAMP, (2) arachidonate release, and (3) activation of mitogen-activated protein kinase and MAP kinase kinase. In contrast, inositol phosphate breakdown was seen only in the
PAF
-stimulated CHO cells. The induction of these signals accompanied no detectable Ras activation. Suppression of the signals by pertussis toxin was almost complete for the somatostatin receptor but partial for the PAF receptor, suggesting that the somatostatin receptor couples only with PTX-sensitive G protein, while the PAF receptor couples with both PTX-sensitive and -insensitive G proteins. A model of G protein-mediated signaling pathways was proposed in which the signals from Gi and those from Gq converge at MAP kinase kinase and lead to arachidonate release. The present system using CHO cells is useful for analyzing signaling pathways from G proteins to MAP kinase kinase and will thereby provide clues for understanding the mechanisms underlying the physiological and pathological events mediated by
PAF
,
somatostatin
, and other G protein-coupled receptors in the central nervous system and other tissues.

Pancreatic tissue specimens of domestic ducks were fixed in

, paraformaldehyde and Bouin's fluid. Sections were immunohistochemically stained with antisera against insulin, glucagon,
somatostatin
and 5-hydroxytryptamine (5-HT). Immunoreactive cells were detected in both exocrine and endocrine pancreas. The endocrine cells were grouped into three islet types: A, B and mixed islets. A islets comprised glucagon,
somatostatin
and 5-HT immunoreactive cells. B islets showed insulin,
somatostatin
and 5-HT immuno-reactive cells. Mixed islets were composed of all four immunoreactive cytotypes. Using a double immunofluorescent technique, it was observed that 5-HT immunoreactivity was present in some of the glucagon and in some of the insulin immunoreactive cells. B islets were observed throughout the pancreas. A and mixed islets were more concentrated in the splenic lobe, and decreased in number in the other lobes. These results were compared with those obtained in other avian species.
