1. The effect of 10 days treatment with growth hormone (GH) (1 mg/kg body wt/day) and somatostatin (SRIF) (0.25 mg/kg body wt/day) subcutaneously on the activity of beta-adrenoceptors in rat hypothalamic, pituitary and cerebral cortical membrane fractions was studied using [3H]dihydroalprenolol ([3H]DHA) as radioligand. 2. The administration of GH significantly increased the beta-adrenoceptor binding affinity and the administration of SRIF decreased the beta-adrenoceptor binding capacity in the hypothalamus. 3. In the pituitary the beta-adrenoceptor binding affinity was significantly decreased after both hormonal applications. 4. In the cerebral cortex the beta-adrenoceptor binding affinity was significantly decreased after the GH treatment and increased after the SRIF treatment. 5. The present study provides direct evidence for GH and SRIF effects on the activity of rat beta-adrenoceptors and supports the view about the involvement of beta-adrenergic mechanisms in the neurotransmitter regulation of GH secretion in the rat.

Recently we reported that insulin resistance and glucose induced insulin release are inversely correlated to age in young healthy siblings of diabetic patients. To confirm this pattern of change with age, the subjects were reexamined after two years. The study was limited to the age groups with the lowest insulin sensitivity, i.e. 14.0-15.9 years for females and 16.0-17.9 years for males. All five girls and four of five boys showed an increased insulin sensitivity as measured by the somatostatin-insulin-glucose infusion test (p = 0.02). All subjects showed a decrease in fasting levels of insulin-like growth factor 1 (IGF-1) (p less than 0.01) during the observation period. All except one showed an increase in the levels of dehydroepiandrosterone sulphate (DHEA-S) (p less than 0.01). These data confirm that insulin sensitivity increases in the late teenage period. The parallelism to the changes of IGF-1 indicate that the pubertal changes in insulin sensitivity may partly be caused by growth hormone. Our data contradict the hypothesis that the low insulin sensitivity of puberty is due to the increased levels of DHEA-S.

Using a combined infusion of somatostatin, insulin and glucose, insulin resistance was assessed in vivo in two groups of females with polycystic ovaries (PCO), obese (PB-PCO) and normal weight (NO-PCO) and in two groups of matched (for age, sex and body mass index) controls (OB and NO). A steady state plasma glucose (SSPG) and insulin (SSPI) was attained after 90 min. OB-PCO and NO-PCO showed higher SSPG with respect to matched controls. The SSPG levels were related to body mass index (r = 0.69; P less than 0.001). The SSPG values were significantly correlated with the fasting insulin levels (r = 0.47; P less than 0.003). Gonadotrophin and steroid peripheral blood concentrations were also evaluated in the PCO females. A significant correlation was found between the SSPG values and the dehydroepiandrosterone sulphate levels (r = 0.46; P less than 0.05) and between the fasting insulin levels and the androstenedione concentrations (r = 0.64; P less than 0.01). Moreover, significant correlation coefficients were found between the glucose to insulin ratio and the A (r = -0.59; P less than 0.01) and the DHEA-S (r = -0.50; P less than 0.05) plasma levels. Finally, no relationship between body mass index and A or DHEA-S levels was found in PCO females considered as a group. We conclude that insulin resistance is present in females with PCO and it is mainly due to the presence of obesity, but other factors such as androgen levels, probably of adrenal sources, must be considered as a cause.

In order to observe the change of brain fatty acid composition and neurotransmitter by DHA supplementation during the development stage of young rats, pregnant rats were randomly divided into four groups: normal control group (O), low (L), medium (M) and high (H) dose-DHA supplemented groups. The fatty acids composition, the levels of 5-hydroxy-tryptamine (5-HT), dopamine (DA), norepinephrine (NE) and somatostatin (SS) in ceretral cortex and hippocampus of offsprings were measured. The body weight of pregnant rats and offsprings were also observed. The results showed that DHA content of brain homogenate and the levels of 5-HT, DA and SS in hippocampus of experimental young rats were significantly higher than those of the control group. And the body weight of young rats in the three DHA-supplemented groups were retarded in different degree. It was concluded that DHA supplementation changed the fatty acid composition and increased the levels of 5-HT, DA and SS in hippocampus. Proper ratio of n-3 and n-6 fatty acids in dietary supplementation should be considered.
