Human immunodeficiency virus (HIV) was detected by assay of reverse transcriptase activity in a "virus pellet" obtained by differential sucrose density centrifugation of cell-free semen from three patients with the acquired immune deficiency syndrome (AIDS), one individual with
AIDS-related complex
(
ARC
), and in an asymptomatic homosexual male. Reverse transcriptase assays indicated virus concentrations in the range of 10(8) particles/ml of semen, an accumulation substantiated by electron microscopic visualization of cell-free virus. This is the first description of cell-free retrovirus in seminal fluid and at a greater concentration than reported for blood or other body fluids or tissues. These results suggest that the male reproductive tract of humans may be a reservoir of HIV expression, and raises the possibility that the cells lining the
epididymal
lumen could be chronically infected with HIV. These are important considerations in formulating treatment and preventive strategies.

1. Four groups of rat pups were reared: mother-reared (MR) control (well-fed) and undernourished (MRC and MRU respectively) and artificially reared (AR) control and undernourished (
ARC
and ARU respectively). Pups for artificial rearing were fitted with a gastric cannula on postnatal day 5 and were fed, by intermittent gastric infusion, expressed rats' milk (days 5-7), mixtures of rats' milk and milk-substitute (days 8-16), and milk-substitute only (days 17-20). Solid food was available to MR pups throughout and to AR pups from day 14. Undernutrition, imposed from postnatal days 5 to 25, was effected initially by underfeeding the mother (MRU) or by infusing restricted quantities of milk (ARU). Weaning was at 21 d and undernutrition from day 21 to day 25 was by restricting the supply of solid food. All rats were fed ad lib. from 25 d. 2. The developmental milestone, eye-opening, was delayed by undernutrition but unaffected by artificial rearing. 3. Growth curves in body-weight during the refeeding phase were influenced most by previous undernutrition and to a lesser extent (also negatively) by artificial rearing. 4. Fourteen measures of body and organ growth were taken at autopsy at 39 weeks. Twelve measures were affected by nutrition and only four by rearing (weight of whole body,

1 The subtypes of alpha1-adrenoceptor mediating contractions of rat vas deferens to endogenous and exogenous noradrenaline and to the exogenous agonists methoxamine, phenylephrine and A61603 have been examined. 2 The effects of antagonists on the shape of concentration-response curves, both tonic and phasic, to the four agonists were analysed. Prazosin produced parallel shifts in all cases. Particularly for RS 17053 against noradrenaline, there was some evidence for a resistant component of the agonist response. High concentrations of RS 17053 (1-10 microM) virtually abolished tonic contractions but phasic contractions were resistant. 3 A series of nine antagonists (the above and WB4101, benoxathian, phentolamine, BMY 7378, HV 723, spiperone) were investigated against contractions to noradrenaline. The correlation with the potency of the series of alpha1-adrenoceptor antagonists against contractions to noradrenaline was significant only for the alpha1A-adrenoceptor ligand binding site (r=0.88, n=9, P<0.01). 4 In

portions (nifedipine 10 microM), the isometric contraction to a single electrical pulse is alpha1-adrenoceptor mediated. The correlation with ligand binding sites for 11 antagonists (the above plus

239 and (+)-niguldipine) was significant only for the alpha1D-adrenoceptor subtype (r=0.65, n=11, P<0.05). 5 In conclusion, tonic contractions of rat vas deferens produced by exogenous agonists are mediated predominantly by alpha1A-adrenoceptors, although a second subtype of receptor may additionally be involved in phasic contractions. Nerve-stimulation evoked alpha1-adrenoceptor mediated contractions seem to predominantly involve non-alpha1A-adrenoceptors, and the receptor involved resembles the alpha1D-receptor.
