The carcinoembryonic antigen (CEA) and ABO (H) blood group antigens of 47 bladder tumors were investigated by immunohistochemistry (CEA by
peroxidase
-antiperoxidase method and blood group antigens by Avidin-biotin-
peroxidase
complex method) and the results were compared with the histopathological diagnosis. Twenty five percent of the bladder tumors had CEA-positive tumor cells. They were found in 11.1% of pT1 cases, in 12.5% of pT2 cases, 70.0% of
pT3
cases and 50.0% of pT4 cases and were also found in 13.3% of grade I cases, in 50.0% of grade II cases and in 40.0% of grade III cases. On the other hand, positive blood group antigen cells were found in 85.2% of pT1 cases, 50.0% of pT2 cases, 20.0% of
pT3
cases and 50.0% of the pT4 cases and also in 80.0% of the grade I cases, 41.1% of the grade II cases, and 20.0% of the grade III cases. The monoclonal antibody Ki-67 recognizes a human nuclear antigen that is present in proliferating cells (G1, S, G2 and M cycling stages of cell division) exclusively, but is absent in G0 cycling stage. We performed immunohistochemical analysis of renal cell carcinomas using Ki-67 antibody, and investigated the relationship between the proportion of Ki-67 positive cells and pathological findings of renal cell carcinomas. The tissues were obtained from 36 patients with renal cell carcinoma who underwent radical nephrectomy. Cryostatfrozen sections were cut at 5 microns and stained with avidin-biotin-
peroxidase
complex method. The percentage of Ki-67 positive cancer cells to the total amount of cancer cells was expressed as growth fraction (GF). We obtained the following results with regard to the relationship between GF and pathological findings of renal cell carcinomas. 1) GFs in pathological grades were 6.58 +/- 4.57% (mean +/- SD) for grade 2 (n = 13) and 1.20 +/- 0.70% for grade 1 (n = 21). GF in grade 2 was significantly higher than that in grade 1 (p < 0.01). GF for Grade 3 (n = 2) were 9.2% and 27.8% respectively. 2) GFs in the pathological stages were 8.12 +/- 7.64% for
pT3
(n = 13) and 1.84 +/- 1.30% for pT2 (n = 23). GF in
pT3

BACKGROUND: The immunohistochemical expression of thymidylate synthase (TS) and thymidine phosphorylase (TP) was examined in a comparative study of the recurrence rates and prognoses of patients with advanced gastric cancer at the same stage.METHODS: We examined the resected specimens of 67 patients with stage IIIB gastric cancer (
pT3
, pN2, M0) under 70 years of age who had undergone curative gastrectomy followed by adjuvant chemotherapy with 5-fluoropyrimidines. Paraffin sections of the resected specimens were stained with human anti-TS polyclonal and anti-TP monoclonal antibodies by the avidin-biotin-
peroxidase
complex (ABC) method.RESULTS: The overall expression of TS and TP was 45.4% and 43.4%, respectively. The postoperative survival curve for the TS-positive group was significantly depressed compared with that for the TS-negative group ( P = 0.0480). The survival curves for TP-positive and TP-negative groups did not show any difference. In regard to the combination of TS and TP expression, the best survival curve was obtained for the TS(-)/TP(+) group, followed by those for the TS(-)/TP(-), TS(+)/TP(-), and TS(+)/TP(+) groups in descending order. With regard to the recurrence site, there was no significant difference in peritoneal recurrence in relation to positivity for TS or TP. Lymph node recurrence, however, was significantly higher in the TS-positive and TP-positive groups, with P-values being 0.0466 and 0.0058, respectively, versus the corresponding negative groups. The incidence of hepatic recurrence was higher in the TP-positive group than in the TP-negative group ( P = 0.0910). As for the total doses of 5-fluoropyrimidines given, more favorable survival curves were obtained for the high dose of negative TS and TP groups, but no significant differences were observed in their positivities.CONCLUSION: The expressions of TS and TP showed different characteristics in overall survival and recurrence rate or site. They should be used for predicting prognosis in comprehension on their properties.

The purpose of the study was to evaluate the expression of IGF-IR in primary tumours and lymph node metastases of oral cancers and the correlation between expression of IGF-IR and some clinicopathological features. Fifty-seven (57) oral cancers were examined by immunohistochemical studies, using the avidin-biotin-
peroxidase
method. Our study included only oral cancers, classified histopathologically as squamous cell carcinoma (7 cases in G1 grade, 44 (G2) and 6 (G3); 23/pT1 stage, 18/pT2, 7/
pT3
and 9/pT4). Positive immunostaining for IGF-IR was noted in 32, out of 57 (56.1%) of oral tumours. We found a tendency (p=0.081) toward an association between IGF-IR expression in the primary tumours and their stage (
pT3
and pT4). A comparison between the primary tumours and matching lymph node metastases revealed that 13, out of 20, (65%) cases showed a convergence between primary tumours and matching lymph node metastases with regard to either negative or positive staining.
