Plasmids containing the varicella zoster virus (VZV) open reading frames (ORFs) 61 and 62 were used in a transient co-transfection assay to test for trans-activation of the VZV and herpes simplex virus type 1 (HSV-1) thymidine kinase (tk) promoters. The trans-activating potential of the polypeptides encoded by these VZV ORFs, designated
p51
and p140, was compared to that of their HSV-1 homologs ICP0 and ICP4, respectively. VZV
p51
was functionally inactive in this system while p140 appeared to be a much stronger
transcriptional activator
than ICP4. Co-transfection of plasmids encoding VZV p140 and HSV-1 ICP0 resulted in a synergistic activation of the reporter gene as has been shown for the combination of ICP4 and ICP0.

The role that the p53 tumor suppressor gene product plays in cellular differentiation remains controversial. However, recent evidence indicates that p53 is required for proper embryogenesis. We have studied the effect of p53 on the expression mediated by the promoter of the rat muscle-specific phosphoglycerate mutase gene (M-PGAM), a marker for cardiac and skeletal muscle differentiation. Experiments involving transient transfection, mobility shift assay, and site-directed mutagenesis demonstrated that p53 specifically binds and transactivates the M-PGAM promoter. The p53-related proteins p51A and
p73L
also transactivated M-PGAM. Moreover, stable expression of a p53 dominant mutant in C2C12 cells blocked the induction of M-PGAM expression during the myoblast to myotube transition and the ability of p53, p51A, and
p73L
to transactivate the M-PGAM promoter. In addition, impaired expression of M-PGAM was observed in a subset of p53-null animals in heart and muscle tissues of anterior-ventral location. These results demonstrate that p53 is a
transcriptional activator
of M-PGAM that contributes in vivo to the control of its cardiac expression. These data support previous findings indicating a role for p53 in cellular differentiation.

/p63 is a novel p53 homologue that has been shown to act as a
transcriptional activator
through the p53-binding sequence of the p21/WAF1 promoter and to induce apoptosis when it is expressed transiently in a human tumor cell line. We developed transcription assay systems for these two related genes in both Saccharomyces cerevisiae and mammalian cells and used them to investigate the functional similarities and differences of these genes. We found that

/p63 trans-activated the previously identified p53 target genes, but the degree of the transactivation by
p51
/p63 differed from that by p53. These results suggest that the cellular signal on

transcriptional activator
by binding to p53-responsive elements in the promoter regions of several p53 downstream genes. Recently, we identified four distinct mutations in the

/p63 gene after screening >200 human tumors and cell lines. Because all of the detected
p51
/p63 mutations were missense mutations, the pathogenic effect of these mutations is difficult to determine without performing a functional analysis. In this study, we examined the transcriptional activity of tumor-derived
p51
/p63 missense mutations using a yeast-based assay and compared the data with that of artificial
p51
/p63 missense mutations at residues corresponding to the positions and substituted residues of p53 mutation "hotspots." Although most of the
p51
/p63 missense mutations at the p53 hotspot residues were unable to transactivate the promoters used in this study, the tumor-derived
p51
/p63 missense mutations retained their ability to transactivate the MDM2 and/or the BAX promoter but not the p21/WAF1 promoter. These results suggest that the
p51
/p63 mutation might be involved in an unknown tumor suppression pathway distinct from that of p53.
