Evidence is presented that, as in cardiomyocytes, vascular endothelial cells use fatty acids, in addition to glucose, as a respiratory fuel. Attention is focused on the cardiac interstitium, lined by vascular cells and cardiomyocytes, which may be enriched with metabolic products from these cells. Also, certain proteins are present in the interstitial fluid (Qi) such as plasma proteins and fatty acid binding protein (FABP). However, the concentration of FABP is so low in Qi that albumin is more important to shuttle long chain fatty acids in the interstitial fluid between cardiomyocytes and the vascular compartment. Under hypoxic conditions (hypo)xanthine, lactate and fatty acids may be expected to accumulate in the interstitium, as well as proteins from adjacent cells, such as xanthine oxidase from endothelial cells. This enzyme, acting upon the elevated level of (hypo)xanthine, giving rise to O2-., may be involved in the damage of the ischaemic heart. The significance of the interstitium in ischaemia and in fibrosis following long standing cardiac lipidosis is briefly discussed, as well as the possible mechanisms involved in fatty acid transport in the heart.

The characteristics and possible functions of the most abundant proteins associated with the bovine milk-fat globule membrane are reviewed. Under the auspices of the Milk Protein Nomenclature Committee of the ADSA, a revised nomenclature for the major membrane proteins is proposed and discussed in relation to earlier schemes. We recommend that proteins be assigned specific names as they are identified by molecular cloning and sequencing techniques. The practice of identifying proteins according to their Mr, electrophoretic mobility, or staining characteristics should be discontinued, except for uncharacterized proteins. The properties and amino acid sequences of the following proteins are discussed in detail: MUC1, xanthine dehydrogenase/oxidase, CD36, butyrophilin, adipophilin, periodic acid Schiff 6/7 (PAS 6/7), and fatty acid binding protein. In addition, a compilation of less abundant proteins associated with the bovine milk-fat globule membrane is presented.

Although some of the physiological roles of milk fat globule membrane (MFGM) proteins are still unclear, there is increasing evidence that the consumption of bovine MFGM proteins has significant nutritional health benefits for humans; therefore, it may be important to be able to estimate the MFGM proteins in complex ingredients. In this study, the absolute quantification (AQUA) technique, which is typically used for the quantification of proteins in proteomic studies, was applied for the quantification of bovine MFGM proteins in butter milk protein concentrate. Six MFGM proteins (fatty acid binding protein, butyrophilin, PAS 6/7, adipophilin, xanthine oxidase, and mucin 1) were simultaneously quantified using high-resolution selected reaction monitoring mass spectrometry. Samples were rehydrated in 6.7 M urea buffer prior to dilution to 2.2 M before tryspin digestion. Direct rehydration in 2.2 M urea buffer or 2.2 M urea/20% acetonitilrile buffer reduced peptide yield digestion. Isotopically labeled peptides were used as internal standards. The coefficient of variation ranged from 5 to 15%, with a recovery of 84-105%. The limit of detection was in the range of 20-40 pg.
