activity in human plasma was measured by quantifying the

-derived uric acid (UA) in plasma using the high-performance liquid chromatography (HPLC) equipped with a UV detector. Chromatographic separation consisted of the mobile phase (a mixture of 0.1%
trifluoroacetic acid
in Milli-Q water and 0.085%
trifluoroacetic acid
in acetonitrile in a mix ratio of 99:1) running through a Zorbax StableBond SB-C(18) column at a flow-rate of 1 ml/min. Deproteinization with heat-treatment of plasma samples after the reaction was used in the assay to avoid splitting of the UA and xanthine peaks caused by acid deproteinization that could interfere the accurate determination of human plasma

activity in our case. Based on the examination of the dependence of
XOR
activity on added amounts of xanthine and reaction times, the amount of xanthine and reaction time for
XOR
activity assay were determined to prevent the errors caused by the limiting effect of substrates and plateau phase of the reaction. Using this method, human plasma
XOR
activities of 25 healthy people were measured. The average human plasma
XOR
activity was 2.1+/-0.8 (x10(-3) U/ml).

1,4,8,11-tetraazacyclotetradecane (cyclam), which is one of the most extensively investigated ligands in coordination chemistry, in its protonated forms, can play the role of host toward cyanide metal complexes. We have investigated the acid-driven adducts formed in acetonitrile-dichloromethane (1:1 v/v) solution by [Ru(bpy)(CN)4](2-) with 1,4,8,11-tetrakis(naphthylmethyl)cyclam (1) and a dendrimer consisting of a cyclam core appended with 12 dimethoxybenzene and 16 naphthyl units (2). [Ru(bpy)(CN)4](2-), 1, and 2 exhibit characteristic absorption and emission bands, in distinct spectral regions, that are strongly affected by addition of acid. When a solution containing equimolar amounts of [Ru(bpy)(CN)4](2-) and 1 or 2 is titrated by
trifluoroacetic acid
, or when [Ru(bpy)(CN)4](2-) is titrated with (1.2H)2+ or (2.2H)2+, [[Ru(bpy)(CN)4](2-).(2H+).1] or [[Ru(bpy)(CN)4](2-).(2H+).2] adducts are formed in which the fluorescence of the naphthyl units is strongly quenched by very efficient energy transfer to the metal complex, as shown by the sensitized luminescence of the latter. The [[Ru(bpy)(CN)4]2-.(2H+).1] and [[Ru(bpy)(CN)4](2-).(2H+).2] adducts can be disrupted (i) by addition of a base (1,4-diazabicyclo[2.2.2]octane), yielding the starting species [Ru(bpy)(CN)4](2-) and 1 or 2, or (ii) by further addition of triflic acid, with formation of (1.2H)2+ or (2.2H)2+ and protonated forms of [Ru(bpy)(CN)4](2-). It is shown that upon stimulation with two chemical inputs (acid and base) both [[Ru(bpy)(CN)4](2-).(2H+).1] and [[Ru(bpy)(CN)4](2-).(2H+).2] exhibit two distinct optical outputs (a naphthalene-based and a Ru(bpy)-based emission) that behave according to an
XOR
and an XNOR logic, respectively.

Vaccinium uliginosum L. (also known as bog bilberry) is a low-growing deciduous shrub classified in the Ericaceae family of plants, which includes numerous Vaccinium berries, blueberries, and cranberries. Berries of the Ericaceae family are known to contain organic acids, vitamins, glycosides, and anthocyanins and have been reported to have antioxidant activity. In order to identify the antioxidative principles of V. uliginosum, we separated water extracts into polyphenol, anthocyanin-rich (pigment), and sugar/acid fractions by using ethyl acetate, acidic methanol (MeOH), and 0.01 N HCl. Antioxidant activities were assessed by 2,2-diphenyl-1-picrylhydrazyl (DPPH), superoxide radical, and hydroxyl radical assays. The crude extract and fractions containing polyphenol and pigment exhibited the greatest antioxidant activities with 50% inhibitory concentration (IC(50)) values of 85.8 microg/mL, 33.2 microg/mL, and 16.7 microg/mL, respectively, for the DPPH assay and 48.1 microg/mL, 83.8 microg/mL, and 51.9 microg/mL for the nonenzymatic superoxide radical assay. The fractions containing polyphenol, pigment, and sugar/acid significantly inhibited
xanthine oxidase
. To investigate the functional compounds from the active fractions, we purified the polyphenol fraction and separated the compounds by using chromatographic techniques. The crude extract was dissolved in MeOH and further purified by reversed-phase high-performance liquid chromatography (HPLC) using MeOH-water (35:65 vol/vol) (with 0.04%
trifluoroacetic acid
) to obtain VU-EA-1 (16.6 mg), VU-EA-2 (8.5 mg), VU-EA-3 (19.8 mg), VU-EA-4 (12.8 mg), VU-EA-5 (6.5 mg), and VU-EA-6 (23.5 mg). The MeOH-washed fraction from the HPLC was concentrated and purified by reversed-phase HPLC using MeOH-water (50:50 vol/vol) to give VU-EA-10 (12.4 mg). Antioxidant activity was assessed by DPPH, superoxide radical, and hydroxyl radical assays. The isolated compounds exhibited dose-dependent antioxidant activity with IC(50) values of 7.6 microg/mL (VU-EA-10) for the DPPH assay, 67.8 microg/mL (VU-EA-4) for the nonenzymatic superoxide radical assay, and 3.7 microg/mL (VU-EA-10) and 7.6 microg/ml (VU-EA-6) for the enzymatic superoxide radical assay and 30% inhibitory concentration values of 0.58 microg/mL (VU-EA-1), 0.57 microg/mL (VU-EA-5), and 0.70 microg/mL (VU-EA-6) for the hydroxyl radical assay. In conclusion, V. uliginosum had potent antioxidative activity, and flavonoids were isolated as the main active principles.
