Present laboratory tests for human
typhoid
vaccines use an intraperitoneal route of challenge given 7 days after injection of increasing doses of standard and test vaccines by the same route. In studies reported here, groups of B6D2 mice were vaccinated intraperitoneally with 2 x 10(8) acetone-killed Salmonella typhi Ty2, with the Vi antigen-free variant O-901, or with Yersinia enterocolitica and Serratia marcescens suspensions. Other groups of mice received 200 mug of purified S. typhi or S. marcescens endotoxin, or their corresponding purified lipid A components. All of the vaccinated mice (except for saline- or thioglycolate-injected controls) exhibited increased protection against the lethal intraperitoneal challenge with S. typhi Ty2. Serial quantitative bacterial counts carried out on peritoneal washouts and on homogenates of the draining mediastinal lymph nodes indicated the development of an antibacterial response by the vaccinated host which was not observed in the control animals. Mice receiving purified endotoxin (
lipopolysaccharide
) exhibited varying degrees of protection, both in terms of increased host survival and the amount of inactivation of the challenge population in vivo. The response seen when the antigenically unrelated S. marcescens
lipopolysaccharide
was injected was little different from that seen when the acetone-killed S. typhi Ty2 whole-cell vaccine was used. This suggests that nonspecific inactivation of the intraperitoneal challenge contributes substantially to the immune response seen in mice vaccinated intraperitoneally with specific
typhoid
antigens.

Distinct increase in amount of total lipids occurred in mice liver tissue after intraperitoneal administration of
typhoid
lipids A and B (the latter was nonhydrolytically isolated from
typhoid
lipopolysaccharides). The alteration referred primarily to neutral lipids, especially to triacyl glycerides, content of which exceeded 3- and more-fold the initial one to the end of a day. The similarity was observed in alteration of triacyl glycerides, phospho- and glycolipids in presence of both lipid preparations. These lipid components resembled intact

from
typhoid
bacteria in their effect on content of lipids in liver tissue. In treatment with these lipid preparations concentration of NEFA was either unaltered or distinctly decreased.

The effect of
typhoid
lipopolysaccharide

) and polysaccharide (PS) on immunological memory in the system of IgM and IgG synthesis and rosette-forming cells was studied. When introduced into animals previously immunized with SRBC, PS stimulated, under certain conditions, immunological memory in the system of IgM synthesis and rosette-forming cells, while the injection of
LPS
induced only an insignificant stimulation of immunological memory. No stimulation in the system of IgG synthesis was observed after the injection of both
LPS
and PS. The suppression of immunological memory was noted in the animals receiving
LPS
as well as in those receiving PS the immunosuppressing effect produced by
LPS
was more pronounced.

Content of phospholipid fractions was altered in mice liver tissue after intraperitoneal administration of

. The intoxication was accompanied by an increase in content of choline-containing phospholipids within 6 and 24 hrs as well as by an alteration in the ratio of the fractions. Effect of lipid A on the tissue phospholipid spectra resembled the effect of the

Sublethal doses of
typhoid
lipopolysaccharide
increased distinctly the content of total lipids in mice liver tissue. Liver obesity was mainly related to accumulation of triacyl glycerols, the level of which exceeded the initial one 6.2-fold within a day. Mono- and diacyl glycerols also accumulated in liver tissue within the same period. Content of free fatty acids was distinctly higher in experimental animals than in control mice. Pronounced increase in free cholesterol content was observed within 3 hrs after the
lipopolysaccharide
administration; but within a day the content of cholesterol was markedly lower in the animals as compared with the control mice. The intoxication was accompanied by a decrease in glycolipid content within 3 and 6 hrs but towards the end of the 24-hrs period content of these lipids did not differ from the initial level in liver tissue. The endotoxin did not alter distinctly the total phospholipid content in liver tissue.

The monoclonal antibodies to porin, an outer membrane protein isolated from Salmonella typhimurium and sandwich enzyme linked immunosorbent assay (ELISA) has made possible the detection of porin from sera of S. typhimurium-infected mice. The specificity of the monoclonal antibodies was ascertained based on their cross-reactivity with porins isolated from S. typhi, Shigella flexneri and Escherichia coli and
lipopolysaccharide

) of S. typhimurium and E. coli. Serum samples were found to be positive for porin as early as 3 days after intravenous and 5 days after oral infection. In addition, a positive correlation was observed between the bacterial load and the concentration of porin detected in the sera. On the other hand, analysis of sera for anti-porin antibody showed diametrically opposite time kinetics with antigenaemia. These results indicate that porin accumulates in the serum of infected mice much earlier than the appearance of antibodies to porin. Thus detection of porin holds promise for early diagnosis of

An ELISA was evaluated for the serodiagnosis of fowl
typhoid
and paratyphoid due to Salmonella enteritidis in chickens. The hot phenol: water
lipopolysaccharide
(
LPS
) extract of Salmonella was used as the antigen. Chicken serum, eggs and discs impregnated with chicken blood were tested for the presence of antibodies against Salmonella factor 'O' 9 antigen. The substrate and chromogen used were hydrogen peroxide and orthophenylenediamine respectively. Serological results from the experimentally and naturally infected chickens showed close agreement between the conventional Serum Tube Agglutination Test (SAT) and serum ELISA while serum ELISA results were in close agreement with the egg and disc ELISA results. It was noted that ELISA was highly sensitive, convenient and versatile. It is concluded that ELISA, especially disc ELISA, ought to replace SAT for seroscreening chickens against S. gallinarum and other Salmonella Group D infections.

The serological response to Salmonella pullorum and S. gallinarum infection in chickens was studied with an indirect enzyme-linked immunosorbent assay (ELISA). In broiler chickens, a more virulent strain of S. pullorum produced a significantly lower serum IgG titer than did a less virulent strain. In laying hens, the serum and egg-yolk IgG titers were very similar. In chickens infected with S. gallinarum, high IgG titers persisted for 30 weeks. In chickens reinfected with this strain, each reinfection was followed by transitory increases in IgG lasting no longer than 2 weeks. showed much higher antibody levels than did those from three uninfected flocks. Using

as the detecting antigen, infections caused by these salmonellae could be differentiated from those caused by other groups. Incorporation of the appropriate flagella antigen in the ELISA allowed differentiation between infections caused by S. pullorum and S. enteritidis.
