The microstructure and permeability of rehydrated 20-100 microm thick partially coalesced (
vinyl
-actetate acrylic copolymer) SF091 latex coatings and a 118 microm thick model trilayer biocatalytic coating consisting of two sealant SF091 layers containing a middle layer of viable E. coli HB101 + latex were studied as delaminated films in a diffusion apparatus with
KNO
(3) as the diffussant. The permeability of the hydrated coatings is due to diffusive transport through the pore space between the partially coalesced SF091 latex particles. Coating microstructure was visualized by fast freeze cryogenic scanning electron microscopy (cryo-SEM). The effective diffusion coefficient of SF091 latex coatings (diffusive permeability/film thickness) was determined as the ratio of the effective diffusivity of
KNO
(3) to its diffusivity in water (D(eff)/D). Polymer particle coalescence was arrested by two methods to increase coating permeability. The first used glycerol with coating drying at 4 degrees C, near the glass transition temperature (T(g)). The second method used sucrose or trehalose as a filler to arrest coalescence; the filler was then dissolved away. D(eff)/D was measured as a function of film thickness; content of glycerol, sucrose, and trehalose; drying time; and rehydration time. D(eff)/D varied from 3 x 10(-4) for unmodified SF091 coatings to 6.8 x 10(-2) for coatings containing sucrose. D(eff)/D was reduced by the flattening of latex particles against the surface of the solid substrate, as well as by the presence of the colloid stabilizer hydroxyethylcellulose (HEC). When corrected for the flattened particle layer, D(eff)/D of HEC-free coatings was as high as 0.20, which agreed with the value predicted from analysis of cryo-SEM images of the coat surface. D(eff)/D decreased by one-half in approximately 5 days in rehydrated SF091 coatings, indicating that significant wet coalescence occurs after glycerol, sucrose, or trehalose are leached from the films. D(eff)/D of SF091 latex trilayer coatings containing viable E. coli HB101 cells decreased as cell loading was increased from 2.2 x 10(-2) for 64 g dry cell weight per liter of coat volume to 5 x 10(-3) for 151 g DCW/L of coat volume. The reduction in coating permeability with increasing cell loading is predicted by Maxwell's equation for D(eff)/D in periodic composites.

The ionomer poly(vinylbenzyltrimethylammonium chloride) has been blended with cross-linked poly(
vinyl
alcohol) to form optically clear composites that can be covalently linked as thin films to oxide surfaces. Films are characterized using spectroscopic ellipsometry with refractive index (n) and extinction coefficient (k) data presented for wavelengths 300 to 1100 nm. A refractive index of 1.54 and average thickness of 709 nm are typical of an air dry film prepared by spin-coating. Dynamic in situ ellipsometry results for films exposed to 0.1 M
KNO
(3) and 1.0 mM Fe(CN)(6)(-3) are presented. Upon initial exposure to 0.1 M
KNO
(3), an air dry film expands by about 160% and stabilizes in size and refractive index at about 18 hours. Ion exchange of film cationic groups with ferricyanide is marked by slight film contraction, presumably due to electrostatic cross-linking by the multivalent anion. These films are useful in the spectroelectrochemical sensor with our newly developed fluorescence detection mode, as demonstrated by results of the reversible incorporation of the fluorescent anion fluorescein.

The success of surgery to remove primary tumors can be compromised by the subsequent outgrowth of metastases. It is recognized that primary tumors secrete antiangiogenic factors that suppress the outgrowth of their daughter metastases. In accord we show here that surgical removal of primary EL-4 lymphomas led to a marked decrease in the levels of circulating angiostatin and
endostatin
, and promoted the growth of distant nodular tumors. Expression vectors encoding angiostatin and
endostatin
, formulated with poly-N-
vinyl
pyrrolidone (PVP), were injected into the tibialis and gastrocnemia muscles, leading to expression of angiostatin and
endostatin
in muscle fibers. High levels of biologically active exogenous proteins were secreted into the circulation. Intramuscular gene therapy with angiostatin and
endostatin
plasmids significantly inhibited tumor vascularity and induced tumor cell apoptosis, and thereby suppressed the growth of secondary subcutaneous and disseminated metastatic tumors in the lung and liver. Simultaneous intramuscular delivery of both angiostatin and
endostatin
plasmids significantly prolonged the survival of mice after removal of primary tumors. These results suggest that intramuscular gene transfer of angiostatin and
endostatin
might serve as a prophylactic cancer-prevention strategy to combat the recurrence of cancer after surgical resection of primary tumors.

In this study, an optimal process to prepare a synthetic filter material (poly(
vinyl
alcohol) (PVA)/peat/
KNO
(3) composite bead) containing nutrients was developed for biofiltration. The optimal preparing condition was that each of the peat and PVA aqueous solutions contains 6.4 g
KNO
(3) and the nitrogen content in the boric and phosphate aqueous solutions must retain higher than 3.94 and 1.52 g N/l, respectively. The equilibrium amount of water-soluble nitrogen dissolved out of the prepared composite bead was between 7.95 and 8.21mg N/g dry solid. The path of water-soluble nitrogen dissolving out of the A-type bead was the water-soluble nitrogen dispersed in the peat phase initially diffused into the outer PVA phase and then it diffused out of the bead surface. And the path of water-soluble nitrogen dissolving out of the H-type bead was the water-soluble nitrogen dispersed in both the peat and PVA phases simultaneously diffused into the outer PVA phase and out of the bead surface, respectively. The microbial growth rate k(g) of the H-type composite bead was higher than that of the A-type composite bead approximately 1.09-1.58 times, and its value was between 0.100 and 0.417 day(-1) as the composite bead was immersed in 0-0.896 M
KNO
(3) solution. The maximum value of k(g) appeared at the composite bead immersed in 0.384 M
KNO
(3) solution and was higher than that of the compost by a factor approximately 1.49. The percentage of removed volatile organic compounds (VOCs) remained at more than 98% during the biofilter operating 230 days as the composite bead was immersed in
KNO
(3) aqueous solution before packing. This composite bed was without the further addition of nutrients during this operating period. It was proved that this composite bead was superior to the compost as a filter material.

The composite films of ferroelectric potassium nitrate (KNO3):poly(
vinyl
alcohol) (PVA) with different weight percentages of KNO3 have been prepared at 200 degrees C using the spray-deposition technique. The remanent polarization (Pr) and peak current density for all composite films was estimated by tracing the polarization-electric field (P-E) hysteresis loop and current density-electric field (J-E) loop, respectively, using a modified Sawyer-Tower circuit. Pure KNO3 is known not to exhibit any ferroelectricity under ambient conditions, but the X-ray diffraction (XRD) studies of PVA:KNO3 composite films reveal the presence of a ferroelectric phase III of KNO3 in the composite films at room temperature. The composite film containing
KNO
(3):PVA in equal proportions shows maximum Pr and peak intensity ratio of approximately 20.10 microC/cm2 and 2.67, respectively, at room temperature. The J-E and capacitance voltage (C-V) characteristics exhibit butterfly features that supports the presence of a ferroelectric phase in the composite films. The field emission scanning electron microscopy (FE-SEM) image of the composite film containing equal proportions of KNO3 and PVA shows the homogenous distribution of spherical grains of KNO3 of size approximately 225 nm.
