The interaction has been studied in aqueous solutions between a negatively charged conjugated polyelectrolyte poly{1,4-phenylene-[9,9-bis(4-phenoxybutylsulfonate)]fluorene-2,7-diyl} copolymer (
PBS
-PFP) and several cationic tetraalkylammonium surfactants with different structures (alkyl chain length, counterion, or double alkyl chain), with tetramethylammonium cations and with the anionic surfactant sodium dodecyl
sulfate
(SDS) by electronic absorption and emission spectroscopy and by conductivity measurements. The results are compared with those previously obtained on the interaction of the same polymer with the nonionic surfactant C12E5. The nature of the electrostatic or hydrophobic polymer-surfactant interactions leads to very different behavior. The polymer induces the aggregation with the cationic surfactants at concentrations well below the critical micelle concentration, while this is inhibited with the anionic SDS, as demonstrated from conductivity measurements. The interaction with cationic surfactants only shows a small dependence on alkyl chain length or counterion and is suggested to be dominated by electrostatic interactions. In contrast to previous studies with the nonionic C12E5, both the cationic and the anionic surfactants quench the

-PFP emission intensity, leading also to a decrease in the polymer emission lifetime. However, the interaction with these cationic surfactants leads to the appearance of a new emission band (approximately 525 nm), which may be due to energy hopping to defect sites due to the increase of

-PFP interchain interaction favored by charge neutralization of the anionic polymer by cationic surfactant and by hydrophobic interactions involving the surfactant alkyl chains, since the same green band is not observed by adding either tetramethylammonium hydroxide or chloride. This effect suggests that the cationic surfactants are changing the nature of
PBS
-PFP aggregates. The nature of the polymer and surfactant interactions can, thus, be used to control the spectroscopic and conductivity properties of the polymer, which may have implications in its applications.

Human haptoglobin (Hp) is classified as three phenotypes: Hp 1-1, 2-1, and 2-2. Previously, we had isolated this protein by affinity columns using either hemoglobin or monoclonal antibody (mAb) prepared against Hp beta-chain (clone 8B1-3A). The isolated Hp from both methods, however, contaminates plasma apolipoprotein A-I (apoA-I). In the present report, we have developed a novel affinity column procedure using an mAb prepared against alpha-chain of Hp (clone 3H8) for Hp purification. Plasma was first chromatographed onto the column followed by a normal wash with a buffer containing 0.12 M NaCl and 0.02 M phosphate, pH 7.4 (

Our study examined whether repeated preventive oral administration of live probiotic bacterial strains Escherichia coli O83:K24:H31 (Ec O83), Escherichia coli Nissle 1917 O6:K5:H1 (Ec Nis) and Lactobacillus casei DN 114001 (Lc) can protect mice against dextran sodium
sulfate
(DSS)-induced colitis. A significant decrease in average symptom score was observed in Ec O83-, Ec Nis- and Lc-pretreated group (p < 0.05). Significant differences in body mass loss between Lc pretreated mice with DSS-induced colitis were found when compared with nontreated mice (p < 0.05).

Administration of Lc significantly decreased the severity of DSS induced histological marks of inflammation (p < 0.05). The aim of this work was to design a new multifunctional nano device (MND) for gene delivery. This MND was equipped with folic acid as ligand, which was conjugated to terminal amido of poly(aminopoly(ethylene glycol)cyanoacrylate-co-hexadecyl cyanoacrylate) (poly(H(2)NPEGCA-co-HDCA)) to synthesize poly(Folate-HNPEGCA-co-HDCA), protamine
sulfate
(PS) as DNA condenser and for nuclear transfer, PEG chain from poly(Folate-HNPEGCA-co-HDCA) for decreasing macrophages recognition and extending half-life, dioleoyl phosphatidylethanolamine (DOPE) for endosomal escape, and we supposed that the latent DOPE fusogenicity could be gently restored along with fast degradation of poly(Folate-HNPEGCA-co-HDCA) in MND membrane within endosome. Our experimental results showed that optimum complexation ( approximately 97%) of DNA was achieved at DNA:PS=1:3 (w/w). The MND showed different loading ratio by lipid film hydration technique with the highest loading ratio about 12%, the particle size range 200-400 nm, surface charge range 8 mV-15 mV. Basic fibroblast growth factor (bFGF) was immobilized onto quartz slides and collagen films by assembly with chondroitin
sulfate
(CS) in a layer-by-layer (LBL) manner. First, the LBL-deposition process on the amino-silanized quartz slides was monitored by UV-vis spectroscopy and water contact angle measurement. By substituting the normal bFGF with rhodamine-labeled one (Rd-bFGF), a linear increase of the absorbance versus bilayer number was recorded. The water contact angle oscillated between the odd CS and the even bFGF layers, demonstrating the alternating change of the surface chemistry. Scanning force microscopy (SFM) revealed that the surface topography was altered slightly after multilayer assembly. In vitro incubation of the CS/bFGF multilayers in

showed that approximately 30% of the incorporated bFGF was released within 8 days. In vitro cell culture found that the fibroblasts showed star-like morphology with plenty of pseudopods on the bFGF-incorporated collagen film after cultured for 1 day, and the collagen films assembled with bFGF possess improved bioactivity than that of the virgin one and the bFGF control. Since the immobilized growth factors can maximally retain their bioactivity, the LBL assembly would be a potential approach to construct a bioactive substrate for biomedical applications.

Puerarin, a natural isoflavonoid found in Chinese Pueraria lobata (Wild.) Ohwi, has received increasing attention because of its possible role in the prevention of osteoporosis. However, the relationship between puerarin and bone formation remains unknown. In the present study, rat osteoblasts isolated from newborn Wistar rats were used to investigate the effect of puerarin on osteoblasts, and its possible molecular mechanism. Data showed that puerarin caused a significant increase in cell viability, alkaline phosphatase (ALP) activity and mineral nodules formation in osteoblasts, suggesting that puerarin had a stimulatory effect on osteoblastic bone formation. This functional improvement by puerarin was accompanied by activation and nuclear translocation of Akt. Furthermore, puerarin-stimulated osteoblastic growth, Akt activation and redistribution were significantly blocked by the specific PI3K inhibitor, LY294002. These results strongly suggested that puerarin stimulated osteoblastic proliferation and Akt activation in a PI3K-dependent manner. In summary, puerarin derived from Chinese Pueraria lobata (Wild.) In this study, the fluorescein isothiocyanate (FITC) labeled otcadecylamine (ODA), otcadecylamine-fluorescein isothiocyanate (ODA-FITC), was synthesized and used as a fluorescence marker to be incorporated into stearic acid solid lipid nanoparticles (SLN) by solvent diffusion method. Approximately 97.9% of added ODA-FITC was incorporated into SLN. Under sink condition, approximately 7% and less than 3% of ODA-FITC leaked from SLN in 24 h, when the ODA-FITC loaded SLNs were dispersed in plasma or phosphate buffered saline (

sulfate
(SDS), respectively. The ODA-FITC loaded SLNs were then subjected to the in vivo transport experiments. The results showed that the transport efficiency of SLN by oral administration was 30%. The SLN could be extensively absorbed, and indicated a linear absorption mechanism in gastrointestinal tract within certain range of concentrations. By the external diversion experiments on lymph, it showed that approximately 77.9% of absorbed SLN was transported into systematic circulation via lymph, which is a major SLN transport pathway in gastrointestinal tract. The rest of absorbed SLN was transported directly into blood, which might be through capillary vessel or intestinal epithelial cell by paracellular pathway. The further experiment demonstrated that the polyethylene glycol monostearate (PEG2000-SA)-modified SLN could also be absorbed in gastrointestinal tract and achieved a prolonged effect in vivo.

Based on the data from these experiments, further experiments tested how different sugars such as raffinose, trehalose, lactose, fructose, and glucose dissolved in mKRB with Equex STM, SDS and egg yolk supplementation affected the post-thaw characteristics of cryopreserved sperm. Cryosurvival of frozen-thawed sperm were judged by microscopic assessment of the sperm motility index (SMI), and acrosome integrity was measured using FITC-PNA staining. Thawed sperm were subjected to 3h of a thermal resistance test. Beneficial effects on the post-thaw survival of sperm were obtained when 0.1M raffinose in mKRB was used with 0.75% Equex STM, 0.05% SDS, and 20% egg yolk. Sperm cryopreserved with this treatment exhibited a higher motility index and maintained greater SMI and acrosome integrity throughout incubation when compared to sperm frozen in various concentrations of other cryoprotectants and trehalose, lactose, fructose, glucose. In conclusion, cryopreservation in an extender solution of raffinose dissolved in mKRB containing Equex STM, SDS and egg yolk greatly enhances the freezability of rat epididymal sperm.
