Several mucolytic agents were evaluated on sputum for testing their viscolytic activity and the bacterial tollerance to each of them. Proteolytic enzymes (trypsin, pepsin,
papain
, pancreatin), KJ, and dithiothreitol (or its derivatives) were better tollerated by common respiratory pathogens (H. influenzae, D. pneumoniae, Klebsiella, etc.) than other mucolytic agents, as acetil-cysteine, cisteamine-HCl, tension active substances, mercaptoethanol, and others. The dithiothreitol showed also one of the strongest viscolytic effect and therefore it was selected for the routinary sputum digestion at the concentration 0.1% in
PBS
pH 7.2. Such a solution was added to sputum specimen in different proportions according to the macroscopic "apparent" viscosity of each specimen. However researches on the comparative viscolytic activity of all the agents hereinafter considered are still in progress.

Formation of rosettes with sheep RBC by blood lymphocytes from young pigs was increased from 24.4% +/- 2.2 (mean +/- S.E.) in
PBS
to 54.1% +/- 1.9 in the presence of dextran. This increase was achieved without inducing appreciable rosette formation with other RBC which do not form rosettes in
PBS
. Lymphocytes which rosette only in dextran are predominant in pigs between 20 and 160 days old, when the peripheral lymphocyte pool is increasing very rapidly. The use of dextran revealed major populations of blood lymphocytes rosetting with SRBC in adult sheep (30.7% +/- 2.0), adult cattle (37.3% +/- 4.2) and adult goats (13.3% +/- 1.2). Proportions of rosette-forming lymphocytes tended to increase with age. In calf lymphoid tissues the distribution of rosette-forming lymphocytes suggested that these were T cells. In the improvement of rosette formation with SRBC, dextran was more effective than foetal calf serum,
papain
treatment of the SRBC or combinations of these treatments.

The only established difference on which to base the separation of X and Y chromosome-bearing spermatozoa is chromosomal constitution. This difference is quantifiable both from chromosome morphology (karyotype) and from DNA content. Flow cytometric techniques were used to measure relative DNA content of the X and Y populations and to flow-sort spermatozoa from Chinchilla laniger. Epididymal spermatozoa were recovered in
PBS
, fixed in 80% ethanol, treated with
papain
and dithioerythritol, and stained for DNA with Hoechst 33342. Sperm nuclei were analyzed and sorted on an EPICS V flow cytometer/cell sorter, modified specifically for spermatozoa. Two clearly resolved peaks (coefficient of variation less than 1.5%) with approximately 7.5% difference in DNA content between X and Y chromosome-bearing spermatozoa were evident. Sperm nuclei were sorted from a portion of the X and Y peaks at a rate of 55 nuclei/sec for each population. Purities of individual X and Y populations averaged 95% as determined by reanalysis of the sorted populations. Successful sorting of Chinchilla X and Y chromosome-bearing spermatozoa into separate populations may aid in the identification of a biochemical marker that could be used to discriminate between the two sperm populations and lead to a practical procedure for sexing spermatozoa.

The antiallergic effect of Fc fragments prepared from human polyclonal IgG was investigated in several experimental models. In an in vitro assay, isolated ileum of cynomolgus monkeys was sensitized with serum from atopic patients. In six of fifteen monkeys the subsequent addition of specific allergens reproducibly resulted in an ileum contraction measured in the Schultz-Dale apparatus. In all six positive monkeys pre-treatment of the isolated ileum with Fc (
papain
) before sensitization inhibited ileum contraction. Fc (plasmin) or Fc (pepsin) was less or not effective, aggregated IgG, monomeric IgG, sulfonated IgG, the Fc-free F(ab')2 moiety, or albumin had no significant or no reproducible inhibitory effect. In an in vivo assay passive cutaneous anaphylaxis was studied in 28 cynomolgus monkeys. Different dilutions in

of the particular specific allergens were subsequently administered to sensitized skin sites, simultaneously to i.v. injection of Evans blue. The degree of the local allergic (passive cutaneous anaphylactia) reaction was evaluated by measuring the diameter of the blue area at the dermal injection sites. About 50% of sera induced positive reaction in monkeys. Compared to the control (application of

), the degree of the positive reactions could be reduced by about 50% if Fc (
papain
) (optimal dose 25 mg/kg body weight) was injected i.v. either 2 h before or after local sensitization. No significant inhibitory effect could be found with the Fab moiety of IgG. Follow-up studies showed that the inhibitory effect of Fc lasted for about 10 days. The pharmacological basis of the observed antiallergic action of Fc is not yet understood.

There have been no reported studies on the Feulgen-DNA cytofluorometry of the cartilage cells. We have attempted to devise a method of cell separation from the epiphyseal and articular cartilages of the rats, and to analyze by cytofluorometry the changes in the ploidy patterns of these chondrocytes during growth and ageing of the animals. Chondrocytes were isolated from the proximal cartilage of tibia by dual enzymatic digestions of the cartilage matrix with
papain
and collagenase, followed by mechanical cell separation with scissors and a micro-homogenizer, and were smeared onto the object glass with
PBS
. These procedures were found to be suitable for the Feulgen-DNA cytofluorometry of the chondrocytes from our repeated studies. We also carried out Feulgen-DNA cytofluorometry combined with 3H-thymidine autoradiography to determine cellular DNA content of the DNA synthetic chondrocytes in the epiphyseal cartilage. It has been clarified that during the growth course of the rats, the chondrocytes of the epiphyseal cartilage consist of many mononuclear diploid cells, a few mononuclear tetraploid cells and of some fraction of the cells having intermediate DNA values between the diploid and tetraploid levels. Those cells with intermediate DNA values, after autoradiographic studies, were found to correspond to DNA synthetic cells, indicating cell proliferative activity. It has been shown that during ageing of the rats, most of the chondrocytes from the articular cartilage are mononuclear diploid cells. The distribution of each cellular DNA content at the diploid level as determined by Feulgen-DNA cytofluorometry was shown to become gradually broader.

Quantitative and morphological analyses (in SEM) of blood platelets collected from the left and right ventricles of the rat heart in the course of experimental lung emphysema were done. Platelet aggregation index was estimated, too. Emphysema was induced by a single intratracheal instillation of
papain
solution in a dose of 20 mg/kg b.w/1 ml

. The animals were sacrificed after 2 and 24 hours and 7, 14, 28 days later. Within 24 hours of the experiment a slight decrease was observed in the number of platelets in the blood collected from the left ventricle compared to the right one as well as to control animals. Also a reduction in platelet aggregation coefficient value was noted. However, in the later period of emphysema progression (after 7th day), a statistically significant increase was found in the number of blood platelets in the left ventricle. A relation was noted between quantitative changes of blood platelets and emphysema progression evaluated morphometrically. The ultrastructural examinations in SEM suggest the occurrence of platelet satellitosis in animals intratracheally injected with
papain
solution. The present results indicate the possibility of a significant contribution of blood platelets to the pathogenesis of experimental lung emphysema.

In the last years pentoxifylline (PTXF) has been successfully used in reducing acute injury of the lung parenchyma in ARDS. The authors have studied the effect of PTXF on degree of
papain
induced lung injury. Papain was administered intratracheally in a dose of 2 mg, 4 mg, 6 mg in 1 ml of
PBS
. The effect of
papain
on bronchoalveolar lavage characteristics was also evaluated. Wistar rats of both sexes were used in the experiment. The degree of lung destruction, reflected by interstitial hemorrhage was assessed by measuring hemoglobin content in the fluid of the lavaged lungs. The hemoglobin levels were assessed spectrofluorometrically with the use of the 414 nm wave length. A reduction of hemoglobin content was seen after PTXF administration only in animals receiving the lowest dosage of
papain
. However in all animals a decrease in the BAL neutrophil count was demonstrated. The protective effect of PTXF on pulmonary tissue in
papain
induced injury models and situations leading to development of ARDS may suggest a similar pathomechanism in both entities.

Polyrotaxanes were synthesized as novel biodegradable polymers with supramolecular assembly and their properties evaluated in vitro. The synthesis of biodegradable polyrotaxanes consists of three steps: preparation of an inclusion complex consisting of alpha-cyclodextrins (alpha-CDs) and amino-terminated poly(ethylene glycol) (PEG); introduction of L-phenylalanine (L-Phc) at each complex terminal via peptide linkages: and hydroxypropylation of alpha-CDs in the polyrotaxanes. Succinimide ester of benzyloxycarbonyl-L-Phe was condensed with the terminal amino groups of the inclusion complex. 1H-NMR and GPC results showed that alpha-CDs were threaded onto a PEG chain and L-Phe moieties were introduced at each terminal of the PEG chain. Further, the amount of threaded alpha-CDs was found to be governed by the molecular weight of PEG. The hydroxypropylation of alpha-CDs improved the solubility of the polyrotaxanes in
PBS
(pH 7.4). The hydroxypropylated (HP-) polyrotaxanes were characterized by terminal peptide cleavage using
papain
. In vitro degradation of HP-polyrotaxanes revealed that HP-alpha-CDs threaded onto a PEG chain were released only when terminal peptide linkages were cleaved. Moreover, threaded HP-alpha-CDs onto a PEG chain was found to be completely released. Kinetics of terminal peptide cleavage were also evaluated by catalytic efficiency (kcat/K(m)). The kcat/K(m) values were found to be independent of the molecular weight of HP-polyrotaxanes but to be affected by terminal hydrophobic moieties. It is proposed that our designed polyrotaxanes are feasible as novel drug carriers.

The aim of this study was the ultrastructural analysis of the changes in the endothelium of myocardial capillaries and the processes observed in their vicinity in consecutive stages of experimental lung emphysema. . Ultrastructural analysis of the changes within the right ventricle and the subendocardial part of the left ventricle of the heart was made by a transmission electron microscope after 1, 3 and 6 months following
papain
administration. Special attention was paid to the analysis of changes found in the vicinity of newly formed myocardial capillaries. Animals killed after 1 and 3 months following
papain
infusion showed blurred borderline between the endothelium of the newly formed myocardial capillaries and perivascular connective matrix. The new vessels had poorly developed basement membrane. Fibrillary structures and/or collagen fibrils were found instead. The latter frequently demonstrated features of disorders in the structure and spatial distribution. Neither fibroblasts nor their processes were observed in the vicinity of the new collagen fibrils. Pericytes were occasionally present. In the later period (6 months from intratracheal
papain
infusion) well formed collagen fibres dominated in the vicinity of myocardial capillaries. The results of the ultrastructural analysis suggest the possibility of active contribution of endothelial cells to the processes of perivascular fibrosis within the myocardium in the course of experimental lung emphysema.
