Kinins and
substance P
have been implicated in the pathogenesis of inflammatory arthritis by virtue of their abilities to induce vasodilation, edema, and pain. The relative biological potencies of these peptides in vivo would depend at least in part upon their rates of catabolism in the joint. We hypothesized that human synovial lining cells may regulate intraarticular levels of kinins and neuropeptides via degradation by cell surface-associated peptidases. We exposed intact human synovial fibroblasts to kinins and
substance P
, in the presence or absence of specific
peptidase
inhibitors, and measured the amount of intact substrate remaining and degradation product(s) generated over time. Aminopeptidase M (AmM; EC 3.4.11.2), neutral endopeptidase-24.11 (NEP-24.11; EC 3.4.24.11), and dipeptidyl(amino)
peptidase
IV (DAP IV; EC 3.4.14.5) were identified on the cell surface of synovial cells. Bradykinin degradation was due entirely to NEP-24.11 (1.39 +/- 0.29 nmol/min per well). Lysylbradykinin was also degraded by NEP-24.11 (0.80 +/- 0.19 nmol/min per well); however, in the presence of phosphoramidon, AmM-mediated conversion to bradykinin (3.74 +/- 0.46 nmol/min per well) could be demonstrated. The combined actions of NEP-24.11 (0.93 +/- 0.15 nmol/min per well) and DAP IV (0.84 +/- 0.18 nmol/min per well) were responsible for the degradation of
substance P
. AmM (2.44 +/- 0.33 nmol/min per well) and NEP-24.11 (1.30 +/- 0.45 nmol/min per well) were responsible for the degradation of the opioid peptide, [Leu5]enkephalin. The identity of each of the three peptidases was confirmed via synthetic substrate hydrolysis, inhibition profile, and immunological identification. The profiles of
peptidase
enzymes identified in cells derived from rheumatoid and osteoarthritic joints were identical. These data demonstrate the human synovial fibroblast to be a rich source of three specific peptidases and suggest that it may play a prominent role in regulating peptide levels in the joint.

The neuropeptide
substance P
(SP) has been localized to amacrine and ganglion cells in the rabbit retina. We have examined the effects of SP and related peptides on rabbit retinal neurons using bath application and intra- and extra-cellular electrophysiological methods in an in vitro retina eyecup preparation.
Substance P
, at concentrations as low as 25 nM, moderately excited most brisk ganglion cells. SP excited some ganglion cells directly during cobalt block of synaptic transmission. Intracellular recordings from amacrine cells demonstrated that some, but not all, were depolarized by SP; pharmacological evidence suggested GABAergic amacrines were probably among those sensitive to SP. SP did not affect horizontal cells or the ERG, suggesting that the effects of this peptide are confined to the inner retina. The effects of SP were strongly potentiated by
peptidase
inhibitors, raising the possibility that endogenously released SP may act quite locally in the rabbit retina. The relative potencies of SP and the related peptides

and eledoisin on different cells suggest that more than one
tachykinin
receptor subtype is present in the rabbit retina. The responses of ganglion cells to SP desensitized with repeated or prolonged applications. Comparison of a cell's light responses before and after the receptors were desensitized revealed no qualitative changes in receptive field characteristics, but quantitative changes in excitability were apparent. SP antagonist analogs, although not potent, specifically blocked the effects of SP on some ganglion cells. The influence of degradation by peptidases on concentration-response relationships for peptide agonists of the
tachykinin
and bombesin-like families was investigated. The combined presence of three
peptidase
inhibitors, phosphoramidon (1 microM), captopril (1 microM) and bestatin (100 microM), had no significant effect on the onset rates or peak contractile responses to these peptides in the rat urinary bladder and guinea-pig taenia caeci preparations, or on their peak potentiation of the contractile response to field-stimulation in the guinea-pig vas deferens preparation. However, rates of offset of the response to tachykinins were markedly prolonged in tissues treated with
peptidase
inhibitors. In experiments designed to estimate clearance of applied peptide from the organ bath, there was an initial rate of loss with the guinea-pig vas deferens and taenia caeci which, measured over the first 5 min, had a half-time of 2-3 min which was then prolonged to 6-8 min in the presence of
peptidase
inhibitors. These results show that although peptide breakdown can be demonstrated in these systems, it seems not to be an important determinant of relative pharmacological activity measured in terms of peak response.

We discovered an enzyme in human platelets that deamidates
substance P
and other tachykinins. Because an amidated carboxyl terminus is important for biological activity, we purified and characterized this deamidase. The enzyme, released from human platelets by thrombin, was purified to homogeneity by ammonium sulfate precipitation, followed by chromatography on an octyl-Sepharose column and chromatofocusing on PBE 94. The purified enzyme exhibits esterase,
peptidase
, and deamidase activities. The
peptidase
activity (with furylacryloyl-Phe-Phe) is optimal at pH 5.0 while the esterase (benzoyl-tyrosine ethyl ester) and deamidase (D-Ala2-Leu5-enkephalinamide) activities are optimal at pH 7.0. With biologically important peptides, the enzyme acts both as a deamidase (

-free acid, oxytocin-free acid) at neutrality, although the carboxypeptidase action is faster at pH 5.5. Enkephalins, released upon deamidation of enkephalinamides, were not cleaved. Gly9-NH2 of oxytocin was released without deamidation. Peptides with a penultimate Arg residue were not hydrolyzed. Some properties of the deamidase are similar to those reported for cathepsin A. The deamidase is inhibited by diisopropylfluorophosphate, inhibitors of chymotrypsin-type enzymes, and mercury compounds while other inhibitors of catheptic enzymes, trypsin-like enzymes, and metalloproteases were ineffective. In gel filtration, the native enzyme has an Mr = 94,000 while in non-reducing sodium dodecyl sulfate-polyacrylamide gel electrophoresis the Mr = 52,000 indicating it exists as a dimer. After reduction, deamidase dissociates into two chains of Mr = 33,000 and 21,000 as determined by sodium dodecyl sulfate-polyacrylamide gel electrophoresis. [3H]diisopropylfluorophosphate labeled the active site serine in the Mr = 33,000 chain. The first 25 amino acids of both chains were sequenced. They are identical with the sequences of the two chains of lysosomal "protective protein" which, in turn, has sequence similarity to the KEX1 gene product and carboxypeptidase Y of yeast. This protective protein complexes with beta-galactosidase and neuraminidase in lysosomes and is vitally important in maintaining their activity and stability. A defect in this protein is the cause of galactosialidosis, a severe genetic disorder. The ability of physiological stimuli (e.g. thrombin or collagen) to release the deamidase from platelets indicates that it may also be involved in the local metabolism of bioactive peptides.

tachykinin
on airway ciliary activity and its modulation by neutral endopeptidase EC 3.4.24.11 (NEP).
Neurokinin A
(
NKA
) caused dose-dependent increases in ciliary beat frequency (CBF), as measured by a photoelectric method, with the maximal increase from the baseline 15.7 +/- 1.7% (mean +/- SEM, p less than 0.01), whereas
substance P
(SP) had no effect. The
NKA
-induced increase in CBF was not inhibited by phentolamine, propranolol, or atropine, but it was abolished by the
tachykinin
antagonist [D-Pro2, D-Trp7,9]SP. Pretreatment of tissue with thiorphan (10(-5) M), a NEP inhibitor, had little effect on CBF responses to
NKA
; however, it significantly potentiated the responses to SP (14.9 +/- 3.0%, p less than 0.01). Other
peptidase
inhibitors, including captopril, bestatin, and leupeptin, did not alter the
tachykinin
-induced CBF response, suggesting that angiotensin converting enzyme, aminopeptidases, and serine proteinases do not modulate ciliary activity in response to tachykinins. These results suggest that
NKA
increases CBF by acting directly on
tachykinin
receptors and that NEP may play a role in modulating the
tachykinin
-induced stimulatory effects on CBF.

Enkephalinase (EC 3.4.24.11) is a naturally occurring, membrane-bound

in vivo and in vitro. Addition of this neutral endopeptidase to a rabbit eye cup model partially inhibits
substance P
-induced contraction of the iris sphincter muscle. Inactivation of
substance P
is reversed by thiorphan, a specific inhibitor of enkephalinase. These results show that enkephalinase degradation of
substance P
produces metabolites that are physiologically inactive in iris contraction. We also observed that atropine acts synergistically with enkephalinase to completely abolish
substance P
-induced iris contraction suggesting that the action of
substance P
on the iris contains an acetylcholine-stimulatory effect which is not lost by enkephalinase treatment.

result in naloxone-sensitive desensitization to this

-induced behavior, possibly through interactions of N-terminal fragments of

with mu opiate binding sites. The present investigation tests the hypothesis that

metabolites play a role in the development of desensitization to
substance P
by using the biting and scratching behavioral paradigm. While
substance P
-induced behaviors are produced by as little as 1 pmol of
substance P
, repeated injections of 7.5 pmol at 60-s intervals was found to be the minimum dose capable of causing desensitization. The C-terminal peptides, substance P3-11 and substance P5-11, elicited
substance P
-like behaviors, but repeated injection of these compounds did not result in desensitization to this behavior. In contrast to C-terminal fragments, intrathecal injection of N-terminal fragments, (substance P1-4, substance P1-7 and substance P1-9), did not elicit any overt

-like behaviors when administered alone, but when co-administered with

, decreased the magnitude of

Modulation of baroreceptor reflex (BRR) by endogenous
substance P
(SP) in the brain was investigated in rats anesthetized with pentobarbital sodium. Intracerebroventricular administration of the undecapeptide (15 or 30 nmol) and its antagonist, (D-Pro2, D-Trp7,9)-SP (30 or 60 nmol) or SP antiserum (1:20), respectively, promoted a significant increase and decrease in the sensitivity of BRR response. Prolonging the endogenous activity of SP with the aminopeptidase blocker, bestatin (200 nmol) or with the endopeptidase-24.11 inhibitor, phosphoramidon (200 nmol) significantly augmented the same reflex. Combining the undecapeptide with either
peptidase
blocker, moreover, promoted additional potentiation of the BRR response. On the other hand, simultaneous administration of bestatin and (D-Pro2, D-Trp7,9)-SP produced a reduction of the augmented effect of bestatin on the sensitivity of BRR response. Bilateral microinjection of SP (600 pmol) or an antiserum against SP (1:20) into the nucleus tractus solitarius (NTS) elicited respectively an enhancement of and reduction in the BRR response. These data suggest that neurons that contain SP may participate in central cardiovascular control by tonically enhancing the sensitivity of the BRR response, possibly via an action on the NTS.
