A new sea urchin lectin from Toxopneustes pileolus, is D(+)galactose (Gal)-, D(+)fucose (Fuc)-specific. Incubation of rat peritoneal mast cells with the lectin in the presence of 0.3 mM
CaCl2
for 10 min significantly and dose-dependently inhibited the histamine release induced by N-acetyl glucosamine (GlcNAc)-specific Datura stramonium agglutinin (DSA), an activator of the Gi-protein-dependent pathway in mast cells. This inhibition by the sea urchin lectin was sugar-specifically reversed in the presence of D(+)Gal or D(+)Fuc but not L(-)Fuc. The sea urchin lectin had no effect on the histamine release induced by compound 48/80, slightly inhibited the histamine release induced by
substance P
and mastoparan, and slightly enhanced the histamine release induced by melittin, but these effects were not dose-dependent. Compound 48/80,

, mastoparan and melittin are mast cell activators without sugar residues. It is suggested that the lectin binds to D(+)Gal residues of DSA to interfere with mast cell activation induced by DSA, a glycoprotein with arabinose and Gal residues. The effects of plant lectins with affinity to D(+)Gal, N-acetyl galactosamine and/or sialic acid and L(-)Fuc on the histamine release induced by DSA, compound 48/80 and
substance P
were also examined.

Substance P
(SP) is possibly involved in the pathophysiology of depression and anxiety. We investigated interactions between antidepressants on SP-induced effects and their potential calcium-blocking activity in the isolated guinea pig ileum. All the antidepressants tested, except pargyline, moclobemide, mianserin, and reboxetine, were able to inhibit in a concentration-dependent manner the contraction induced by 100 nmol/L SP. Clomipramine, fluoxetine, maprotiline, and amitriptyline (all at 3 mumol/L) flattened the concentration-response curves to SP, resulting in a reduction of up to 59%, 63%, 32%, and 23%, respectively, of the maximum contractile effect. All the antidepressants tested (3 mumol/L), except pargyline, moclobemide, and mianserin, produced a rightward parallel shift of the concentration-response curve to
CaCl2
. The L-type selective calcium blocker nifedipine and the T-type selective mibefradil showed similar behaviour against both agonists used, SP and
CaCl2
. The relative order of potency was nifedipine (pA2, 7.6 +/- 0.1) > clomipramine (pA2, 7.0 +/- 0.1) > fluoxetine (pKB, 6.5 +/- 0.1) = mibefradil (pKB, 6.6 +/- 0.1) > amitriptyline (pKB, 6.3 +/- 0.1) = maprotiline (pKB, 6.2 +/- 0.1) > fluvoxamine (pKB, 5.9 +/- 0.1). The data reported in the present study suggest that the antidepressants tested did not behave as competitive antagonists versus NK1-receptor subtypes, but their inhibitory action seems to be related to their calcium-blocking properties.

An 8-year field experiment was conducted in the Taihu Lake region of eastern China to investigate the effects of incorporation of straw and manure on the yield and phosphorus (P) accumulation in the paddy soil, and to evaluate the potential risk of P loss from soil to environment. The experiment had four fertilization treatments, i. e., chemical fertilizers alone (
NPK
), chemical fertilizers plus rice/wheat straw (
NPK
+ S), chemical fertilizers plus 7.5 t x (hm2 x a)(-1) wet pig manure (
NPK
+ M7.5), and chemical fertilizers plus 15.0 t x (hm2 x a)(-1) wet pig manure (
NPK
+ M15). Among the four treatments, no significant differences were observed in the yield of rice or wheat. Long-term application of chemical fertilizers plus pig manure significantly increased the soil total P, the degree of P saturation (DPS), and the concentration of extractable P forms, including Olsen-P, Mehlich 3 extractable P,
CaCl2
extractable P, and water extractable P, which became a potential source of eutrophication in Taihu Lake. In contrast to chemical fertilizers plus pig manure, there were no significant differences in the concentrations of extractable P forms between the
NPK
+ S and
NPK
treatments. We concludes that chemical fertilizers [P 45 kg x (hm2 x a)(-1)] plus rice/wheat straw should be recommended in the paddy soil in the Taihu Lake region under the rice-wheat rotation system.
