The action of an eledoisin-hexapeptide analogue (EH) upon learning and memorising processes of 48 male Wistar laboratory rats aged between 5 and 6 months was studied and is reported in this paper. The animals suffered from neurogenic hypertension which had been experimentally induced by applying
emotional stress
. A comparison between the action of EH (Lys-Phe-Ile-Gly-Leu-MetNH2) and that of
Substance P
(Arg-Pro-Lys-Pro-Gln-Gln-Phe-Phe-Gly-Leu-MetNH2) on conditioned-reflex learning in the intact rat had been reported by the authors in one of their previous papers [7]. The following results were obtained with regard to EH and its action upon rats with neurogenic hypertension. The learning process was favoured, as it had been by 2 or 3 weeks of exercise. Defective learning and memorizing process as well as impaired behavioural patterns, interpreted as neurotic phenomena, were normalized by doses of 250 microgram/kg and 500 microgram/kg. Blood pressures were reduced, depending on dosage. The action of the EH analogue used on the central nervous system was stronger than that on blood pressure. Discontinuance of peptide application was followed by the phenomenon of "state-dependent learning". The results are likely to suggest possible involvement of such peptide sequences in the regulation of processes which are relevant to the whole. That effect is of particular interest, as
Substance P
is under discussion as a transmitter or modulator in mammals.

The influence of delta sleep-inducing peptide (DSIP, 60 and 120 nmole/kg, intraperitoneally) on the content of
substance P
(SP) in the hypothalamus of rats was studied in male rats of the August line. It was demonstrated that the administration of DSIP significantly increases the average content of SP in the hypothalamus, as well as its content in animals resistant to and predisposed to
emotional stress
. A daily one-time administration of DSIP before placing the rats in conditions of stress increases the content of SP in the hypothalamus which was decreased during
emotional stress
. The preliminary one-time administration of DSIP to animals subjected to a stressor influence also increases the SP content in the hypothalamus. It was established that a one-time administration of DSIP in a dose of 60 nmole/kg sharply decreases the classical manifestations of stress such as the hypertrophy of the adrenals and involution of the thymus.

Leukocyte trafficking in normal and diseased skin appears to be initially governed by endothelial surface glycoproteins that promote adhesive interactions with circulating leukocytes. In a separate study, we have demonstrated that one of these glycoproteins, endothelial-leukocyte adhesion molecule-1 (ELAM-1), is rapidly induced on postcapillary dermal venules as a direct consequence of experimentally-elicited degranulation of adjacent mast cells (Proc Natl Acad Sci USA 86:8972-8976, 1989). A principle endogenous mediator of mast cell degranulation is the neuropeptide
substance P
. In this study, we exposed organ cultures of neonatal human foreskins for 45 min to
substance P
or to a
substance P
analogue (D-pro4, D-trp7,9)SP(4-11) that binds to the identical mast cell surface receptor but which does not provoke histamine release. Dermal mast cells were uniformly degranulated only in explants exposed to
substance P
, as judged by ultrastructural analysis. After subsequent culture in medium alone for 6 h, superficial venules of explants exposed to
substance P
showed evidence of ELAM-1 induction, as documented histochemically using H4/18 monoclonal antibody. ELAM-1 was not induced by
substance P
analogue. Furthermore, preincubation of explants with analogue or with the mast cell inhibitor, cromolyn sodium, abrogated the ability of
substance P
to induce ELAM-1. From these results we suggest that
substance P
endogenously released by dermal nerve fibers upon physiologic or electrical stimulation may be important in the regulation of endothelial-leukocyte interactions in vivo. This concept provides further evidence for a neurogenic and psychogenic modulation of the immune response, and may be relevant to the course of naturally occurring dermatoses (e.g., psoriasis) that are commonly exacerbated by
emotional stress
.

The tendencies in the
substance P
influence on the expression of individual and group behaviour of adult males of rhesus macaques have been studied on the background of cardiopathogenic
emotional stress
(CES) and without it. CES stimulated the general activity of rhesus macaques in the individual cages, while the
substance P
injection without CES increased the frequency and duration of pathologic behavioral patterns. The maximal influence of
substance P
was expressed on the 3-4 day of experiments. On the 5th day the intensification of locomotion and social activity was found during the settling of all individuals in the large cage. The definite regularity of connection between the value of arterial pressure before and after experiments and the individual's social ranks is found. The injection of
substance P
on the CES background and without it the second-ranking individuals stood more hard, the leader and the most subordinated individuals stood easier.

The influence of the delta-sleep inducing peptide (DSIP, 60 and 120 nmol/kg, intraperitoneally) on the content of
substance P
(SP) in rats hypothalamus was studied on males of August line. DSIP administration significantly increased the mean SP content in the hypothalamus and also its content in animals, stable and predisposed to
emotional stress
. Daily DSIP administration before putting the rats in conditions of stress increased the SP content in the hypothalamus decreased at the
emotional stress
. Preliminary single DSIP administration to the animals subjected to stress also increased the SP content. Single DSIP administration in a dose of 60 nmol/kg sharply reduced classical stress manifestations, such as hypertrophy of adrenals and thymus involution.

Single intraperitoneal injection of
substance P
in a dose of 125 mcg/kg induced a significant increase of noradrenaline and dopamine level in the hypothalamus and the midbrain of intact rats. Under conditions of immobilization
emotional stress
, the
substance P
eliminated the stress induced decrease of hypothalamic noradrenaline and increase of its level in the midbrain; in other words the
substance P
normalized the noradrenaline level. Modulatory effect of a single injection of the
substance P
had a long-term character and was synchronized with an earlier found increase of resistability of rats to chronic
emotional stress
.

A study was made of the content of
substance P
in the blood and hypothalamus of Wistar rat brain in acute and chronic
emotional stress
and after intraperitoneal injection of
substance P
in a dose of 250 mg/kg. A possibility was demonstrated of inducing long-term changes in the content of
substance P
in the hypothalamus after a single injection. Exposure to a single 24-hour stress was followed by an increase in the
substance P
content in the hypothalamus.

The effect of
substance P
--an endogenous brain peptide--on the resistance to chronic
emotional stress
was studied in rats. A single administration of the peptide (125 or 250 mkg/kg) elicits a highly significant increase of rats resistance to chronic
emotional stress
and reduces the raise of adrenals weight, hemodynamic fluctuations and emotional reactivity.

The immune system is important in the pathogenesis of psoriasis and
emotional stress
has precipitated psoriasis in many patients. Neuropeptides, alpha-Melanocyte stimulating hormone (alpha-MSH), beta-endorphin, met-enkephalin and
substance P
(SP) act as immunomodulators, and their secretion increases during periods of stress. To see whether these neuropeptides themselves might be related to psoriasis and/or to the aggressiveness of the disease, we evaluated the plasma neuropeptide levels in 13 patients with active psoriasis (patients with new lesions and/or pre-existing lesions that had become larger during the month before the study), in 11 patients with stable psoriasis and in 10 healthy controls. Plasma concentrations of neuropeptides were evaluated by RIA (immunoradiometric assay for beta-endorphin). Data were compared by the Student t-test for unpaired data. There were no significant differences between the plasma levels of any of the neuropeptides between active psoriatic patients and stable psoriatic patients, nor between the plasma levels of neuropeptides of psoriatic patients and those of control subjects. It seems unlikely that circulating neuropeptide levels are of primary importance in the manifestation of the psoriatic skin lesions.

The paper provides evidence for the new hypothesis of the nature of congestive excitation in the brain region during
emotional stress
. In the author's opinion,
emotional stress
-induced congestive excitation is determined by disintegration of molecular chemical properties in the brain regions, by the altered sensitivity of brain neurons to neurotransmitters and neuropeptides. The baseline molecular integration of the brain regions, its change and fixation in a steady status have been shown to be governed by special neuropeptides. Additional administration of
substance P
, delta-sleep-inducing peptide, beta-endorphin to stress-predisposed animals has been ascertained to normalize the chemical properties of brain neurons and to enhance
emotional stress
resistance in the animals.
