The distribution of substance P- (SP-) and calcitonin gene-related peptide-like immunoreactivities (CGRP-LI) was studied in the human carotid body at the light and electron microscopic level. Of the different compartments of the carotid body, the glomic lobules received the densest innervation by SP/CGRP double-labelled axons, followed by interlobular arteries and interlobular connective tissue. Ultrastructurally, preterminal SP/CGRP-LI axons are unmyelinated and measure 0.09-1.02 micron in diameter with a median diameter of 0.28 micron (calculated conduction velocity: 0.5 m/s). Within the glomic lobules, vesicle-containing varicosities are numerous around glomus cells, but only few of them directly contact (non-synaptically) glomus cells. The findings are consistent with the existence of two populations of SP/CGRP-LI C-fibers, one of them probably representing chemoreceptor C-fibers, the other belonging to the ubiquitous vascular sensory innervation. In addition to its localization in nerve fibers, CGRP-LI also occurs in dense core vesicles of a subpopulation of glomus cells. In this regard, the human carotid body is unique among mammalian species studied so far, which severely hampers the interpretation of pharmacological data obtained in laboratory animals in terms of human physiology.

Mustard oil, which stimulates small diameter afferents, was used to evoke the expression of the oncogene c-fos in the lumbar spinal cord. C-fos-like immunoreactivity was concentrated in, but not limited to, neuronal nuclei of laminae I and II of the lumbar dorsal horn. Double-label immunocytochemistry was used to determine if neurons which expressed c-fos-like immunoreactivity received axonal input from enkephalin-, substance P- or serotonin-immunoreactive neurons. The analysis of vibratome and semithin plastic-embedded tissue sections demonstrated that the majority of c-fos-like immunoreactive neurons received input from enkephalin-, substance P- or serotonin-immunoreactive axonal varicosities.

Impingement of plical synovial tissue in a facet joint could cause pain. Plical tissue was removed during surgery for recurrent disc herniation or spinal stenosis. The presence of nerves was studied with silver impregnation, immunofluorescence, and avidin-biotin-peroxidase complex (ABC) immunostaining. Heterologous antisera to protein gene product (PGP) 9.5, substance P, calcitonin gene-related peptide (CGRP), and galanin were used to stain nerves. After silver impregnation, nerve-like structures were observed perivascularly. Such nerves located close to blood vessels were also immunoreactive for PGP 9.5, a more general cytoplasmic neural marker, whereas only few perivascular small varicosities were seen with antisera to substance P and galanin and none with antiserum to CGRP. In addition, PGP-9.5-, substance-P-, and galanin-immunoreactive nerves were occasionally seen very near to fat globules. Very few peptide-immunoreactive nerve varicosities were seen with immunofluorescence, and none of the PGP-9.5-immunoreactive nerves that were observed with ABC immunostaining were immunoreactive for neuropeptides as well. One mechanism for pain production could be mechanical compression of fatty tissue, but it is considered more likely that nerves in this particular tissue are mainly involved in local vasoregulation and that they are not sensory nociceptive nerves.

The peptidergic innervation of guinea-pig coronary arteries was investigated by means of immunohistochemical, ultrastructural and in vitro pharmacological techniques. A network of nerves was demonstrated in all major epicardial arteries by means of an antiserum to the neuronal marker protein gene product 9.5. The majority of nerve fibres possessed neuropeptide Y (NPY) and tyrosine hydroxylase (TH) immunoreactivity, the number and distribution of nerves immunoreactive for NPY being similar to that of nerves containing TH immunoreactivity. Numerous nerve fibres displaying immunoreactivity for substance P, neuropeptide K and calcitonin gene-related peptide (CGRP) were also found. In double-stained preparations substance P immunoreactivity was co-localized with CGRP and with neuropeptide K immunoreactivities in the same varicose nerve fibres. Ultrastructural studies revealed the presence of numerous axon varicosities at the adventitial-medial border. NPY immunoreactivity was localized in large granular vesicles in nerve varicosities which also contained numerous small granular vesicles. Large granular vesicle-containing nerves also displayed immunoreactivity for dopamine-beta-hydroxylase. With an in vitro method, the vasomotor responses to perivascular peptides were characterized in epicardial and intramyocardial arteries. In epicardial arteries neither noradrenaline nor NPY elicited a contractile response. Only in some intramyocardial arteries was an NPY-mediated contraction demonstrated. No potentiating effect of noradrenaline and NPY was observed in either epicardial or intramyocardial arterial segments. In contrast, CGRP, substance P and vasoactive intestinal peptide (VIP) all produced a concentration-dependent relaxation of both epicardial and intramyocardial arteries. These results suggest that peptide-containing nerves associated with guinea-pig coronary arteries may predominantly be involved in mediating vasodilation.

The peptidergic innervation of human dental pulp was studied with indirect immunofluorescence and immunoperoxidase techniques. Pulpal nerve fibres displaying immunoreactivity for cholecystokinin, calcitonin gene-related peptide, C-terminal flanking peptide of neuropeptide tyrosine, leucine-enkephalin, methionine-enkephalin, neuropeptide K, neuropeptide tyrosine, peptide with N-terminal histidine and C-terminal isoleucine, somatostatin-28, substance P and vasoactive intestinal polypeptide were observed. Immunoreactive axon varicosities were detectable within radicular and coronal nerve trunks and within the nerve plexus of Raschkow in the para-odontoblastic region. Many peptidergic nerve fibres were observed in association with blood vessels of various sizes. Substance P- and calcitonin-gene-related peptide-immunoreactive axons were visible in the odontoblastic layer. The occurrence of VIP- and PHI-immunoreactive fibres lends support to the hypothesis that human tooth may be supplied by parasympathetic nerves. The immunocytochemical results here shown provide a morphological basis to previous experimental studies concerning the possible roles of neuropeptides in nociception mechanisms, control of the blood flow and modulation of the inflammatory response in dental tissues.

The immunocytochemical localization of corticotropin-releasing factor (CRF), thyrotropin-releasing hormone (TRH), substance P (SP), neuropeptide Y (NPY), and neurotensin (NT) were studied in the human locus coeruleus (LC) using the avidin-biotin complex (ABC) technique. The brains were obtained from 3 adult male human subjects of 36-61 years old. All of these peptides studied were present in nerve fibers and varicosities throughout the entire nucleus, where they distributed unevenly. The highest density of immunoreactive neuronal networks was generally observed in the middle and to a lesser extent in the caudal part of the nucleus, while only scattered fibers were seen in the rostral part of the LC. Among the investigated neuronal peptides, the CRF-immunopositive network was the most dense, less dense immunostaining with TRH and NPY antisera while only few SP- and NT-immunoreactive fibers were present in the nucleus. Cell bodies immunostained for the above peptides were not seen in the LC. Per analogiam with rats, fibers immunostained in the LC may be of both intrinsic and extrinsic origins.

By the use of the indirect immunofluorescence and in situ hybridization techniques, the distribution of calcitonin gene-related peptide (CGRP)-like immunoreactivity (LI) and CGRP mRNA was studied in the spinal cord as well as in the midline raphe nuclei and the hypoglossal nucleus in the medulla oblongata of the monkey (Macaca fascicularis). In the spinal cord only a few large neurons in the motor nucleus contained CGRP-LI, while a majority of the neurons in the hypoglossal nucleus contained CGRP-LI. A relatively dense innervation by CGRP-immunoreactive (IR) fibers was also seen close to cell bodies and proximal dendrites of large neurons in the motor nucleus, especially in its ventral part. 5-Hydroxytryptamine (5-HT)-, substance P- and thyrotropin-releasing hormone (TRH)-IR varicosities were also observed in a similar position around large neurons in the motor nucleus. Double labeling disclosed that the majority of CGRP-IR axon terminals also contained 5-HT-LI. Expression of CGRP mRNA was found in neurons in the medullary midline raphe nuclei and in large neurons in the motor nucleus at the cervical spinal cord level. In adjacent sections of the medulla oblongata, CGRP-labeled neurons in the midline raphe nuclei also expressed preprotachykinin mRNA. The present results show that CGRP- and 5-HT-LI coexist in fibers within the motor nucleus of the monkey spinal cord and that this coexistence is probably due to the presence of CGRP in the descending bulbospinal, serotonergic pathway.

Partially purified nerve varicosities prepared from canine small intestinal myenteric, deep muscular and submucosal plexuses were found to contain, by radioimmunoassay, gastrin-releasing polypeptide (GRP), substance P, Leu-enkephalin, Met-enkephalin, vasoactive intestinal polypeptide (VIP) and neurokinin A, but did not contain detectable amounts of neurokinin B. In all three plexus preparations, VIP was present in the highest concentration. In contrast to other species, GRP and the enkephalins were found to be present in relatively high concentrations in the submucosal plexus and GRP was present in low concentrations in the deep muscular plexus. Equal concentrations of substance P and neurokinin A were found in the myenteric and deep muscular plexus preparations but greater concentrations of substance P relative to neurokinin A were found in the submucosal plexus preparations. On reverse phase HPLC, a major peak of immunoreactivity occurred at the retention times of standard preparations for all six neuropeptides measured. Significant heterogeneity was found for GRP- and VIP-like immunoreactivity, especially in the submucosal plexus preparations. These partially purified canine small intestine nerve varicosity preparations may prove of value in studying release mechanisms for, and the posttranslational processing of, neuropeptides.

An analysis of Nissl stained sections of the spinal cord taken from four species of elasmobranch showed that seven distinct cytoarchitectonic laminae are present. These laminae are compared with laminae described previously in the spinal cord of other vertebrates. The distribution of immunoreactivity to serotonin, substance P, somatostatin, calcitonin gene-related peptide, neuropeptide Y, and bombesin was determined in the brown stringray (Dasyatis fluviorum), the eagle ray (Aetobatis narinari), the shovelnose ray (Rhinobatis battilum), and the black-tip shark (Carcharhinus melanopterus). In all species, dense immunoreactivity to most substances tested was found in the outer part of the substantia gelatinosa. Many fibres and varicosities immunoreactive to substance P, calcitonin gene-related peptide, and bombesin were found in this region and smaller numbers of fibres were found in the nucleus proprius. Immunoreactivity to somatostatin consisted of coarse fibre bundles that entered the dorsal horn at the nucleus proprius and radiated dorsally to the substantia gelatinosa. Axons and varicosities immunoreactive to serotonin and neuropeptide Y were found in all regions of the dorsal horn but were concentrated in the outer part of the substantia gelatinosa. The distribution of immunoreactivity to met-enkephalin in the shovelnose ray was concentrated in the lateral third of the substantia gelatinosa and to a lesser extent in the nucleus proprius. The distribution of these substances is compared with that described in other vertebrates. Although the sensory information reaching the elasmobranch spinal cord is limited, compared with that of mammalian species, the distribution of these neuroactive factors in the dorsal horn of the two groups is strikingly similar.

The effects of capsaicin on the function and structure of muscularis mucosa of opossum esophagus were studied. In this tissue there are numerous nerves containing a substance P-like immunoreactive substance (SPLS), and electrical field stimulation (EFS) leads to a phasic response that appears to be due to release of acetylcholine, followed by a tonic response at higher frequencies of stimulation that appears to be due to release of a SPLS. The acetylcholine released by EFS and exogenous muscarinic agonists inhibits release of this SPLS (8). In the present study capsaicin (5 X 10(-5) M) was shown to cause a tonic submaximal contraction in most cases. This was prevented by substance P tachyphylaxis and by pretreatment with the partial agonist [D-Pro2, D-Trp7,9]substance P, the antagonist [D-Arg1, D-Trp7, Leu11]substance P, and tetrodotoxin. This response to capsaicin could not be repeated even after 2 h. Capsaicin also abolished the tonic response to high-frequency EFS without affecting phasic responses and reduced markedly the enhanced tonic response after atropine had abolished the phasic response. This occurred with or without a preliminary contraction to capsaicin. This tonic response to high-frequency EFS recovered completely 1-2 h after washing out capsaicin. Then, a further administration of capsaicin had no direct effect but again abolished tonic responses to EFS. Capsaicin reduced responses to exogenous substance P or carbachol only partially. When tissues were fixed and studied after capsaicin had abolished tonic responses to EFS, specific damage to nerve varicosities or synaptic vesicles in nerve varicosities could not be demonstrated.(ABSTRACT TRUNCATED AT 250 WORDS)
