Cells of sarcoma 180 and of Ehrlich's carcinoma were maintained by serial transplantation in male and female Swiss mice. Either estrogen, progesterone, or testosterone were injected im at doses of 1 mg/mouse. Ascitic fluid was aspirated at intervals of 1, 3, 6, 24, and 48 hours following hormone injections. Enzyme activities were analyzed by subjective grading according to the intensity of staining reaction. Estrogen produced enhancement of alkaline phosphatase activity in both types of cells in both sexes of mice. Progesterone produced increased alkaline phosphatase activity in both types of cells from female hosts but an inhibitory effect in male hosts' cells. Testosterone produced no change in enzyme activity in tumor cells of female hosts but in male hosts it inhibited enzyme activity of sarcoma 180 cells and activated activity in carcinoma cells. The effect of all 3 hormones on acid phosphatase activity was activation. With adenosine triphosphatase, estrogen stimulated the activity in both types of tumor in both sexes. Progesterone stimulated cells from male hosts with little or no effect on cells from female hosts. This enzyme was resistant to testosterone. Succinate dehydrogenase activity under similar conditions was different. Estrogen reduced this activity and progesterone produced some inhibition of activity. Testosterone inhibited the sarcoma cells but had no effect on carcinoma cells of either sex. Others have shown that sex hormones affect the enzyme activities beyond the target tissues, particularly in the liver, kidney, and pancreas. Different responses of the enzymes seemed to depend on the endogenous hormonal status of the mice.

A 54-year-old man was admitted because of right supraclavicular lymphadenopathy of some weeks duration. Computed axial tomography revealed a large multinodular lesion in a supraclavicular lymph node. The patient then had a supraclavicular lymph node biopsy. Light microscopy showed a tumor whose structure was suggestive of an interdigitating cell sarcoma. Enzyme and immunohistochemical analysis showed that the tumor cells possessed membranous adenosine triphosphatase activity, intracytoplasmic S100 protein, surface CD1a and CD4 antigens, and HLA-DR antigen. Ultrastructural examination showed that the cells exhibited many interdigitating cytoplasmic extensions, but no Birbeck granules. DNA content analysis of the tumor cells proved that the cells were malignant. These data are consistent with derivation from a lymph node interdigitating cell.

For the purpose of clarifying cellular differentiation of epithelioid sarcoma, studies based on various methods were performed. Enzyme histochemical studies showed that epithelioid sarcoma tumor cells have characteristics intermediate between epithelial cells and the large plump cells of synovial sarcoma-incomplete epithelial differentiation. For alkaline phosphatase and adenosine triphosphatase particularly, positive cells and negative cells coexisted, as in the large plump cells of synovial sarcoma. Immunohistochemical studies for alpha 1-antitrypsin, alpha 1-antichymotrypsin, vimentin, and keratin also showed that epithelioid sarcoma tumor cells are very similar to the large plump cells of synovial sarcoma and have incomplete epithelial differentiation. For example, the examinations of serial sections and double staining methods revealed that keratin-positive cells are always vimentin-positive in epithelioid sarcoma and in the monophasic area of synovial sarcoma. Electron-microscopically, bundles of intermediate filaments and filopodia toward the intercellular lumen were observed, as in the monophasic area of synovial sarcoma. The results of enzyme-histochemical and immunohistochemical studies of non-neoplastic synovial lining cells, performed here for the first time, are also discussed.

A case of alveolar soft part sarcoma was studied by light and electron microscopy and by electron microscopic enzyme histochemistry of adenosine triphosphatase (ATPase) and 5'nucleotidase(5'Nase). The tumor showed distinct alveolar pattern and diastase resistant PAS positive crystalline inclusions were found in the cytoplasm. Ultrastructurally, characteristic rhomboid crystals and dense granules were observed and they were positive for Mg++- and Ca++-ATPases but negative for 5'Nase. Tumor cell membrane also showed positive activity of ATPase in addition to 5'Nase. These results would support the myogenous derivation of alveolar soft part sarcoma.

The increased rate of glucose uptake found in cells transformed by Rous sarcoma virus was shown to be enhanced relative to the changes in uptake induced in nontransformed cells by deprivation of glucose (deprivation derepression). Glucose-specific uptake sites were distinguished from glucose-galactose sites in nontransformed cells, and the capacities for glucose uptake and for galactose uptake were increased to about the same extent by the exclusion of glucose from the cell culture medium. Deprivation derepression occurred without a requirement for new RNA or protein synthesis, suggesting that preexisting inactivate uptake sites were activated. Deprivation derepression could be mimicked by the treatment of cells with adenosine triphosphatase activators, and adenosine triphosphate levels were reduced in glucose-deprived cells and in cells treated with adenosine triphosphatase activators. Cells transformed by the Bryan strain of Rous sarcoma virus were unresponsive to addition of high concentrations of glucose, to glucose starvation, or to treatment with adenosine triphosphatase activators, and the relative capacity for glucose uptake in these transformed cells was enhanced much more than the capacity of galactose uptake. It was concluded that cells infected by the Bryan strain of rous sarcoma virus in the process of transformation selectively synthesize more sites specific for glucose uptake. Lower levels of adenosine triphosphate found in transformed cells possibly contribute to a chronic derepression of uptake sites.

A tumor in a 37 years old male is described in which the tumor cells appeared to be derived from interdigitating cells normally found in the T-cell area of lymph nodes. The patient presented with superior vena caval obstruction due to a mediastinal mass, followed by lymph node enlargement and skin lesions leading to death within 4 months. The tumor cells lacked immune markers for lymphocytic cells. They showed Ia-like antigens and high adenosine triphosphatase activity, while acid phosphatase and alpha-naphthyl acetate esterase activity was absent. Their fine morphology was strikingly similar to that of interdigitating cells. A combination of these data led us to the conclusion that this tumor represents a specific subtype of the tumors derived from the mononuclear phagocyte system, namely a sarcoma of interdigitating cells.

A case of epithelioid sarcoma was studied by electron microscopy and by light and electron microscopic enzyme histochemistry comparing with several control soft tissues. In addition to previously reported ultrastructural features, such as abundant 10 nm cytoplasmic filaments, desmosome-like cell junctions and small cystic spaces surrounded by filopodia or microvilli of the tumor cells, we encountered 10 nm cytoplasmic filaments showing electron dense condensation with a concentrically oriented or whorled pattern and a finger-print-like arrangement and 5'-nucleotidase activity of tumor cell membrane. Among the control soft tissues, 5'-nucleotidase activity was found only in synovial and endothelial cells. Both tumor and synovial cells showed no activity of adenosine triphosphatase, while marked activity of the enzyme was found in endothelial cells. These results support the concept that epithelioid sarcoma is derived from mesenchymal cells undergoing differentiation toward synovial cells during neoplastic transformation.
