Chemical parameters comprising urea and creatinine nitrogen, cations (Na+, K+, and Ca2+), chloride, phosphorus, protein, cholesterol and enzymes, aminotransferases, alkaline and prostatic acid phosphatases, gamma-glutamyltransferase, creatine kinase, and lactate dehydrogenase were ascertained for semen from groups A (vasectomized), B (oligospermic), and C (normospermic) men, 19 to 55 years of age. Of the parameters, the vasectomized group underwent definite depressions in potassium ion, alkaline phosphatase, aspartate aminotransferase, gamma-glutamyltransferase, and lactate dehydrogenase as compared with the normospermic group; the last three enzymes and, possibly, the urea-creatinine ratio were decreased for the oligospermic group vs. the normospermic men. In the comparison of groups A and B, only the decrements in alanine aminotransferase and lactate dehydrogenase were statistically significant. In corroboration of past reports, CK-BB comprised the main isoenzyme of semen creatine kinase.

The objectives of the study were to establish the effects of five successive intra-abdominal injections of sterigmatocystin (STG), administered at 11, 13, 15, 17, and 19 days of age, on the growth pattern of chicks and their organs, and on the concentration of certain blood and liver constituents. The STG, when administered at dosages of .5 and .7 mg per injection, markedly reduced chick growth and affected organ weights. In general, there was an increase in the relative size of the crop, proventriculus, gizzard, large intestine, kidney and pancreas and a decrease in relative size of the bursa of Fabricius. Liver, heart, and spleen size was not affected. Peritonitis was observed in chicks given the high dosage STG. The STG elevated the activity of serum aspartate aminotransferase and the number of circulating granulocytes and depressed concentrations of total serum proteins, albumin, potassium, and the total number of circulating white blood cells and granulocytes. The STG treatment decreased the concentration of dry matter, DNA, RNA, and protein in the liver, affected glycogen concentration differentially, and had no effect on lipid concentration. Liver and kidneys also showed degenerative changes as detected histopathologically. The results of these studies suggest that STG affects several tissues including the digestive system, liver, kidney, pancreas, and the immunological system.

The pharmacokinetics and safety of amphotericin B infusion were studied in 13 infants and children (age range 3 weeks to 18 years; median age 11 years) treated with the drug for proved (n = 11) or suspected (n = 2) fungal infections. The dose during the first day was 0.5 mg/kg, followed by a daily dose of 1 mg/kg for the rest of the treatment period in most patients. The drug was infused over 4 to 6 hours. During the first day, serum concentrations were above the target therapeutic level of 0.3 microgram/ml in all patients at 2 and 6 hours from the start of the infusion, in 12 of 13 patients at 12 hours, but in only 6 of 13 patients at 24 hours. On the third day, all concentrations were greater than 0.3 microgram/ml throughout the 24-hour period, and in 12 of 13 patients were greater than 0.5 microgram/ml. The same kinetic profile prevailed on days 7 to 10 of therapy, with a tendency for increasing concentrations. Elimination half-life was 9.93 +/- 1.5 hours (mean +/- SEM), clearance rate 26 +/- 5 ml/kg.hr, and distribution volume 378 +/- 25 ml/kg. The half-life inversely correlated with patient's age. Pharmacokinetic values calculated during the first day were not different from those calculated on day 3. Significant decreases in hemoglobin, platelets, and serum potassium concentration were recorded along with significant increases in serum creatinine, urea, and aspartate transaminase values. Because of the large pharmacokinetic variability and the high rate of serious adverse effects, individualized dosing of amphotericin B based on therapeutic drug monitoring should be considered.

In the fall of 1983, 322 western calves, in five different groups, were bled on arrival at two Ontario feedlots. Calves receiving treatment for respiratory disease, within 35 days of arrival, were denoted as cases. The hematology and serum chemistry parameters of cases were compared to those of controls. Cases had significantly (p less than 0.05) lower hematocrits, fewer platelets, and more band cells on arrival, than did controls. Cases also had lower serum phosphorous, magnesium, potassium, iron and alkaline phosphatase levels, and increased bilirubin and aspartate aminotransferase levels relative to controls. Based on the results of multivariable logistic regression, hematocrit, platelet numbers, serum phosphorous, iron and aspartate aminotransferase levels were the most significant parameters for the prediction of respiratory disease. Reference values for stressed feedlot calves were created. Most parameters were distributed in an approximately normal manner, however the group to group variation in most parameters was significant.

Concentrations of sodium, potassium, chloride, inorganic phosphorus, total magnesium, total calcium, iron, urea, creatinine, total protein, albumin, total bilirubin, alkaline phosphatase (ALP), alanine transaminase (ALT), lactate dehydrogenase (LD), creatine kinase (CK), gamma-glutamyltransferase (GGT) and aspartate transaminase (AST) were determined in serum specimens collected from 53 free-ranging mountain reedbuck (Redunca fulvorufula) during live capture using nets. Considerable variations in the concentrations of the enzymes ALP, LDH, CK, GGT and AST were found as well as in the concentrations of creatinine, bilirubin and iron. This wide variation in results seriously questions the usefulness of similar blood investigations on heterogenous groups of mechanically restrained animals.

Concentrations of sodium, potassium, chloride, inorganic phosphorus, total calcium, total magnesium, albumin, total protein, cholesterol, urea, creatinine, cortisol as well as the activities of alkaline phosphatase, alanine transaminase, aspartate transaminase, gamma-glutamyltransferase, creatine kinase and lactate dehydrogenase were determined in serum specimens collected from 100 free-ranging warthogs Phacochoerus aethiopicus within five minutes after they were killed with a shotgun. Average concentrations for the following chemical constituents were found: sodium (145 mmol l-1), potassium (8.6 mmol l-1), chloride (102.5 mmol l-1), phosphorus (2.31 mmol l-1), calcium (2.93 mmol l-1), magnesium (1.23 mmol l-1), albumin (26.4 g l-1), serum proteins (62.2 g l-1), cholesterol (1.82 mmol l-1) and urea (8.74 mmol l-1). The cortisol concentrations ranged from 55-340 nmol l-1 (n = 30). Wide variations were recorded in the concentration of creatinine as well as in the activities of the various enzymes.

Four experiments were carried out with 10 to 12 day old leghorn chicks weighing approximately 93 to 101 g. The chicks were injected intraperitoneally with sterigmatocystin (STG) dissolved in olive oil. The LD50 values as established in the first two experiments were 10.0 and 14.0 mg/kg body weight with most of the deaths occurring between 9 and 21 h following injection. Histopathological studies demonstrated that there was hemorrhage, foci of degeneration and necrosis with fibroblastic proliferation in sinusoids of the liver while the kidneys showed tubular degeneration and necrosis. Biochemical analysis of blood sera demonstrated that STG caused a marked elevation in the activities of lactate dehydrogenase, aspartate aminotransferase, and alanine aminotransferase, and a depression of creatine kinase, but no effects on gamma-glutamyl transferase, amylase and lipase. Free and conjugated bilirubin were elevated in the sera while total protein, albumin, glucose, potassium, chloride and phosphorous concentrations were depressed. In addition, total white blood cells and circulating agranulocytes were depressed while circulating granulocytes were elevated. STG did not significantly affect the concentration of uric acid, cholesterol, triglycerides, calcium, magnesium and sodium in blood.

A 7-year-old adult male ferret had progressive hair loss that was bilaterally symmetric. Also clinically evident were severe dehydration, polydipsia, muffled heart sounds, weak femoral pulses, hepatomegaly, lethargy, weakness, temporal muscular atrophy, dyspnea, and weakness. The blood profile of the ferret indicated profound leukopenia, eosinopenia, and high phosphorus, BUN, creatinine, and potassium concentrations, as well as high aspartate transaminase activity; the albumin concentration was low. The serum cortisol concentration was 8.1 micrograms/dl. Necropsy and histologic findings confirmed a diagnosis of hyperadrenocorticism, complicated by dilatative cardiomyopathy, chronic active hepatitis, and renal disease.

The suitability of isolated rat hepatocytes for the investigation of the toxicity of chemical mixtures has been studied. Cadmium chloride and chloroform were used because both had been previously investigated in hepatocytes and both produce hepatotoxicity after in vivo administration. The addition of the two chemicals caused an increase in cytotoxicity as assessed by loss of intracellular potassium ion and aspartate aminotransferase. There was even a toxic response form the mixture at concentrations where the chemicals alone yielded no such response. The metabolic parameter, lactate to pyruvate ratio, was less consistently affected. The nature of the interaction could, under the varying conditions employed, be described as one of synergism, potentiation, or addition. The data support the proposed role of isolated hepatocytes in screening chemical mixtures for toxic effects.
