31 healthy Thai males, 22 Thai male regular drinkers not suffering from any clinical signs or symptoms of alcoholism, and 52 patients from a neurological hospital in Bangkok suffering from the effects of chronic alcohol consumption were investigated. Alcohol consumption in asymptomatic drinkers ranged from 7 to 134 (median 44) g/d ethanol, and for the patients 22 to 517 (median 197) g/d ethanol, as assessed by questionnaires. The symptomatic alcohol drinkers had consumed alcohol for 2 to 35 years and the hospitalized patients for 5 to 40 years. Only the median levels of serum triglycerides and serum glutamyl transferase (gamma-G) were significantly increased and vitamin B1 deficiency was found with higher frequency in the group of alcohol drinkers without clinical signs compared with the healthy non-alcohol drinkers. Statistically significant correlations were demonstrated in the group of asymptomatic alcohol drinkers only, between alcohol consumption and the Quetelet's index, gamma-G, and alkaline phosphatase levels. Alkaline phosphatase also correlated significantly with gamma-G. In the group of hospitalized patients, compared with healthy males statistically significantly higher median values of systolic and diastolic blood pressure, serum triglyceride, gamma-G, aspartate aminotransferase (GOT), alanine aminotransferase (GPT), alkaline phosphatase, haemoglobin, hematocrit, folate and total protein were found. The median levels of cholesterol, bilirubin, vitamin B2, B6 and B12 in the hospitalized group were lower than, but not significantly different from the other two groups.

The pyridoxamine (PM), pyridoxal (PL) and pyridoxol (PN) in liver, the urinary excretion of 4-pyridoxic acid (4-PA), the red blood cells enzymatic activities (aspartate aminotransferase, GOT and alanine aminotransferase, GPT) were determined in rats fed diets containing different levels of PN, HCl (0,23-170 micrograms/rat/d) during 13 days. PL and PM hepatic levels, GOT and GPT activities decreased significantly in groups A and B receiving respectively 0,23 and 11 micrograms PN, HCl/rat/d. No differences were noticed in food intake and growth. The urinary excretion of 4-PA shows a consistent trend with vitamin B6 intake. As the PM/PL-ratio was maximum in group A, PL reflects the actually available vitamer, whereas PM should be regarded as "trapped" PL. The GOT and GPT activities can be considered as reliable parameters of the pyridoxine status of the rat.

Studies on the influence of long-lasting hyperthyroidism on enzyme activities and total protein level in the blood plasma of adult Leghorn hens showed that: 1. Protein level during whole experimental period showed inconsiderable variability irrespective of T4 dose. 2. Activity of aspartate aminotransferase (GOT) and alanine aminotransferase (GPT) increased. Changes were dependent on T4 dosage level. 3. T4 had no effect on activity of aldolase.

The influence of intestinal microflora on the hepatotoxic effects of dimethylnitrosamine (DMN) or dimethylamine (DMA) plus NaNO2 was studied by comparing the degree of liver necrosis and the levels of serum alanine aminotransferase (GPT) and aspartate aminotransferase (GOT) in germ-free and conventional male Wistar rats (320 to 340 g). In one experiment, both germ-free and conventional rats were intubated with DMN in respective doses of 8, 9, and 10 mg/kg of body weight, while in another experiment, both groups were intubated with DMA (1500 mg/kg) plus NaNO2 (100 mg/kg). In both experiments, 48 hr after intubation, there was a marked difference in the degree of liver necrosis and the levels of serum GPT and GOT between the groups. In particular, a dose of 8 mg of DMN or 1500 mg of DMA plus 100 mg of NaNO2 produced severe liver necrosis in the majority of germ-free rats, while the same dose did not produce any detectable liver necrosis in the majority of conventional rats. At a dose of 8 mg, serum GPT and GOT levels were raised to 22 and 15 times normal values, respectively, in germ-free rats, but only to about twice the normal values for both levels in conventional rats. At the combination dose of DMA plus NaNO2, the levels of serum GPT and GOT were raised to 40 and 30 times normal values, respectively, in germ-free rats, while both levels remained almost normal in conventional rats. Thus, the results indicated that the liver of the germ-free state was far more susceptible to the acute toxic effects of DMN as well as DMA plus NaNO2 administration at a certain dose range than was the liver of the conventional state, suggesting the influence of the absence of microflora.

Quails hatched from eggs incubated at physiological temperature (37.5 degrees C--normal quails) and elevated (39.3 degrees C--warm quails) were injected with L-thyroxine (T4) at the dose of 600 micrograms/kg of body weight, every 48 hr for 17 days. 2. Twenty-four hours after the last injection activity of aspartate aminotransferase (GOT), alanine aminotransferase (GPT) was determined in liver homogenates and lactate dehydrogenase and isocitrate dehydrogenase in homogenates of heart and kidney. 3. Significant increase of the activity of GPT in liver homogenates was observed in normal and warm quails up to 252.9 and 186.8% of control, respectively). 4. The activity of aspartate aminotransferase increased significantly in liver homogenates of T4-treated normal quails, while such changes in the warm quails were not observed. 5. Activities of lactate dehydrogenase (LDH) and isocitrate dehydrogenase (ICDH) in heart and kidney homogenates in both T4-treated groups of birds did not change.

An elevation of serum aspartate aminotransferase (GOT) and alanine aminotransferase (GPT) may be produced in patients treated with i.v. full-dose HEPARIN. We studied the influence of low-dose s.c. HEPARIN (5,000 IU X 2) in 34 patients with acute myocardial infarction (AMI) and in 7 with cerebrovascular accidents or calf thrombophlebitis. Twelve patients (all males) with AMI showed a secondary elevation of GOT and GPT at about the sixth or seventh day after the commencement of therapy that persisted throughout the period of treatment. Four patients (two males and two females) with cerebrovascular accidents or thrombophlebitis showed similar increases of GOT and GPT.

We studied the effect of carbamoylphosphate (CP) on L-aspartate aminotransferase (GOT) and L-alanine aminotransferase (GPT), compared to its effect on L-threonine deaminase (TD). GPT and GOT were slightly inhibited by CP, while TD was strongly inhibited. GPT and TD, but not GOT, were inactivated when preincubated with CP. Only GOT was enhanced by pyridoxal 5'-phosphate (PLP), but not when the coenzyme was preincubated with CP. When the enzymes were resolved by p-chloromercuribenzoate (PCMB) treatment to apoenzymes, only GOT retained 47% of the original activity. Reconstitution of the apoenzymes with PLP also followed different course; activities of GPT and TD were completely restored while GOT remained partially inactivated. Treatment of apoenzymes with CP resulted in impairment of their reconstitution except GPT, activity of which could be completely restored. When PLP was pre-treated with CP before reconstitution, however, even GPT was only partially restored. The data indicated that CP affect activities of these enzymes at different levels, holoenzymes, PLP and probably apoenzymes. Under a concentration of PLP, activity of GOT would be most enhanced, followed by TD then GPT. In the presence of CP, this effect would be eliminated.

We examined the effects on alanine aminotransferase and aspartate aminotransferase of different aminothiols (L-cysteine, D-cysteine, cysteamine, L-cysteine ethyl ester, L-cysteine methyl ester) and several vitamin B-6 derivatives (pyridoxal, pyridoxamine, pyridoxol, pyridoxol 5'-phosphate), before and after treatment with KOCN, which transforms these molecules into the corresponding carbamoyl derivatives. Only GPT, and not GOT, was specifically inhibited by L-cysteine and, to a lesser extent, by D-cysteine. The association reaction: PLP + apo GPT<-->holo GPT was inhibited by the vitamin B-6 derivatives, and this inhibition was prevented by pretreatment of the vitamin B-6 derivatives with KOCN. All the observed effects occurred at pH 7, 37 degrees C, at mM and even lower concentrations of reagents. Hence, they all potentially play a physiological role, in the regulation of the PLP dependent enzymes and of the vitamin B-6 levels in the cell.

Cystathionine gamma-lyase activity in the sera of rats subjected to experimental hepatotoxicity after intraperitoneal administration of carbon tetrachloride (CCl4) was measured and compared with activities of aspartate aminotransferase (GOT) and alanine aminotransferase (GPT), which have been clinically used for detecting liver damage. In the experimental subjects, serum levels of cystathionine gamma-lyase showed a similar behavior to GOT and GPT, increasing markedly with respect to the controls after administration of CCl4 and reaching a maximum at 24 hours. No such cystathionine gamma-lyase activity was detected immunochemically in the control subjects. These data suggest that measurement of serum cystathionine gamma-lyase activity could be used as a sensitive and specific marker of hepatic cytolysis.

A control survey was conducted to check the accuracy of automated analyzers used in the evaluation of clinical chemistry parameters in nonclinical toxicology studies. Pooled serum samples from male Sprague-Dawley rats were delivered refrigerated to each facility 98 laboratory facilities throughout Japan within 18 hours after sample preparation and analyzed. Commercially available normal human serum samples from a single lot were also analyzed at the same time. Survey results were divided into three categories. (1) Parameters with small coefficient of variation (CV) values for both rat and human serum samples included protein, glucose, cholesterol (CHO), urea nitrogen (UN), sodium (Na), potassium (K), chloride (Cl), calcium (Ca), and inorganic phosphate (IP). Definition of normal values in rats should be straight forward for these parameters. (2) Parameters with large CV values, but with a relatively good correlation between rat and human values include triglycerides (TG), glutamic oxaloacetic transaminase/aspartate aminotransferase (GOT/AST), glutamic pyruvic transaminase/alanine aminotransferase (GPT/ALT), and alkaline phosphatase (ALP). Measurements based on different principles gave different mean values, and this values contributed to the increase in CV values. Assessment of normal values would require a consideration of the measurement principles. (3) Parameters with large CV values only in rat serum samples included albumin (albumin/globulin ratio: A/G ratio), creatinine (CRE), and total bilirubin(BIL). Reactivity was different in rat albumin (ALB), depending on the reagents used. This difference needs to be corrected with values available by electrophoresis, or adjusted by rat ALB values, because of the lack of an appropriate measurement method. The enzyme method gave low values for rat CRE, which suggests the need for further examination of this method. The BIL values were extremely low in rat samples. It seems to be necessary to select appropriate methods to measure clinical pathology parameters correctly for rats. There was no deviation in values due solely to the mechanical operations of the analytical equipment. Non-standard initial settings of the equipment (equipment originally intended for human samples, but now applied to animal samples) was the main cause of the wide range of analytical values seen.
