Two trials were conducted to determine the effect of monensin in broiler litter on sheep receiving the broiler litter in their diets. Broiler litter from chickens fed monensin as a coccidiostat, and from chickens receiving no coccidiostat, was included at a level of 30% in 2 sheep diets. In a further 2 treatments, monensin (15 mg kg-1) was added to each of the 2 diets to give a 2x2 factorial experimental design. In the first trial, copper (20 mg kg-1 feed) was added to the diets. These lambs were fed individually at a slightly restricted level of intake. No differences between treatments were observed in feed intake, average daily gain or efficiency of feed utilisation or in the concentrations of zinc, iron and manganese in the liver, glutathione peroxidase in erythrocytes and creatine kinase concentrations in the plasma. Hepatic copper content and copper retention in the livers of the sheep receiving the added monensin were significantly higher (P less than 0.05 and less than 0.01 respectively) than in those not receiving added monensin. The aspartate transaminase and alkaline phosphatase concentrations in the plasma of these sheep were also higher (P less than 0.05) than in those not consuming added monensin. In the second trial, the lambs were group-fed according to treatment and received the diets on an ad lib basis. The mean intakes of the groups receiving the diets with the added monensin, were lower than the intakes by the other groups.(ABSTRACT TRUNCATED AT 250 WORDS)

Plasma alanine aminotransferase, aspartate aminotransferase, creatine kinase and alkaline phosphatase activities were studied in clinically healthy Danish landrace and dwarf kids in seven herds from birth to 12 months of age. The purpose was to evaluate the influence of age, breed and herd on reference values. The mean enzyme levels +/- standard deviation (s) in neonatal dwarf kids were 0.09 +/- 0.04, 1.23 +/- 0.24, 2.79 +/- 1.50 and 18.3 +/- 11.0 mu kat/l respectively. The respective values in landrace kids were 0.13 +/- 0.06, 1.06 +/- 0.22, 2.44 +/- 1.60 and 37.6 +/- 23.6 mu kat/l. In 8-12 months old dwarf kids they were 0.30 +/- 0.11, 1.49 +/- 0.13, 3.28 +/- 0.44 and 11.1 +/- 2.4 mu kat/l respectively and 0.23 +/- 0.05, 1.12 +/- 0.34, 3.68 +/- 1.63 and 14.1 +/- 8.40 mu kat/l respectively in landrace kids of the same age. The 5th to 95th percentile intervals of the enzyme activities were within mean +/- 2s for most age groups in both breeds except alkaline phosphatase. The means and medians were close to each other for the values of alanine aminotransferase, aspartate aminotransferase and creatine kinase but not for alkaline phosphatase. Alanine aminotransferase, aspartate aminotransferase and creatine kinase levels were low at birth and increased with age, whereas for alkaline phosphatase it was vice versa. Significant differences were observed in mean enzyme activities between kids of different ages (within breeds), breeds (in same age kids) and herds (within same breed and age kids). Sex variations (within the breeds) were not observed. It was concluded that plasma enzyme activities are dependent on age, breed and environment.

Several clinical chemical blood variables were compared, in order to evaluate the differences between Na heparinized plasma and serum samples. Samples from 45 healthy horses were used. No differences between the two sample substrates were found for aspartate aminotransferase, lactate dehydrogenase, lactate dehydrogenase-isoenzymes, creatine kinase, alkaline phosphatase, bilirubin, cholesterol, urea, total protein, alpha-globulin, gamma-globulin, albumin, calcium (Ca), phosphate (P), sodium (Na) and potassium (K). gamma-Glutamyltransferase and beta-globulin were significantly higher in heparinized plasma than in serum (each p less than 0.05) while magnesium (Mg) was lower (p less than 0.05). From the horse group used for the study, thoroughbreds in racing condition had significantly higher aspartate aminotransferase, gamma-glutamyltransferase, alkaline phosphatase, bilirubin, P and Mg as well as lower Ca and K values than riding horses, irrespective of the sample substrate used. It was concluded that expect for gamma-glutamyltransferase, beta-globulin and Mg, there was no significant difference between the clinical chemical variables of Na heparinized plasma and serum samples.

1. Plasma calcium, magnesium, inorganic phosphorus, sodium and potassium concentrations, alanine aminotransferase, aspartate aminotransferase, alkaline phosphatase and creatine kinase activities were determined in young and adult non-pregnant non-lactating, early and late non-lactating pregnant and early, mid- and late non-pregnant lactating Danish landrace goats in five herds. The purpose was to determine the influence of pregnancy and lactation on the levels of these parameters and the effect of age and parity on the changes. 2. Calcium, phosphorus, alanine aminotransferase, alkaline phosphatase and aspartate aminotransferase decreased in late gestation. Magnesium and creatine kinase decreased in early lactating goats but increased in subsequent lactation periods. Sodium and potassium fluctuated little during pregnancy and lactation. Calcium, magnesium and potassium profiles were inversely, while phosphorus was directly, proportional to parity. 3. There were significant differences in most ions and enzymes between goats of different herds (within the same physiological state). 4. The transferases and creatine kinase were higher in young goats than in old ones, while alkaline phosphatase was unpredictably high or low in individual goats. 5. Alterations in the level of plasma electrolytes and enzyme activities occur due to pregnancy and lactation and the degree depends on age and parity, influenced also by environment.

The content of hormones that regulate carbohydrate metabolism was studied during the early neonatal period in 80 full-term neonates with intrauterine hypotrophy. Early application to the breast (2 to 6 hours after the birth) was shown to promote the normalization of the hormonal content. The levels of blood serum C-peptide in the newborn depend on the degree of the rise of the mother's body weight during pregnancy and the presence of toxicosis. The levels of cortisol, somatotropic hormone, immunoreactive insulin and C-peptide were determined by the degree of morphological immaturity of the tissues whereas the content of STH and cortisol by the intensity of hypotrophy as well. The moment of the birth and the early neonatal period of children with intrauterine hypotrophy is characterized by a decrease of the activity of lactate dehydrogenase, alpha-hydroxybutyrate dehydrogenase, creatine kinase and aspartate aminotransferase.

Nineteen purebred Beagles of various ages (4, 5, 13, and 47 weeks) were inoculated with North American Trypanosoma cruzi isolates obtained from an opossum (Tc-O), an armadillo (Tc-A), or a dog (Tc-D). Dogs were grouped on the basis of clinical outcome of infection. During the acute stage of disease, dogs of group 1 (n = 7 inoculated with Tc-O or Tc-A) died or were euthanatized because of the severity of disease. Dogs of group 2 (n = 5 inoculated with Tc-O or Tc-A) developed acute disease, but survived to develop chronic disease. Dogs of group 3 (n = 7 Tc-D-inoculated dogs) developed neither acute nor chronic disease. Dogs of group 4 (n = 4--2 dogs 13 weeks old and 2 dogs 47 weeks old) served as noninoculated controls. Clinical signs associated with severe acute myocarditis developed in dogs of groups 1 and 2 between postinoculation day (PID) 15 and 28. Generalized lymphadenopathy and lymphocytosis were observed in all dogs of groups 1, 2, and 3 between PID 14 and 17. Serum alanine transaminase and aspartate transaminase activities and urea nitrogen concentration were high, and glucose concentration was low prior to death of dogs in group 1. Serum activities of isoenzymes of creatine kinase were significantly (P less than 0.05) high in only 1 dog (group 1), whereas serum lactate dehydrogenase isoenzyme activities were not significantly high in any dog.(ABSTRACT TRUNCATED AT 250 WORDS)

Infused beta 2 agonists have been shown to cause focal myocardial necrosis. Serum creatine kinase activity was compared in 13 patients with brittle asthma currently being treated with subcutaneous terbutaline and an age and sex matched control group of patients with moderate asthma having inhaled treatment only. The median serum total creatine kinase activity for patients receiving subcutaneous terbutaline (211 units/l) was greater than that for the control group (120 units/l). The cardiac specific isoenzyme component of creatine kinase was not raised in either group, and the electrocardiograms and serum aspartate aminotransferase activity were normal. Electromyograms in five patients receiving subcutaneous terbutaline with high creatine kinase activity showed changes consistent with myositis in two, one of whom was subsequently shown to have a metabolic myopathy, which is thought to be long standing. No pathological changes were seen in the myocardium at necropsy in a patient who died from an acute attack of asthma while taking subcutaneous terbutaline. These results suggest that the raised creatine kinase activity seen in patients receiving this treatment is unlikely to be myocardial in origin.

The clinical significance of the serum enzymes creatine kinase (CK, EC 2.7.3.2), lactate dehydrogenase (LD, EC 1.1.1.27) and aspartate aminotransferase (EC 2.6.1.1), and the isoenzymes CK 1-3 and LD 1-5, in acute myocardial infarction (AMI) is reviewed. Particular attention is given to electrophoretic analysis of the isoenzymes (and the CK isoforms/subforms) following AMI and thrombolytic therapy. Other protein markers for the monitoring of AMI, including myoglobin and muscle contractile proteins, are also discussed and the potential for the detection of new marker proteins using high-resolution two-dimensional electrophoretic methods is demonstrated. Whilst emphasis is placed upon electrophoretic methods the value of complementary immunoassays is acknowledged in order to maintain a balanced perspective.

Ten minutes after an intravenous flooding dose of phenylalanine to rats, plasma sodium and calcium concentrations were slightly reduced (by 2-7%) but no effects on potassium or phosphate were observed. Creatine kinase activities were significantly increased by phenylalanine injection (by 39%), but alkaline phosphatase, alanine aminotransferase, lactate dehydrogenase and aspartate aminotransferase activities were unaltered. Plasma concentrations of total proteins, albumin, cholesterol, triglycerides, urea, creatinine and glucose were also unaffected. In the presence of anaesthesia, phenylalanine injection had almost identical effects, although the increase in creatine kinase activities did not reach statistical significance. Anaesthesia for 10 min reduced plasma potassium concentrations (by 27%), and calcium (by 5%), though phosphate and sodium were unaltered. The activities of lactate dehydrogenase, creatine kinase and aspartate aminotransferase were reduced by between 36-52%, but alkaline phosphatase and alanine aminotransferase activities were unaltered by anaesthesia. Plasma concentrations of total proteins and albumin were also reduced (both by 9%), but glucose concentrations were increased (by 33%). Anaesthesia had no other significant effects on cholesterol, triglycerides, urea or creatinine concentrations. The qualitative effects of anaesthesia in the presence of raised free phenylalanine concentrations were similar. It was concluded that, except for creatine kinase, determinations of plasma constituents in phenylalanine-injected rats could be made without overt interpretational errors. However, caution is required in interpreting data on plasma constituents from anaesthetized rats.

Data were obtained and analyzed in 229 patients admitted to the coronary care unit from November 1988 through July 1989. The patients were classified into 2 groups: patients without or with only mild left ventricular failure (Killip class I or II) during their hospital stay (group I), and patients who were in Killip class I or II on admission but developed cardiogenic shock during hospitalization (group II). Discriminant function analysis was performed using the following variables: patients' age, history of previous myocardial infarction, diabetes mellitus, blood lactate, urea, creatinine, creatine kinase, aspartate aminotransferase, lactate dehydrogenase concentrations, and chest x-ray cardiothoracic ratio. Variables that were found to significantly discriminate the 2 groups of patients were age, previous infarction, x-ray cardiothoracic ratio, blood urea and lactate concentrations. The risk index was computed, and blood lactate was the variable with the greatest predictive power for shock development. The sensitivity, specificity and predictive value of the risk index, taking various cutoff points, were calculated. With a cutoff value of 1, sensitivity was 65%, specificity 91%, positive predictive value 36% and negative predictive value 97%. With a cutoff value of 2, sensitivity was 53%, specificity 99%, positive predictive value 82% and negative predictive value 96%.
