The coagulation inhibitor protein C was measured in 151 patients with various liver diseases. The protein C level was significantly decreased in patients with alcoholic cirrhosis (n = 73) compared to patients with steatosis (n = 24) (40 +/- 2%) vs. 88 +/- 4%, mean S.E., p less than 0.001). It was also decreased in cases of acute liver damage (n = 8) and in patients with non-alcoholic cirrhosis (n = 15) (35 +/- 7% and 36 +/- 4%, respectively). A significant correlation was found between protein C and Normotest, antithrombin, heparin cofactor II, (r = 0.83, r = 0.82, r = 0.81, respectively, p less than 0.001). There was also a significant correlation between protein C and serum concentrations of
albumin
(r = 0.61, p less than 0.001), but a negative association to bilirubin (r = -0.56). No significant association was found between protein C and
aspartate aminotransferase
, alaline aminotransferase, and gamma-glutamyltranspeptidase. In conclusion, protein C is low in advanced liver diseases and gives the same amount and type of information as Normotest, antithrombin and heparin cofactor II.

Serum chemistry values were obtained from 64 adult San Joaquin kit foxes (Vulpes macrotis mutica) in western Kern County, California (USA). The goal of the study was to establish normal chemistry values for this endangered species. No significant differences were found for mean values of alanine aminotransferase (217.1 IU/l), alkaline phosphatase (44.2 IU/l), cholesterol (145.6 mg/dl), total protein (5.8 g/dl), creatinine (0.63 mg/dl), calcium (8.2 mg/dl),
albumin
(3.0 g/dl), glucose (129.2 mg/dl), amylase (196.8 IU/l), sodium (153.7 mEq/l) and phosphorus (5.42 mg/dl) between sexes or seasons. Significant differences were noted for
aspartate aminotransferase
, blood urea nitrogen and potassium between seasons. Possible disturbances in normal hepatic and renal functions were noted.

Simultaneous multiple automated analyses of liver function can be performed quickly and cheaply, but their usefulness in mass screening is questionable. Reference intervals are frequently applied without regard to race and sex, despite the fact that reported values may vary considerably in relation to these factors. Serum analyte results for greater than 5000 black and white men and women in the CARDIA Study showed clinically and statistically significant differences by race and sex for values of
aspartate aminotransferase
, gamma-glutamyltransferase, alkaline phosphatase, total bilirubin, total protein, and
albumin
; these differences were not explained by differences in age, body mass, reported ethanol intake, smoking, or oral contraceptive use. Results for at least one of these six tests were out of range in 38% of the men and 19% of the women. Sex- and race-specific reference intervals are recommended to decrease the frequency of values reported as abnormal in otherwise healthy young adults.

Specific biochemical and physiological tests of liver function were used to assess 20 consecutive patients undergoing prolonged head and neck surgery with halothane or isoflurane anaesthesia. Hepatic function was assessed by measurement of serum concentrations of total bilirubin and
albumin
, and plasma activity of pseudocholinesterase, gamma-glutamyl transferase (GGT),
aspartate transaminase
(
AST
), alkaline phosphatase (ALP) and hepatic glutathione S-transferase. Plasma clearance of indocyanine green was used as an estimate of hepatic blood flow. No major differences were observed in serum concentrations of GGT, ALP, bilirubin,
albumin
or pseudocholinesterase. Serum
AST
activity in those patients receiving halothane was increased at 24 h and at 48 h compared with those who received isoflurane (not statistically significant). Glutathione S-transferase activity was increased significantly in the halothane group throughout the period of study, compared with those who received isoflurane. Similarly, there was a significant difference between the two groups as measured by plasma clearance of indocyanine green: in the halothane group there was a slower disappearance rate of the dye from plasma at specific times than in the patients who received isoflurane. Our data support the use of isoflurane rather than halothane for prolonged anaesthesia.

Eight hundred broilers were examined to define the reference values of nine blood constituents (total proteins,
albumin
, total and free cholesterol, triglycerides,
aspartate aminotransferase
(
AST
), alkaline phosphatase (ALP), calcium, and phosphorus). The broilers, coming from 20 different commercial poultry operations, were equally divided into different groups according to age (21 and 45 days), strain (Arbor Acres and Hybro), sex, and sampling season (summer or winter). Age and sampling season were the variation sources that most influenced the values of the hematochemical variables examined. The interaction of age with strain influenced total proteins total and free cholesterol, triglycerides, and ALP. The interaction of age with sampling season influenced all hematochemical constituents except triglycerides. Reference limits defined by .975 and .025 fractiles were computed for each blood constituent according to the significance of different variation sources and their combinations. Reference limits defined herein could be used as indicators of metabolic and health conditions of a poultry farm.

The effect of the pyoverdin Pf (an iron chelating agent isolated and purified from Pseudomonas fluorescens CCM 2798) was studied on iron overloaded rat hepatocyte cultures. Iron overload was obtained by addition of 5-80 microM ferric nitrilotriacetate to the culture medium. Twenty-four hours after iron treatment, a significant increase in
aspartate aminotransferase
and lactate dehydrogenase in the culture medium was observed. This corresponded to intracellular decrease in the activity of these two enzymes and correlated with a decrease in
albumin
secretion and an increase in total free malondialdehyde production. The iron toxicity was inhibited by desferrioxamine B. Pyoverdin Pf added to the hepatocyte cultures served as an effective agent to prevent iron toxicity induced in overload. The observed effect of the pyoverdin Pf was as potent as that of desferrioxamine B.

An improved understanding of medical problems of alcoholic patients can be gained from commonly encountered laboratory test results. Liver function tests--such as measures of alkaline phosphatase, gamma-glutamyl transpeptidase,
aspartate aminotransferase
, alanine aminotransferase, and lactate dehydrogenase--may provide evidence of altered hepatic activity of different types, such as obstruction and hepatocellular injury. Other test results may indicate impaired hepatic function, such as measurements of
albumin
, bilirubin, prothrombin time, and blood urea nitrogen. Alterations are also common in electrolytes, blood glucose, magnesium, phosphate, uric acid, and acid-base balance. Disturbances in hematologic function are not infrequent in alcoholic patients, including anemias from many causes, altered granulocyte responses, and thrombocytopenia.

Differences in haematological and clinicochemical profiles of blood of apparently healthy slaughter pigs collected at the farm and at slaughter were investigated in relation to the severity of pathological-anatomical lesions. For this purpose blood-samples of castrated male pigs were collected once at seven different farms and from the same pigs one to three days later at the slaughterhouse. In general, as far as the investigated blood variables are concerned, there were distinct and significant differences in the mean values between farm and slaughter blood-samples. As a rule the mean values of the investigated blood variables were higher in the slaughter blood than in the farm blood. The effects are most pronounced for leucocyte concentration and for the activity of the enzymes creatine kinase (CK), lactate dehydrogenase (LDH),
aspartate aminotransferase
(
AST
) and gamma-glutamyltransferase (gamma-GT). For the blood variables
albumin
, lymphocytes and magnesium there were significant, but not so strong interactions between the sites of blood-sampling and the groups, which were classified according to the severity of pathological-anatomical lesions. Despite the fact that there were significant differences in the mean values of blood profiles of the slaughter pigs between the sites of blood sampling, this effect was only manifest as a difference in the level of the values of the blood variables. This means that clinicochemical and haematological profiles from groups of pigs at slaughter reflect the herd's health status at the farm and can be used when monitoring it.(ABSTRACT TRUNCATED AT 250 WORDS)

A study was conducted to investigate morphologic as well as metabolic characteristics of microcarrier-attached hepatocytes in culture, and also to evaluate the effect of intraperitoneal transplantation of the microcarrier-attached hepatocytes on acute hepatic failure in rats induced by D-galactosamine (GalN). Rat hepatocytes were isolated by collagenase perfusion, and cultured on collagen-coated microcarriers. Protein synthesis estimated by [14C] leucine incorporation was four-fold higher in microcarrier culture than in cell suspension. The rates of
albumin
, transthyretin and bile acid syntheses in hepatocytes cultured on microcarriers were similar to those in monolayer culture. When microcarrier-attached hepatocytes were intraperitoneally transplanted into rats with Galn-induced acute liver failure, a marked improvement in survival rate was observed as compared with control rats which received injections of microcarriers alone (80% vs 0% beyond 6 days of transplantation). Mean serum
glutamate oxaloacetate transaminase
(SGOT), serum glutamate pyruvate transaminase (SGPT), methionine and glucose levels were similar in both groups, while serum bilirubin and ammonia levels were lower (P less than 0.1, P less than 0.05) in rats transplanted with the microcarrier-attached hepatocytes. Immunohistochemical examinations revealed that the transplanted hepatocytes around microcarriers had
albumin
synthesis activity, whereas almost no
albumin
synthesis was demonstrated in recipient liver. In conclusion, intraperitoneal transplantation of the microcarrier-attached hepatocytes will provide sufficient metabolic support, representing detoxication of ammonia (and presumably bilirubin) and synthesis of
albumin
, to allow GalN-damaged liver function to restore. Microcarrier culture of isolated hepatocytes seems to be one of the most appropriate tools for an artificial liver support.

Prothrombin time, serum albumin, aminotransferases and liver size were evaluated in 40 consecutive cases of kwashiorkor. Eleven (27.5%) of the 40 patients died. Eight out of the 11 patients who died had a prolonged prothrombin time of more than 3 s above the control compared to only 4 out of the 29 who survived (p = 0.005). Mean serum
aspartate aminotransferase
(
AST
), alanine aminotransferase (ALT)
albumin
, globulin and liver size were abnormal but similar in both groups. These results may indicate a predictive mortality value of prothrombin time in kwashiorkor.
