The aim of this study was to produce large liver tumors reliably, and to diagnose the tumors during development. Therefore, New Zealand white rabbits were treated with N-nitrosodiethylamine orally three times per week by gavage and were examined by clinical-chemical assay at regular intervals during the average treatment period of 14 months. The total cumulative dose was 1200 mg N-nitrosodiethylamine over 14 months. After a short treatment period the initial dose of 3 mg/kg had to be reduced to 1.5 mg/kg. In all 11 treated animals (100%) liver tumors were seen at the end of the study. Four control animals did not show any neoplastic changes. Clinical parameters investigated were for an assessment of liver function, total protein, urea, creatinine, alanine aminotransferase,
aspartate aminotransferase
, alkaline phosphatase, albumin and neuraminic acid as well as some serum electrolytes. The in vivo diagnosis of liver tumors based on changes in these parameters proved to be relatively unreliable. The liver enzyme tests and urea concentration only yielded significant changes when the liver tumors were very large. Changes in neuraminic acid levels were the most reliable indicator for the presence of a liver tumor in this animal model. In the 11 treated animals, serum values of this marker increased towards the end of the study by an average of 300 mg/dl. The induced tumors were mainly hepatocellular carcinomas. Only in 1 animal was a hepatocellular adenoma found. Further primary tumors diagnosed were six adenomas in the kidneys and two

adenomas, as well as nasal cavity tumors (two papillomas, one carcinoma, one adenoma and one adenocarcinoma). In 70% of the treated rabbits the hepatocellular carcinomas had metastasized to the lungs.

In order to assess the systemic toxicity of Zwittergent 3-14, a detergent used in the protein-detergent complex vaccines for its attractive immunogenic properties, a subacute toxicity study was performed. In this 4-week toxicity experiment five female and five male rats per group were injected intramuscularly with 0.25 ml of 0, 75, 750 and 7500 micrograms Zwittergent ml-1 sterile saline solution. Body weight and food intake were recorded weekly. At day 24 urine was collected for semiquantitative (pH, protein, ketone bodies, bilirubin and occult blood) and quantitative analyses (protein, creatinine and volume). At the end of the experiment blood was sampled for haematological [haemoglobin (Hb), packed cell volume (PCV), erythrocytes, leucocytes, reticulocytes and thrombocytes and differential white blood cell count] and biochemical analyses (alanine aminotransferase and

, ovaries and testes were weighed. The underlined organs and the musculus quadriceps including the injection sites were examined histopathologically. Indications for systemic toxicity were noticed in the high-dose group and included occult blood in urine, elevated protein/creatinine ratio concomitantly with an increased urine volume and increased relative kidney weight indicating a slight disturbance of the kidney function. Some changes in haematological parameters (decreased PCV and increased numbers of thrombocytes eosinophils and monocytes) and a decreased glycogen content in the liver were recorded in the high dose group. ) was 21.6 +/- 5.7 (+/- SD) IU/l, of alanine aminotransferase (ALT) 40.4 +/- 13.0 IU/l and of the

/ALT ratio 0.6 +/- 0.2. These values showed only minor changes over years. In 96 bitches with the cystic hyperplasia-pyometra complex there was a very significant increase of the
AST
, decrease of the ALT and increase of the
AST
/ALT ratio. The changes were more pronounced in 62 clinically ill bitches with typical endometritis post oestrum, in 18 dogs with gram negative organisms in the
uterus
and in 53 bitches with white blood cell (WBC) levels higher than 40 X 10(9)/1. Renal failure had no influence on the specific changed values. The changed values returned either temporarily to normal after prostaglandin (PGF2 alpha)-treatment or definitely after ovariohysterectomy.

The overall rate of inactive ovaries diagnosed by two consecutive rectal examinations was 8.5 per cent for 7751 lactations. Inactive ovaries had an inverse association with increasing parity and were directly associated with twinning, retained placenta, primary metritis and high milk yield after calving in heifers. No independent associations were established with stillbirth, a low milk yield in the last 120 days before calving, a long dry period, ketonuria or a high serum
glutamate oxaloacetate transaminase
activity in the first week after calving. The presence of inactive ovaries in the previous lactation was a significant predictor of the trait. Cows with either metritis or inactive ovaries were at a greater independent risk of not conceiving within 150 days after calving and the combined risk associated with both factors was higher than their sum. It is concluded that while damage to the

repeatability and an energy shortage after calving are responsible for ovarian inactivity, this trait was not associated with fatty liver. Such an association could be due to an indirect causal relationship, the outcome of post parturient uterine diseases which are associated independently with overfeeding before calving and inactive ovaries.

Female BALB/c and B6C3F1 mice were examined after a 3-wk exposure to dietary estradiol (0, 400, 800, 1600, and 3200 ppb) in a purified (AIN-76A) or a natural-ingredient (NIH-07) diet. The use of AIN-76A was associated with a 9-13% greater (p less than 0.001) body weight and a 36-43% higher (p less than 0.001) serum cholesterol in both mouse genotypes when compared to mice fed NIH-07. Conversely, when fed NIH-07, both mouse genotypes had a 20-22% higher (p less than 0.003) serum urea nitogren and 2-3.5% higher erythrocyte count (p less than 0.001) and hemoglobin concentration (p less than 0.04) than when fed AIN-76A. Reduced erythrocyte parameters suggest that chronic feeding of the purified diet might result in anemia. No significant compound or diet-related differences were noted for serum creatinine, alanine aminotransferase,

, or gamma-glutamyl transferase. Although there was no diet effect on absolute or differential white blood cell count, estradiol caused a decrease in the total white blood cell count (p less than 0.014) and an increase in the percentage of polymorphonuclear leukocytes (p less than 0.014) in BALB/c and decreased the percentage of lymphocytes (p less than 0.005) in B6C3F1 females. In addition, estradiol increased uterine weight and inhibited thymic and splenic weights in one or both genotypes. The concentrations of acid cathepsin and beta-glucuronidase are significantly decreased in pregnancy and increase 3-4-fold during post-partum involution. 3. The concentrations of beta-galactosidase and acid phosphatase are not decreased in pregnancy and increase only 2-3-fold during involution. 4. The concentrations of these four acid hydrolases increase linearly during the first 4 days post partum and reach their peak values at the same time that wet weight and collagen content fall to their lowest point. 5. The concentration of deoxyribonuclease is depressed in pregnancy but does not rise above normal in the post-partum period. 6. The activity of enzymes with a regulatory function in the pathways of glycolysis, glyconeogenesis and NADP-generation, and the tissue content of DNA, protein, glycogen, triglycerides (TG), phospholipids (PL), cholesterol and dry matter were investigated in placentas from deliveries accompanied by fetal distress as a result of umbilical cord compression or placental dysfunction in toxemic pregnancies. In placentas from cases of fetal distress due to umbilical cord compression, there was increased activity of pyruvate kinase, 6-phosphogluconate dehydrogenase and NADP-malate dehydrogenase, and decreased activity of phosphoenolpyruvate carboxylase. The activity of
aspartate aminotransferase
was unchanged, and that of glucose-6-phosphate dehydrogenase was slightly elevated. The tissue content of dry matter, DNA, TG and PL was increased, whereas the protein, cholesterol and glycogen concentrations remained unaltered. In placentas from deliveries accompanied by fetal distress due to placental dysfunction, pyruvate kinase, when calculated per mg protein, was the only enzyme with decreased activity. TG, PL, glycogen and dry matter content were increased, DNA concentration was decreased, and protein and cholesterol remained unchanged. It is suggested that the divergent placental metabolic patterns found in the two fetal distress groups are related to the different levels of disturbed oxygen passage along the
uterus

The efficacy of Tiron (4,5-dihydroxybenzene 1,3-disulfonic acid disodium salt) was examined in the treatment of beryllium-induced maternal and developmental toxicity in rats. Single administration of beryllium nitrate at a dose of 50 mg/kg (i.m.) on day 13 of gestation caused reductions in fetal and placental weights, the number of implantation sites and number of corpora lutea, as well as causing post-implantation loss, stunted growth, increase in the number of resorptions, and also a disturbed sex ratio. Maternal toxicity was demonstrated by reduction in body weight gain. Administration of beryllium also showed significant alteration in the hematological and biochemical indices of the mother as well as the fetus. Marked decreases were recorded in hemoglobin percentage, blood sugar levels, serum protein contents and serum alkaline phosphatase activity. By contrast, significant elevation was found in the activity of transaminases (
aspartate aminotransferase
and alanine aminotransferase). Tissue protein contents, glycogen contents, activities of alkaline phosphatase, adenosine triphosphatase and succinic dehydrogenase of kidney, lungs and

, and maternal and fetal liver all showed significantly decreased values after beryllium exposure, and remarkable elevation was observed in acid phosphatase, glucose-6-phosphatase and hepatic lipid peroxidation. These parameters were restored considerably with administration of 471 mg/kg i.m. Tiron from days 14 to 18 of gestation. Atomic absorption spectrophotometry also revealed a high concentration of beryllium in different organs of pregnant rats. Interestingly, a small amount of metal ion was also detected in the fetus and reduced accumulation of beryllium was noticed after Tiron treatment.

The aim of the study was to prove a correlation between creatine kinase (CK; EC 2.7.3.2.) and
aspartate aminotransferase
(AST; EC 2.6.1.1.) activities in serum and the severity of endometritis. We (i) determined clinical and clinical-chemical (CK, AST, bilirubin) parameters on 87 cows with abomasal displacement (DA), (ii) measured CK, AST and glutamate dehydrogenase (GLDH; EC 1.4.1.2.) in serum and uterine tissue samples in 10 slaughter cows, and (iii) compared the serum reaction (CK, AST, bilirubin) of six healthy, non-pregnant cows after an inter-auterine application of a mild irritating 0.2% peroxyacetic acid (Uterofertil) with that of four healthy cows after an intrauterine application of 0.9% sodium chloride solution. Uterine tissue contains high activities of CK (2940 +/- 1140 U/g protein) and AST (159 +/- 25 U/g protein). Cows with DA have increased serum CK and AST activities, which correlate with the degree of endometritis. The DA without endometritis also comes along with slightly increased CK (quartiles 181, 259 and 288 U/l) and AST (101, 138 and 199 U/l) activities. In pregnant cows these activities are higher than in non-pregnant cows. Irritation of the
uterus
with Uterofertil leads to increased serum CK but not AST. After the exclusion of evaluated CK as a result of muscular damage or hypocalcaemia, this enzyme can be used as a screening parameter in the diagnosis of endometritis. In each clinical case it is necessary to determine if increased AST activities are muscle-, liver- or

-dependent.

in different age groups (12, 18 and 24 months) of naturally menopausal rats. The rats were given the subcutaneous injection of 17beta-estradiol, progesterone and combination of estradiol and progesterone for 1 month. The activity of SOD, Ala-AT and Asp-AT was measured in the brain (cerebral hemisphere, CH), heart, liver, kidney and

. The activity of SOD decreased with age in all the tissues taken particularly in liver. After HRT the enzyme activities were increased as compared to age-matched controls in all the tissues of aging rats. The activities of transaminases (Ala-AT and Asp-AT) showed a decrease with age in all the tissues and administration of estradiol and combination of estradiol and progesterone further decreased both the aminotransferases. Our study elucidates that increased activity of SOD contributes in protection of cells from oxygen toxicity by catalyzing the dismutation of free radicals in tissues. Furthermore, the HRT probably decreases gluconeogenesis and proteolysis by decreasing the activities of Ala-AT and Asp-AT in aging rat tissues.
