The possible role of mesenteric mast cell degranulation as the mediator of the initial UCS effects in the complex sequences leading to conditioned taste aversion (CTA) was studied. Both LiCl and Compound 48/80, a potent mast cell degranulator, produced CTA to 10% sucrose. Whereas the Compound 48/80 groups displayed massive mast cell degranulation, neither the LiCl treated nor saline control groups demonstrated any histologically determinable alterations. Administration of the antihistamine chlorpheniramine at dosages known to block radiation-induced CTA before the sucrose CS-UCS pairings did not block either LiCl- or Compound 48/80-induced CTA; the antihistamine actually facilitated the aversion. However, pretreatment with the antihistamine did not alter mesenteric mast cell morphology.

Studies were performed in animals to establish whether antigen prefeeding could present homocytotropic antibody synthesis and the induction of gastric ulcer resulting from mucosal anaphylaxis. A single digestive exposure to 1 or 10 mg ovalbumin induced a state of specific immunologic tolerance. Inhibition of the formation of specific reagins was shown by the study of mast cell degranulation. Tolerant animals presented a reduced incidence of gastric ulcer after subsequent mucosal challenge. These results are important for the development of methods of prevention and treatment of allergic diseases.

An asthmatic atopic patient who developed bronchoconstriction during repeated forced vital capacity manoeuvres and after exercise is reported. Since increases in total airway and upper segment resistances and drop in FEV1 and Vmax 50 were induced, it is considered that bronchoconstriction involved most of the airways. Partial prevention of this effect could be demonstrated after inhalation of an adrenergic bronchidilator or disodium cromoglycate. Atropine completely prevented the flow response but did not alter the total airway resistance increase. Bronchoconstriction is interpreted as partially related to mast cell disruption and cholinergically mediated stimuli.

Significant alterations in the circulatory properties of platelets have been documented during IgE-induced systemic anaphylactic shock in the rabbit. Within 30 to 60 sec after i.v. antigen challenge, platelet aggregation occurs in both the venous and arterial circulations. The platelet aggregates then sequester in small blood vessels of various organs, particularly in the lung. The organ sequestration results in the development of a profound thrombocytopenia within 3 to 5 min after antigen challenge. Fifteen min later deaggregation of platelets occurs and the platelets return to the peripheral circulation within normal, prechallenge levels by 60 min. Additional experiments demonstrated that platelet depletion before antigen challenge abrogates the lethal effects and significantly reduces the pathophysiologic manifestations of IgE-induced systemic anaphylaxis. We conclude that the IgE-induced platelet alterations, probably induced by the intravascular release of basophil and perhaps mast cell-derived platelet-activating factor (PAF), play a major role in the pathogenesis of systemic anaphylaxis in the rabbit.

Comparative counts of Alcian Blue-Basic Fuchsin-stained mast cells of the facial skin and bone marrow have been made in young rats of different sexes and strains, fed a diet deficient in magnesium (0.8 to 1 mg/100 g dry weight) for 4 weeks. Normal rats fed a magnesium-supplemented diet (65 mg/100 g dry weight) had about three times as many mast cells in the tibial metaphysis as in the facial skin. In both males and females fed the Mg-deficient diet, the marrow mast cells increased five to six times, while their number was concomitantly decreased in the skin. The marrow mast cells became also polymorphic, an indication of a possible preferential renewal site. Gonadectomy in the males had no effect on the above pattern. The administration of large doses of testosterone to males and estradiol to females depressed the mast cell population increase in the bone marrow and at the same time, moderated the loss of skin mast cells.

Two types of mast cell can be identified histochemically in the dermis of the rat's external ear. One type is recognized by the binding of concanavalin A (con A) to the cytoplasmic granules (con A-positive cells) while in the other type (con A-negative cells), the granules do not bind con A. The granules in both types are stained metachromatically by toluidine blue. Antidromic stimulation of the great auricular nerve for 2 min results in an increased proportion of degranulating mast cells in the auricular dermis and both types of cell are affected to an approximately equal extent. In discussion of this observation, it is argued that both the con A-positive and the con A-negative mast cells are probably involved in the mediation of vasodilatation due to axon reflexes in injured skin. The proportions of degranulating mast cells determined in histological preparations varied with the fixatives (Carnoy and glutaraldehyde-formaldehyde) used, but the increased degranulation due to antidromic nervous stimulation could be detected after either fixation.

The relative number of mast cells in the ear lobes' skin (pinna) of nude (athymic) nu/nu and normal (thymic) nu/+ heterozygotes of Balb/c mice was similar. The results obtained contradict some suggestions about the general influence of the thymus on the number of mast cells in the skin and suggest the existence of some local factor(s) in regulation of skin mast cell numbers.

The histology of early feeding lesions of the cattle tick B. microplus has been studied using 32P labelled larvae to standardize the duration of attachment. Critical studies were made on 3-h lesions in six separate experiments on different groups of British breed animals. Each group consisted of three animals--one previously unexposed to ticks, one of high resistance and one of low resistance. The degree of mast cell disruption, eosinophil concentration and degranulation, and the extent of epidermal vesiculation were all significantly greater at the site of attachment on highly resistant hosts. In previously unexposed animals there was no mobilization of eosinophils nor mast cell breakdown and no epidermal vesiculation. Possible immune mechanisms producing mast cell disruption and the infiltration and concentration of eosinophils are suggested, and the effect of eosinophil degranulation on larval attachment and feeding is discussed.

A new technique for the study of the mechanism of enzymes has been developed. An enzyme, modified by an active-site directed reagent, is digested by one or more proteases. The resulting mixture of oligopeptides is then analyzed directly by gas chromatography-mass spectrometry without the use of separation or isolation procedures. A comparison with unmodified enzyme identifies the modified residue as well as quantifies the reaction. This approach has been applied to the identification of Glu-270 in the active site of carboxypeptidase A using a carbodiimide as modification reagent. Studies on the possible incorporation of 18O (from 18O-enriched water) into Glu-270 or other acidic residues near the active site of carboxypeptidase A show that the oxygens of the carboxyl groups of these residues are not exchangeable.

Mast cells have been counted in sections of iliac bone from 61 control subjects at necropsy. Mast cells were found in all but three, and the range was 0-33-7, median 1-95 per mm2 marrow. The majority (82%) had less than 4-99 mast cells per mm2 marrow; in 37-7% there was less than 1 mast cell per mm2 marrow. In a group of 45 patients with chronic renal failure there was a significant increase in the numbers of mast cells (P less than 0-001) with a range of 0-96-55-63, median 9-55 per mm2 marrow. Mast cells were common in the areas of marrow fibrosis associated with osteitis fibrosa but this was not the sole cause of the increase since there was also an excess of mast cells in the non-fibrous parts of the marrow. There was a tendency towards greater numbers of mast cells in those cases with most marked osteitis fibrosa in association with the prominent marrow fibrosis, but there was no significant relationship between mast cell numbers and other features of oesteitis fibrosa such as the number of osteoclasts and the amount of woven bone formation. There was no relationship between the numbers of mast cells and the amounts of total bone, ostoid, percentage mineralization of cancellous bone, or the presence of osteomalacia.
