This simple, reproducible colorimetric method for determining the activity of
carboxypeptidase A
(EC 3.4.17.1) is based on measuring the absorbance at 505 nm of a quinoneimine dye produced from the action of this enzyme on the new substrate p-hydroxybenzoyl-glycyl-L-phenylalanine. The enzyme acts on the substrate to produce p-hydroxybenzoyl-glycine and L-phenylalanine. The former is then hydrolyzed by hippuricase (EC 3.5.1.14) to produce p-hydroxybenzoic acid and glycine. Finally, oxidative coupling of p-hydroxybenzoic acid with 4-aminoantipyrine by sodium periodate forms a quinoneimine dye. The Km for the reaction with this substrate is 3.6 mmol/L; the optimum pH is 7.8. Our within-run and between-run CVs are 4.3% and 6.6%, respectively. The activity of
carboxypeptidase A
in serum correlates well with that of
lipase
(r = 0.96) and immunoreactive elastase-1 (r = 0.76).

Alterations in pancreatic function and structure were examined in suckling mice infected intraperitoneally with reovirus type 3. The results were compared to pancreatic zymogen enzyme activities and histology in adult mice infected with the same virus. No effect of the rovirus type 3 on the adult mice could be elicited. In contrast, the suckling mice infected by the reovirus type 3 revealed a definite change in pancreatic zymogen enzymes. However, the zymogen enzymes were affected in a nonparallel fashion and three groups of enzymes with different responses were noted. Amylase and
lipase
activities were significantly diminished (P less than 0.001) at 6 days of viral infection. The endopeptidases, trypsin (P less than 0.025) and cymotrypsin (P less than 0.001) activities were increased significantly in the infected group. The exopeptidases,
carboxypeptidase A
and B in the infected animals were not changed significantly compared to the control. It seems reasonable that the reovirus type 3 infection in the suckling mouse causes diminished
lipase
and amylase activities that might contribute to the pathogenesis of viral enteritis.

It has been noted that from days 18 to 22 (birth) during the second intrauterine period of morphogenesis of the rat pancreas the accumulation of enzymes increases dramatically. We studied the effect of altered maternofetal blood flow on the development of the rat pancreas during the critical second period. Our studies indicate that during pancreatic cytodifferentiation, reduction in maternofetal blood flow not only reduces the weight of the pancreas (68% of control) and diminishes the total activities of enzymes but that the changes in specific activities of the enzymes do not appear to be coordinate. The specific activities of amylase decreased to 59,000 units from the control value of 103,000 units (P less than 0.01) and
lipase
decreased to 4000 units from a control value of 7350 units (P less than 0.001). In contrast, the specific activities of trypsin (ogen), chymotrypsin (ogen) and (pro)-

and B are not changed. These results suggest that reduction in maternofetal blood flow caused a selective decrease of fetal rat amylase and

The transplantable pancreatic acinar carcinomas of rat established recently provide useful model systems to examine the composition of secretory proteins as well as the secretory process in transformed pancreatic exocrine epithelium. The neoplastic acinar cells exhibit considerable variation in the extent of cytodifferentiation. In the present study the enzymatic profile of this heterogeneous tumor cell population has been investigated by the indirect immunofluorescent technique using antibodies against six pancreatic enzymes. By immunofluorescence, all neoplastic cells stained positively for the six enzymes tested: amylase,

, chymotrypsinogen, trypsinogen, and ribonuclease. Some variability in the intensity of immunofluorescence was noted, suggesting possible quantitative differences in the content of a given enzyme among tumor cells. These observations suggest that neoplastic acinar cells with or without secretory granules contain secretory proteins, but to a variable extent.

The diminution of the three pancreatic enzymes may be related to the destruction of pancreatic endocrine tissue that occurs in these diabetic animals.

Mouse bone marrow mast cells sensitized with monoclonal IgE and activated with specific antigen released 2.8 +/- 0.5 ng of platelet-activating factor (1-0-alkyl-2-acetyl-sn-glyceryl-3-phosphorylcholine) (PAF-acether)/ 10(6) cells. The PAF-acether was identified by its ability to aggregate fully aspirin-treated washed rabbit platelets in the presence of an adenosine diphosphate (ADP)-scavenger complex, by its co-chromatography with [3H]-labeled semi-synthetic PAF-acether and synthetic 1-0-octadecyl-2-acetyl-sn-glyceryl-3-phosphorylcholine, and by its inactivation by phospholipases A2, C, and D and not by
lipase
A1. The antigen-initiated release of PAF-acether, leukotriene C4 (LTC4), and leukotriene B4 (LTB4), and the secretion of the granule marker beta-hexosaminidase were not diminished by washing the cells before challenge, indicating that they were due to the interaction of antigen with the IgE fixed on the cell membrane and not to phagocytosis of immune complexes formed in the fluid phase. The parallel antigen-induced dose-response relationship, along with the superimposable time-course of the extracellular appearance, of beta-hexosaminidase, PAF-acether, and both leukotrienes indicated that the origin of these diverse mediators was from a common cell type with IgE-Fc receptors. Ethanol extraction of antigen-stimulated bone marrow-derived mast cells revealed the early transient appearance of a cell-associated platelet-aggregating activity, the action of which on platelets, like PAF-acether, was independent of ADP and arachidonic acid metabolism. The cell-associated activity contained a novel product that eluted at 13 min during high performance liquid chromatography (HPLC) (solvent hexane:n-propanol:water, 46:46:8), permitting resolution from PAF-acether and lyso-PAF-acether (1-O-alkyl-sn-glyceryl-3-phosphorylcholine), which eluted at 29 min and 30 min, respectively. The cell-associated material, which differs from lyso-PAF-acether, the putative precursor of PAF-acether, in being active in the bioassay on platelets may represent a newly recognized intermediate in the generation of PAF-acether. 1. Pancreatic juice was collected from six pigs of 48 kg initial weight fitted with a collection catheter in the pancreatic duct and a return catheter in the duodenum. 2. Measurements of flow and composition of the juice were made during 24 h periods after adaptation to isonitrogenous diets based on barley, wheatings and fish meal (diet BWF) or starch, sucrose, casein, maize oil and cellulose (diet SSC), given in a change-over design. Measurements were also made during the periods of adaptation to a change from one diet to the other. 3. Mean flow-rates for pigs adapted to diets showed a highly significant four-fold difference between diets; values were 4962 ml/d for diet BWF and 1273 ml/d for diet SSC. The hourly volumes of juice were very variable and showed no clear response to feeding and no consistent diurnal pattern for either diet. 4. Alcohol-induced hypersecretion probably contributes to chronic alcoholic pancreatitis. Feeding of raw soybean flour or soybean trypsin inhibitor also stimulates protein secretion of the pancreas. Therefore, we tested whether or not the pancreatic damage is increased by additional feeding of raw soybean flour in rats fed 20% ethanol. After 11 months, we classified the morphological lesions of the pancreas into seven stages of severity calculated by means of a discriminating procedure. In order to characterize the secretory capacity of the pancreas, we measured the outputs of
lipase
, phospholipase, A, alpha-amylase,
carboxypeptidase A
, chymotrypsin, and bicarbonate. Compared with the alcohol-fed animals, the rats fed with alcohol and soya exhibited a lower average degree of morphological damage in the pancreas. Hypertrophy and hyperplasia of the parenchyma and accumulation of secretory products within the acinar cells were main features. On the other hand, some separate regions of the pancreas showed intraductal secretion precipitates as well as plugs, which were sometimes associated with atrophy of acinar cells. Feeding with soybean diet grossly reduced the alcohol-induced enzyme hypersecretion. In the early phase of alcohol-induced pancreatic damage, long-term soybean flour diet thus reduces morphological lesions and hypersecretion of the rat pancreas, whereas protein synthesis in the acinar cells appears increased. However, the precipitation of secretory products on ductal epithelium, the increased formation of plugs, and the more frequent acinar atrophies suggest the development of significant tissue injuries.
