Pairs of monoclonal/polyclonal antibodies directed against interleukin-1 (IL-1) alpha, IL-1 beta and tumour necrosis factor (TNF) alpha were used for immunocytochemical identification of cytokine-containing cells in cryostat sections of human fetal thymuses and thymomas. In the fetal thymus immunoreactivity for IL-1 alpha was mainly confined to the medulla and was detected in S-100 positive interdigitating reticulum cells. The pattern of immunoreactivity for IL-1 beta was similar to that for IL-1 alpha, but the number of positive cells was much lower. Cells positive for TNF alpha were extremely rare in the fetal thymus. In 11 thymomas macrophages were constantly present and were regularly distributed throughout the tumour, whereas S-100 positive interdigitating reticulum cells were fewer and were characterized by a zonal distribution. Thymoma-associated macrophages were negative for IL-1 beta and were poorly reactive for IL-1 alpha, only a few positive cells being detected in five of the cases. Some macrophages with immunoreactivity for TNF alpha were detected in seven cases; they formed rosettes with surrounding lymphocytes or were located in a perivascular position. A marked immunoreactivity for TNF alpha was constantly detected in mast cell granules, which were observed in nine thymomas but not in fetal thymus. Positive immunoreactivity of interdigitating reticulum cells for IL-1 alpha was confirmed in five reactive lymph nodes and was also observed in Langerhans' cells in dermatopathic lymphadenitis. Mouse cell lines of different lineages have been established which constitutively secrete large quantities of recombinant mouse interleukins (mIL2, mIL3, mIL4 or mIL5). An existing bovine papilloma virus-based expression vector, pBV-1MTHA, was modified to allow transformed X63Ag8-653 myeloma cells, NIH 3T3 fibroblasts and C127 mammary tumor cells to stably carry multiple copies of the vector, to express the inserted cDNA encoding a single interleukin constitutively, and to secrete the interleukin in high quantities. Cell lines transformed with mIL2 cDNA stably carried 30-100 copies of the plasmid per cell and constitutively secreted biologically active mIL2 in quantities similar to those produced by murine EL4 thymoma cells or rat spleen cells stimulated with mitogens. Deletion of the 3' untranslated region containing AT-rich sequences from the mIL2 cDNA resulted in a 100-fold increase in the constitutive production and secretion of mIL2 by the transformants. Addition of a heavy metal further increased the production 2 to 6-fold. Cells transformed with 3'-deleted mIL3 cDNA constitutively secreted 300-1000 times higher activities of mIL3 than the myelomonocytic leukemia line WEHI3. mIL4 produced by the similar transformants induced [3H]thymidine uptake of a T cell line, a mast cell line and B leukemia cells, and enhanced the production of IgG1 by B cells. IL4 titers were 150 times higher than those produced by the concanavalin A-stimulated T cell line 2.19. mIL5 was secreted by similar transformants at 10-fold higher titers than those produced by concanavalin A-stimulated 2.19 T cells, as judged by the proliferation and maturation of B cell leukemia BCL1. The expression vectors should be useful in establishing eukaryotic cell lines producing proteins from full length cDNA clones at higher rates. The established cell lines secreting IL2, 3, 4 or 5 at high rate should be useful sources for these interleukins in the investigation of their function in the immune system.

Thymoma was diagnosed in 15 dogs at Angell Memorial Animal Hospital between 1972 and 1983. All thymomas developed in the cranial portion of the mediastinum. An autoimmune paraneoplastic syndrome was observed in 10 (67%) of the dogs and included myasthenia gravis, nonthymic neoplasms, and polymyositis. Clinical signs were variable and inconsistent, depending on whether they were attributable to the cranial mediastinal mass or to the paraneoplastic syndrome. Eleven dogs were necropsied. Two thymomas had gross characteristics of malignancy. In 3 cases, a cell consistent with a subclass of mast cell was found and in 1 thymoma, melanocytes were observed.

T1/ST2 is a member of the interleukin (IL)-1 receptor superfamily, possessing three immunoglobulin domains extracellularly and a Toll/IL1R (TIR) domain intracellularly. The ligand for T1/ST2 is not known. T1/ST2 is expressed on Type 2 T helper (Th2) cells, and its role appears to be in the regulation of Th2 cell function. Here, we have investigated T1/ST2 signal transduction, using either transient overexpression of T1/ST2 or a cross-linking monoclonal antibody to activate cells. We demonstrate that T1/ST2 does not activate the transcription factor NF-kappaB when overexpressed in murine thymoma EL4 cells, or in the mast cell line P815 treated with the anti-T1/ST2 antibody. However, a chimera comprising the extracellular domain of the type 1 IL-1 receptor and the intracellular domain of T1/ST2 activates NF-kappaB both by overexpression and in response to IL-1. This artificial activation requires the IL1RAcP recruited via the extracellular portion (IL1R1) of the chimera. T1/ST2 is, however, able to activate the transcription factor activator protein-1 (AP-1), increase phosphorylation of c-Jun, and activate the MAP kinases c-Jun N-terminal kinase (JNK), p42/p44 and p38. Anti-T1/ST2 also induces the selective expression of IL-4 but not IFN-gamma in naive T cells. Importantly, this effect is blocked by prior treatment with the JNK inhibitor SP600125 confirming that JNK as a key effector in T1/ST2 signaling. The lack of effect on NF-kappaB when T1/ST2 is homodimerized identifies T1/ST2 as the first member of the IL-1 receptor superfamily so far studied that is apparently unable to activate NF-kappaB, consistent with evidence indicating the lack of a role for NF-kappaB in Th2 cell function.

Of 1146 caprine necropsy or biopsy specimens submitted from 1987 through 2011 to the Veterinary Diagnostic Laboratory at Oregon State University, 100 goats (8.7%) had 102 tumors. Detailed records were available for 89 cases. Fifty-five goats were female, 17 were castrated males, and 12 were intact males. Breeds included 21 Nubian, 16 Pygmy, 10 Pygora, 8 Alpine, 4 Angora, 4 Saanen, 2 Toggenburg, and 9 crossbred goats. Dwarf, Nubian, and Saanen goats were overrepresented and Alpine and Boer goats underrepresented among cases with neoplastic disease in comparison to submissions overall. Age ranged from 7 months to 19 years (median, 7 years). Histopathology was performed on 97 tumors. Lymphoma (n = 17) was the most common tumor, followed by cutaneous squamous cell carcinoma (n = 10) and thymoma (n = 9). Most lymphomas were multicentric. All 7 mammary neoplasms were adenocarcinomas. Five of 7 vascular proliferations were hemangiosarcomas. All 4 melanocytic tumors were classified as (malignant) melanoma. Rarely reported caprine tumors included a choroid plexus carcinoma, 2 rhabdomyosarcomas, and 3 pheochromocytomas. Cutaneous round cell tumors were provisionally diagnosed as 2 histiocytomas and 5 mast cell tumors. Single cases of previously unreported caprine tumors included amyloid-producing odontogenic tumor, myxosarcoma, sebaceous carcinoma, apocrine sweat gland adenoma, and thyroid carcinoma. Nonneoplastic entities included 2 cases of mammary fibroadenomatous hyperplasia and single cases of vascular hamartoma, cervical adenomatous hyperplasia, and cervical leiomyofibromatosis. The results of this 25-year retrospective study indicate that lymphoma in particular and tumors in general are common in goats.
