Androgen secreting Leydig cells in the adult are differentiated with a very low turnover, however, Leydig cell tumours can arise spontaneously or after treatment with toxins. This study in the rat investigated whether changes in components of programmed cell death could be involved. In contrast to their absence in differentiated Leydig cells, antiapoptotic Bcl-2 and proapoptotic Bax were expressed in tumours. Bak and Bcl-xl were found in both tumour and normal Leydig cells. Apoptosis was induced in subcutaneous implants of Leydig cell tumour by ethane dimethanesulphonate (EDS) which is known to kill differentiated Leydig cells. The marked regression of the tumour following EDS treatment was transient and re-growth occurred between 6 and 14 days later. Tumour regression and growth was associated with a similar weight pattern in the seminal vesicles caused by changes in serum testosterone. During tumour regression, clusterin and Bax proteins were elevated but Bak, Bcl-xl and Bcl-2 were unchanged. Fas-R, Fas-L and Bax were upregulated after tumour regression had taken place. These data show that Leydig cell tumours possess many of the apoptosis related gene products and can die by apoptosis, however, regulation is clearly different in differentiated and mitotic Leydig cells.

Prostate cancer (Pca) is the most prevalent cancer diagnosed and the second leading cause of cancer-related deaths among men in the Poland. The present study was designed to analyse the relationship between expression of Bcl-2 protein in prostate cancer (PCa) following radical prostatectomy to chosen anatomoclinical and morfological parameters of the tumours. Tumours from 28 patients were assessed by immunohistochemistry. Tissue sections were fixed in 10% buffered formaldehyde solution, embedded in paraffin and stained immunohistochemically with the anti-human Bcl-2 antibody (Dako/Clon124). The immunolocalization of Bcl-2 was performed using the Labelled Streptavidin Biotin (LSAB) method. No correlation was found between Bcl-2 protein expression and pT stage, lymph node metastases, Gleason score, seminal vesicles invasion, positive or negative resection margins as well as capsular penetration and preoperative PSA serum level.

The present study was conducted to investigate the possible protective effects of lycopene (LP) and ellagic acid (EA) on aroclor (AR) 1254-induced testicular and spermatozoal toxicity associated with the oxidative stress and apoptosis in male rats. The control group was treated with placebo. LP (10 mg/kg/every other day), EA (2 mg/kg/every other day) and AR (2 mg/kg/day) groups were given alone LP, EA and AR respectively. One of the last two groups received AR + LP, and the other treated with AR + EA. Body and reproductive organ weights, epididymal sperm characteristics, testicular tissue lipid peroxidation levels, antioxidant enzyme activities, histopathological changes and apoptosis via Bax and Bcl-2 genes were investigated. AR administration caused statistically significant decreases in body-weight, epididymal sperm concentration, testicular superoxide dismutase activity, diameters of seminiferous tubules, germinal cell layer thickness and Johnsen's testicular score, and increases in relative weights of testis, epidydimis and seminal vesicles, rates of abnormal sperm and apoptotic cell expression along with degeneration, desquamation and disorganization in spermatogenic cells, and interstitial oedema and congestion in testicular tissue. LP and EA treatments to AR-treated rats markedly decreased abnormal sperm rates, testicular thiobarbituric acid reactive substances level, and increased the glutathione (GSH) level, GSH-peroxidase, catalase activities and epidiymal sperm concentration as compared with the alone AR group. Additionally, the AR-induced histopathological damages were totally or partially recovered by LP or EA administrations respectively. AR damages the testicular tissue and spermatozoa by impairing the oxidant/antioxidant balance and by increasing the apoptotic spermatogenic cell rates. However, both LP and EA have modulator effects on AR-induced reproductive dysfunction in male rats.

In this study, it was aimed to investigate the possible protective effects of gilaburu (Viburnum opulus L., Glb) fruit extract on testis and sperm damages induced by docetaxel (DTX) and paclitaxel (PTX) chemotherapeutics in rats. Sixty adult male Wistar albino rats were divided into 6 equal groups, 10 animals each. The groups were created as control (weekly i.p. saline injection), Glb (weekly i.p. saline injection and daily oral 100mg/kg Glb), DTX (weekly i.p. injection of 5mg/kg DTX), PTX (weekly i.p. injection of 4mg/kg PTX), DTX+Glb (weekly i.p. injection of 5mg/kg DTX and daily oral 100mg/kg Glb) and PTX+Glb (weekly i.p. injection of 4mg/kg PTX and daily oral 100mg/kg Glb). Following 10-week chronic application, spermatological, biochemical, histopathological, cytopathological and immunohistochemical examinations were performed. DTX and PTX caused significant decreases in absolute and relative weights of all reproductive organs, testosterone level, sperm motility, concentration, Bcl-2 anti-apoptotic immunopositive cell scores of testes and spermatozoa as well as catalase activity in epididymal tissue, superoxide dismutase and glutathione peroxidase activities of testicular and epididymal tissues when compared with the control group. Both drugs also significantly increased abnormal sperm rate, testicular and epididymal malondialdehyde level, Bax pro-apoptotic immunopositive cell scores of testes and spermatozoa, and caused remarkable testicular histological and cytological damages. However, Glb consumption mitigated the PTX-induced decreases in absolute weights of epididimis, seminal vesicles, ventral prostate and both taxanes-induced disturbances in sperm characteristics, imbalances in oxidant/antioxidant system, increments in germ cell apoptosis and testicular histo-and cyto-pathological damages. It was concluded that long-term Glb consumption alleviates the taxanes-induced damages in reproductive system of male rats.

The muskrat (Ondatra zibethicus L.) is a seasonal breeding animal. Adult male muskrat has a pair of scent glands between the muscle and skin above the urogenital tract, which secrete muskrat musk during the breeding stage. Compared with other rodents, the muskrat prostates and seminal vesicles are very well developed throughout the breeding stage. Studies have shown that intraperitoneal injection of muskrat musk can readily promote the development of the prostates and seminal vesicles in the male mice, suggesting a tight correlation between musk secretion and the development of the prostates and seminal vesicles in the muskrat. In this study, RNA-seq technology was used to analyze the gene expression patterns were further investigated by gene ontology (GO) analysis and KEGG pathway analysis. The results showed that 1629 genes were significantly and differentially expressed and were involved in a variety of signal transductions and energy metabolism pathways. The differential expression patterns of the OBP2 gene, and members of the Bcl-2 family and tumor necrosis factor receptor superfamily suggest that the prostate development in muskrat is controlled by a variety of molecular mechanisms; and the musk secreted by the muskrat scent glands might participate in the regulation of such developmental processes.

In epididymis, cimetidine induces androgenic failure due to reduced sex hormone-binding globulin stromal levels and blockade of androgen receptor (AR) nuclear import. UCHL1, a hydrolase of ubiquitin-proteasome system (UPS), seems to play a role in autophagy and apoptotic pathway. However, the role of UPS and autophagy in epididymis has not been clarified. We evaluated UCHL1 and autophagy in epididymal cauda epithelium under androgenic deficiency induced by cimetidine, focusing on the interplay among these processes and apoptosis. The integrity of epididymal muscular layer was also evaluated. Male rats received cimetidine (CMTG) or saline (CG). Seminal vesicles were weighed, the expression of androgen-responsive genes Crisp1 and connexin 43 (Cx43) in cauda epididymis was evaluated, and cauda fragments were processed for light and transmission electron microscopy. The epithelium height and muscular thickness were measured. TUNEL, immunohistochemistry for caspase-3 and Cx43, and immunofluorescence for AR, Bcl-2, UCHL1, MAP LC3A, and p62/SQSTM1 (autophagic markers) were performed. Bcl-2, UCHL1, and Cx43 were detected by Western blot. In CMTG, the reduction in seminal vesicles weight accompanied by downregulation of Crisp1 and Cx43 confirmed epididymal androgenic failure. These results were associated with muscular atrophy, apoptosis and weak Cx43 and AR immunoexpression, supporting the androgenic dependence of muscular integrity. The high UCHL1 levels and reduction in Bcl-2 reinforce UCHL1 role in epithelial cells death. The intense immunoexpression of LC3A and p62/SQSTM1 indicates autophagic disturb, which in association with high UCHL1 levels, points to a role of UPS and autophagy in the regulation of epididymal epithelial cells viability under androgenic control.
