The interaction of arginine vasotocin (AVT) and norepinephrine (NE) upon pineal gland indoleamine synthesis was investigated. Rat pineal glands were incubated for 10 h in Krebs--Ringer bicarbonate plus 2 mg/ml glucose, 1 mg/ml bovine serum albumin, [14C]tryptophan, NE (10(-6) M), and log doses of AVT ranging from 100 ng to 10 microgram. Incubation media were extracted for [14C]serotonin while the other [14C]indoleamines, melatonin, hydroxyindoleacetic acid (HIAA), methoxyindoleacetic acid (MIAA), N-acetylserotonin (NAS), hydroxytryptophol (HTOL), and methoxytryptophol (MTOH) were separated by thin-layer chromatography. Serotonin metabolism was decreased by 0.1 microgram AVT and NAS decreased by 1.0 microgram AVT. Melatonin synthesis was decreased by both 0.1 and 1.0 microgram AVT. AVT also decreased the conversion of [14C]serotonin to MIAA and to HTOL. The data indicates that AVT decreased NE-stimulated pineal indoleamine synthesis in vitro and further suggests that AVT may participate in the intracellular regulation of melatonin synthesis.

Interaction of bonive serum albumin with copolymers 2-methyl-5-vinylpyridine, acrylic and methacrylic acids in different proportions in aqueous solutions was studied. It was shown, that in a wide range of ration of the components soluble cooperative complexes were formed. Ability of amphotheric copolymers to form complexes with bovine serum albumin depend both on the composition of the copolymers and on the pH of the medium. The non-polar interactions of the protein globules with hydrocarbonic 2-methyl-5-vinylpyridine chain fragments of the polyampholyte play an important role in the association of similarly charged particals. It was shown that besides this effect of chain fragments of 2-methyl-5-vinylpyridine a significant contribution in the association is made by non-polar interactions, created by methyl groups in copolymer 2-methyl-5-vinylpyridine and methacrylic acid. The association mechanism and structure of the formed complexes was studied. It was shown that complex formation is accompanied by non-equal distribution of the chain copolymer between the protein globules and appearance of asymmetric particles of the complex. A correlation between the ability of polyampholytes to bind protein and their physiological action was found.

1. In 18 patients with ischaemic heart disease left ventricular ejection fraction, measured by two different nuclear angiographic methods, has been compared with ejection fraction measured by single-plane contrast angiography. 2. The first nuclear angiographic technique involves detection of variation in the radio-activity from the left ventricle during the initial passage of a bolus of 99Tcm-labelled human serum albumin injected intravenously; the second is our own modification of a "gated" method, which accumulates the radioactivity detected during the continuing recirculation of the plasma bound radioisotope, so presenting an "averaged" ventricular volume curve. 3. Ejection fraction, measured by the "bolus" method, is lower than that measured either by contrast ventriculography or by the "gated" method. This may be due to a damping effect. 4. Ejection fraction measured by the "gated" method is well correlated with that measured by contrast ventriculography (r = 0-89). 5. Our modification of the "gated" method, which presents the changes in ventricular volume throughout the cardiac cycle, without needing computer facilities, is a useful non-invasive means for assessment of left ventricular function.

A differential method is described for measuring dielectric constants and losses in aqueous protein solutions at millimetrerange wavelengths. Employment of the method allows to improve the accuracy of determining the degree of hydratation. A method has also been suggested for taking into account the contribution of ions to the dielectric constant of solutions. The differential method was used to study hydratation of nine globular proteins. The data obtained are compared with the corresponding values provided by other experimental techniques and with theoretical predictions based on some models of hydratation. Good agreement is obtained with results provided by the isopiestic and NMR techniques. The discrepancy shown for hemoglobin is discussed in the paper. As has been shown, the dielectric method registers a monomolecular surface layer of water only. With pH varying between 4.0 and 3.2, a significant increase is observed in the hydratation of serum albumin. Presumably, this effect is connected with a N--F conformational transition.

1. Distribution of zinc between the metalloprotein alpha 2-macroglobulin and albumin was determined in samples of serum obtained from twenty-three women in their third trimester of pregnancy and eighteen women who were not pregnant. 2. The decrease observed in total serum zinc in the group of pregnant women could be accounted for in large part by a decreased concentration of albumin-bound zinc. 3. The concentration of serum albumin was lower in the pregnant women, thus hypoalbuminaemia, which is often invoked to explain hypozincaemia in pathological situations, may in part account for hypozincaemia in pregnancy. 4. The affinity of albumin for zinc added to serum from pregnant women was less than that of albumin from non-pregnant women. This was determined by competition experiments between albumin and glycine for zinc. Decreased affinity of serum albumin for zinc may also contribute to the hypozincaemia associated with pregnancy.

The presence of histamine receptors on lymphocyte membranes was investigated using conjugates of histamine and macromolecules tritiated or iodinated with I-125. Histamine-RSA conjugate binds to lymphocytes and causes patching and capping of the bound conjugate. It was found, however, that free histamine did not inhibit the binding of histamine-rabbit serum albumin to mouse lymphocytes, nor did His-RSA interfere with the binding of free histamine. In addition conjugates between RSA and other small molecules, such as ethylamine, ethanolamine, tyramine and glycine, were found to bind to the same sites on lymphocyte membrane as did His-RSA. Ethylamine-RSA like His-RSA when coupled to Sepharose, was capable of removing antibody producing cells from spleen cells of mice immunized against sheep red blood cells. In addition, when spleen cells from such immunized mice were passed through ethylamine or histamine-RSA-Sepharose and the unbound cells were subsequently injected into X-irradiated mice, a 1.8 fold increase in the immunological response was noted. We conclude that the selective binding to lymphocytes of the various ligand-macromolecular conjugates may be due to some general properties of the cell membrane and not to any specific receptors. Nevertheless, these conjugates can be used as a tool to remove selectively antibody producing cells as well as some regulatory cells.

The antibody response of genetically inbred rats to poly(Glu52Lys33Tyr15) is controlled by a complex polygenic system which includes at least two autosomal genes and a sex influence, which may also be genetically determined. The genetic control of the quantity, binding constants, and specificity of the antibody formed linked to the major histocompatibility locus. Factors other than the major genetic ones and the sex influence also affect the quantity of antibody formed, since animals of the same genotype can make significantly different amounts of antibody, depending upon the crosses by which they acquire the major histocompatibility alleles. After immunization with poly(Glu52Lys33Tyr15) the low responders make fewer antibody-producing cells, are not capable of mounting a delayed hypersensitivity reaction to the polypeptide and appear to be deficient in their ability to produce the specific IgM antibody. Immunization of the low responders with antigen aggregated with methylated bovine serum albumin enhances the quantity of antibody formed, increases the binding constants and crossreactivity of the antibody and enhances the delayed hypersensitivity response. In contrast to the findings with the L-amino acid polypeptide, there does not appear to be any genetic control over the antibody response to the D-amino acid enantiomorph poly(DGlu52DLys33DTyr15), which is minimal in all strains.

In the system with constant content of the chlorophyll a and increasing amounts of human serum albumin, dependence of pigment incorporation into the complex upon interaction of its aqueous associates with protein solutions was studied by applying the gel filtration on Sephadex G-75 and by measuring light scattering and rate of sensitized photoreduction of the methyl red by ascorbic-acid. The curves were obtained after extraction of the chlorophyll by acetone from dry pigment-protein films formed after desiccation of the aqueous systems. Sigmoid character of the above dependences, their linearization in Hill's coordinates and the value of cooperativity coefficient close to 2 testifies in favour of the cooperative character of the complex formation, two pigment molecules reacting with a single protein molecule. Measurement of adsorption isotherms and their treatment with use of the Brunauer-Emmett-Teller theory of polymolecular adsorption make it possible to evaluate the maximum molar ratio of the pigment to the protein in the complex (close to 2). The pigment-pigment interaction suggests that the chlorophyll molecules adsorbed on the protein are in the state of loosely packed dimers. Deaggregation of aqueus pigment associates by the protein in the course of complex formation results in a considerable increase of the protosensitizing chlorophyll activity.

The binding of [3H] delta 6-testosterone photoaffinity-labelled rat androgen-binding protein (rABP) has been studied in an enriched fraction of plasma membranes of epithelial epididymal cells in immature (15 days) and adult rats (40 days). The binding was maximal in less than 30 min and more rapid at 4 degrees C than at 34 degrees C. It was calcium and pH dependent. Scatchard plots of the binding data gave curvilinear plots with two types of binding sites corresponding to a K(ass1) of 18.2 nM-1 and K(ass2) of 1.6 nM-1 (2.2 x 10(11) sites/mg protein and 5.4 x 10(11) sites/mg protein, respectively). In adult rats, only one type of binding site was found, with a K(ass) of 3.7 nM-1 (4.5 x 10(11) sites/mg protein). The number of receptors was 5-fold lower in the cauda than in the caput of the epididymis. The pretreatment of the isolated intact cells with streptozotocin induced a 45% reduction of the binding. Only unlabelled rABP and hSBP (human sex steroid-binding protein) but not other proteins (lactotransferrin, serotransferrin, asialofetuine, fetuine and bovine serum albumin) competed with the labelled ligand to bind plasma membranes. The membrane fraction was solubilized by triton X-100. Its incubation with labelled rABP and hSBP provoked the elution of the tracer as an aggregate into the void volume fraction of superose 6B mini-gel filtration columns. Structural homology between hSBP and rABP could be responsible for the common behaviour of the steroid-carrier molecules for the ABP receptor of rat epididymal epithelial cells.

Several early (Phase I) and late (Phase II) estrogenic effects of 9-ene-tetrahydrocannabinol (THC) were examined in the adult mouse uterus. An injection of THC (2.5 or 10 mg/kg body wt) in ovariectomized mice neither stimulated uterine water imbibition or accumulation of [125I]bovine serum albumin (Phase I responses) at 6 h, nor antagonized these Phase I responses elicited by estradiol-17 beta (E2). With respect to Phase II responses, although single injections of THC (2.5, 5.0 and 10 mg/kg body wt) alone were ineffective in influencing uterine weight at 24 h or incorporation of [3H]thymidine at 18 h, this drug interfered with these responses elicited by E2 in a dose-dependent manner. In contrast, an injection of THC in progesterone (P4)-primed ovariectomized mice modestly enhanced (61%) uterine incorporation of [3H]thymidine. However, E2-stimulated uterine thymidine incorporation in P4-primed ovariectomized mice was antagonized by THC treatment. Effects of THC on blastocyst implantation were examined. Single or multiple injections of various doses of THC neither induced implantation in P4-primed delayed implanting mice, nor interfered with E2-induced implantation. Furthermore, daily injections of THC (10 mg/kg body wt) during the peri-implantation period had no apparent adverse effects on implantation, or on experimentally induced decidualization (deciduomata). The data suggest that THC is neither pro- nor antiestrogenic with respect to Phase I responses. However as regards Phase II responses, THC is modestly pro-estrogenic in the P4-treated uterus, but is anti-estrogenic in the presence of E2. These estrogen agonistic/antagonistic effects of THC on uterine Phase II responses do not adversely affect the process of implantation and decidualization.
