The murine B-cell hybridoma B9 requires interleukin-6 (IL-6) for its survival and proliferation in vitro. We show here that withdrawal of IL-6 from B9 cultures results in programmed death, concomitant with arrest of the cells in the G1 phase of the cell cycle. Unlike several other systems that undergo programmed cell death, no induction of transcripts corresponding to the testosterone-repressed message-2 or transglutaminase genes is observed during this process. Upon readdition of IL-6 to G1-arrested B9 cells, viability is maintained and entry into S phase occurs after a lag period of 10 to 12 hr. Northern blot analysis showed that the immediate-early mRNAs normally induced shortly after growth factor stimulation in quiescent fibroblasts (c-fos, c-jun, Egr-1, c-myc, JE, and KC), and other growth-related genes (2F1, c-Ha-ras, and
p53
), are either not induced or remain unchanged during G1 to S phase progression. A correlation was found, however, between the temporal pattern of expression of several G1/S phase genes (dihydrofolate reductase, thymidine kinase, transferrin receptor, and
histone H3
) and DNA synthesis. These results demonstrate that IL-6-induced viability and growth of hybridoma (and, presumably, plasmacytoma) cells is mediated via novel signal transduction pathways.

A definite association between the transcription of N-myc oncogene and proliferation-related genes,

, was found in a set of 12 primary untreated neuroblastomas and a metastasis of one of these at relapse. Molecular analysis allowed us to discriminate between actually proliferating and non-proliferating tumors, and suggested a link between N-myc and proliferation. Flow cytometric analysis of DNA distribution was less reliable for assessing tumor proliferative activity. Our data also seem to indicate a down-regulation of c-myc by N-myc in human neuroblastoma.

Mouse F9 teratocarcinoma cell lines can be induced to differentiate into either parietal endoderm or embryoid bodies which contain visceral endoderm-like cells. The nature of the early molecular events involved in these two differentiation pathways has not yet been fully elucidated. Moreover, since the process of differentiation is often accompanied by changes in cell growth, it is often difficult to determine which of the events that do occur during the early stages of differentiation are a direct result of the process of differentiation and which events are indirect results that occur as a consequence of altered cell growth. In the experiments reported here we have attempted to distinguish between these two possibilities by examining the patterns of expression of a representative group of growth-associated genes (i.e., c-myc,
p53
, and
histone H3
) when F9 cell aggregates are induced to differentiate into embryoid bodies containing visceral endoderm. By analysis of the patterns of growth-associated gene expression in both retinoic acid treated and nontreated F9 cell aggregates, we were able to classify early events as differentiation-specific events (events which occurred only following retinoic acid treatment of aggregates) or nondifferentiation-specific events caused by reduction in cell growth (events which occurred even when aggregates were not treated with retinoic acid). Our results show that F9 cells differentiated into embryoid bodies containing visceral endoderm-like cell exhibit an early reduction in both growth and c-myc mRNAs which is neither retinoic acid-specific nor differentiation-specific. However, following this initial response to aggregation, constant levels of c-myc mRNA are maintained despite continued reduction in growth. Thus, it appears that alteration in c-myc expression is a differentiation-specific event along the pathway to formation of visceral endoderm. Interestingly, however, the nature and time course of this alteration in c-myc expression in F9 cells' differentiation into visceral endoderm is different from that observed in F9 cells differentiated into parietal endoderm.

Chronic and blastic phase chronic myelogenous leukaemia cells have been studied by northern and Southern blot analysis. DNA from matched chronic and blastic phase cells obtained from the same patient demonstrated that the rearrangement site within the breakpoint cluster region did not change at the time of blastic crisis. A search for a mutation in a controlling region of the first exon of c-myc also failed to demonstrate any new abnormality at the time of blastic crisis. , MRS) were detected, these differences could be ascribed to differences in the proportions of immature cells during the chronic and blastic phases. The data suggested that the c-myc transcripts in blastic phase cells and in immature chronic phase cells differ in that the latter contain some c-myc transcripts that are not polyadenylated. Differences in c-myc transcript half-life could contribute to the differences in the behaviour of chronic phase and blastic phase immature cells.

We have studied a panel of 10 genes and cDNA sequences that are expressed in a cell cycle-dependent manner in different types of cells from different species and that are inducible by different mitogens. These include five sequences (c-myc, 4F1, 2F1, 2A9, and KC-1) that are preferentially expressed in the early part of the G1 phase, three genes (ornithine decarboxylase,

, and c-rasHa) preferentially expressed in middle or late G1, and two genes (thymidine kinase and

) preferentially expressed in the S phase of the cell cycle. We have studied the expression of these genes in nonpermissive (tsAF8) and semipermissive (Swiss 3T3) cells infected with adenovirus type 2. Under the conditions of these experiments, adenovirus type 2 infection stimulates cellular DNA synthesis in both tsAF8 and 3T3 cells. However, four of the five early G1 genes (c-myc, 4F1, KC-1, and 2A9) and one of the late G1 genes (c-ras) are not induced by adenovirus infection, although they are strongly induced by serum. Our results indicate that: (a) MPO gene expression (steady state mRNA levels) is strictly confined to the first stages of myeloid differentiation, reaching its peak at the promyelocyte stage and becoming undetectable in mature granulocytes and monocytes; (b) cells devoid of any detectable MPO enzymatic activity such as leukemic basophils have a high content of MPO mRNA; and (c) MPO gene expression is not related to the growth activity of the cell population. Finally, our results show that the pattern of expression of growth-regulated genes in the neoplastic myeloid disorders AML, CML, and CML blast crisis is remarkably different.

C3H/Km flora-defined mice were used to investigate the effect of exposure to pulsing electromagnetic field (PEMF) after total body x-ray irradiation. Prolonged exposure to PEMF had no effect on normal nonirradiated mice. When mice irradiated with different doses of x-ray (8.5 Gy, 6.8 Gy, and 6.3 Gy) were exposed to PEMF 24 h a day, we observed a more rapid decline in white blood cells (WBC) in the peripheral blood of mice exposed to PEMF at all the x-ray dosages used. No effect of exposure to PEMF was observed on the survival of the mice irradiated with 6.3 Gy and 8.5 Gy; in mice irradiated with 6.8 Gy, 2 out of 12 survived when exposed to PEMF as compared to 10 out of 12 control mice that were irradiated only. At day 4 after irradiation autoradiographic studies performed on bone marrow and spleen of 8.5-Gy-irradiated mice showed no difference between controls and mice exposed to PEMF, whereas on 6.8-Gy mice the bone marrow labeling index was lower in mice exposed to PEMF. In mice irradiated to 6.3 Gy we observed that the recovery of WBC in the peripheral blood was slowed in mice exposed to PEMF and their body weight was significantly lower than in control mice that were irradiated only. The spleen and bone marrow of the mice irradiated to 6.3 Gy and sacrificed at days 4, 14, 20, and 25 after irradiation were analyzed by autoradiography to evaluate the labeling index. Half of the spleens from mice sacrificed at day 25 after irradiation were used to evaluate the RNA content. Autoradiography showed that in the spleen and bone marrow of control mice, there were more cells labeled with [3H]thymidine at days 4 and 14 and less at days 20 and 25 after irradiation in comparison with mice irradiated and exposed to PEMF. RNAs extracted from the spleens at day 25 after irradiation showed a slight increase in cycling cells among spleens of mice exposed to PEMF. We suggest that the exposure to PEMF immediately after x-ray irradiation results in increased damage.

gene, was compared in bone marrow cells from multiple myeloma patients and normal controls by measuring the amount of specific RNA by Northern blot analysis. Four samples contained at least 72% of myeloma cells, one sample 43%, and one 11%. Expression of the protooncogenes was similar in normal and myeloma bone marrow cells, whereas that of
histone H3
gene was significantly reduced (between 10 and 15 times) in samples containing at least 43% of malignant plasma cells and not detectable in those containing more than 72% of neoplastic cells. Protooncogene levels of expression were compared to those of the H3 gene to distinguish the increased expression of a growth-regulated gene due to a true deregulation from overexpression reflecting solely an increase in the fraction of cycling cells. histone H3
were markedly increased in multiple myeloma cells; the highest ratios were found in the patients with the highest number of malignant plasma cells. These results suggest that the expression of three growth-regulated oncogenes (c-myc, c-myb,
