The polymerase chain reaction (PCR) was used to show that basic fibroblast growth factor (FGFb) and epidermal growth factor (EGF) receptor messenger ribonucleic acids (RNA) are produced in human lacrimal tissue. Transforming growth factor beta-1 (TGF beta-1) messenger RNA was not detected. These results and the previous identification of EGF in the lacrimal gland suggest that EGF could have autocrine or paracrine functions in lacrimal tissue. FGFb could also serve autocrine or paracrine functions in lacrimal gland physiology. It is also plausible that FGFb is released by the lacrimal gland into the tears and that the growth factor may have regulatory effects on the cells of the ocular surface.

Expression of epidermal growth factor (EGF) receptor was examined in 32 cases of pleomorphic adenoma of lacrimal gland by means of an immunohistochemical method. While normal lacrimal glands were all negative for the antigen, EGF receptor was positive in 10 pleomorphic adenomas and the positive staining was mainly limited in the tumor cells in trabecular or duct-like arrangement or squamous metaplastic epithelium. These data suggest that the expression of EGF receptor be significantly higher in pleomorphic adenoma than in normal lacrimal gland, and also suggest that EGF receptor be expressed in the neoplastic cells which are considered to be of duct origin.

Using a polyclonal antibody to the c-erbB-2 oncogene product (ErbB-2 protein), an immunohistochemical study on the expression of ErbB-2 protein in lacrimal gland tumors was performed. The expression of ErbB-2 protein was observed in 4 (33.3%) of 12 cases of pleomorphic adenoma, but was not in two cases of adenoid cystic carcinoma and one case of normal lacrimal gland. The immunohistochemical reaction was localized in the cell membrane and cytoplasm, and was more intense in the former. There were strongly stained cells in the epithelium of ductal cell lineage and in the solid area, and some of the cells in the myxoid/chondroid areas were weakly stained. The relation ErbB-2 protein with the malignant lacrimal gland tumors in general is not clear, but pleomorphic adenomas of lacrimal gland could have some relation with this protein.

This study was designed to demonstrate the presence of the epidermal growth factor (EGF) receptor in the rat exorbital lacrimal gland. EGF receptor gene transcription was demonstrated 1) by reverse transcription-polymerase chain reaction analysis of lacrimal gland and acinar cells from RNA with a set of specific primers deduced from the rat EGF receptor sequence and 2) by Northern blot analysis of rat lacrimal gland mRNA. Lacrimal acinar cell preparations contain a low but detectable amount of specific 125I-EGF binding sites and efficiently internalize the ligand on binding at 37 degrees C. A sheep polyclonal antibody, directed against the human EGF receptor, detects a protein of 170 kDa by Western blot analysis of membrane proteins of the whole gland. This protein can be immunoprecipitated by the same antibody from whole gland membrane proteins as well as from solubilized acinar cells. Incubations of acinar cells in the presence of EGF results in an increased content of tyrosine-phosphorylated residues in immunoprecipitated 170-kDa protein. Taken together, these results demonstrate for the first time both EGF receptor gene transcription and protein expression in a lacrimal tissue, i.e., the rat exorbital lacrimal gland. These results also suggest a specific cellular location of the EGF receptor in a cell population contained in acinar cell preparations.
