Pinus mugo needles were sampled at different altitudes (1420, 1590 and 1920 m a.s.l.) to analyse levels of oxidative stress and changes in maximum photochemical efficiency of PSII.
Polyacrylamide
gel electrophoresis demonstrated that almost all superoxide dismutase activity represented Cu/Zn superoxide dismutase, and only 4-6% represents
Mn superoxide dismutase
. In extracts from plants sampled at 1590 and 1920 m a.s.l., lower activity of Cu/Zn superoxide dismutase was found. Comparing these data with immunoblots, it can be concluded that the differences in superoxide dismutase activity was related to protein amount. In needles from higher altitudes, a decrease in catalase activity was detected, as opposed to the protein amount, which was higher in needles from the higher stands. Considering the decrease in catalase and Cu/Zn superoxide dismutase activities in needles collected at 1590 and 1920 m a.s.l., we suggest that higher levels of oxidative stress may induce changes in photochemical efficiency of PSII.

The study on the effects of different concentration (0, 10, 50, 100, 200 and 400 micromol x L(-1)) cadmium (Cd) on the antioxidative system in Ganoderma lucidum mycelia indicated that with increasing concentration of Cd, the fresh mass and the proline, total polysaccharides, and reduced polysaccharides contents of G. lucidum mycelia decreased, but non-protein thiol (NPT) content increased. At 400 micromol x L(-1) of Cd, the NPT content increased dramatically, being 5.7 times higher than control. Within the range of test Cd concentrations, the activities of CAT, GR and POD increased first and decreased then, with the peak at 100 micromol x L(-1) of Cd, while the activities of LOX and SOD increased with increasing Cd concentration, with the maximum at 400 micromol x L(-1) of Cd.
Polyacrylamide
gel electrophoresis analysis revealed that 100-400 micromol x L(-1) of Cd induced two additional isozymes bands of
Mn-SOD
, 10-200 micromol x L(-1) of Cd increased the intensity of constitutive isozymes of CAT, POD, SOD and LOX, while 400 micromol x L(-1) of Cd decreased the intensity of isozymes of POD significantly.

Iron superoxide dismutase (Fe-SOD; EC 1.15.1.1) was isolated from the nitrogen-fixing cyanobacterium Anabaena cylindrica Lemm.
Polyacrylamide
gel electrophoresis separated the purified protein into three closely running, enzymatically active bands. The molecular weight of the enzyme was estimated by gel filtration to be about 40 kDa. Polyclonal antibodies were produced by immunization of rabbits with the isolated enzyme, and were purified on a column of protein A-Sepharose. The Fe-SOD antibody reacted with the purified Fe-SOD and also specifically recognized the protein in extracts of A. cylindrica. In the extracts, anti-Fe-SOD did not cross-react with
Mn-SOD
, an enzyme which belongs to an SOD class displaying high homology of primary and three-dimensional structure with respect to Fe-SOD. Iron superoxide dismutase was localized in heterocysts by immunogold labeling and transmission electron microscopy. These results are the first in-situ evidence for the presence of SOD in the cells specialized for nitrogenase activity.

In heterotrophically grown Scenedesmus obliquus, the specific activity of superoxide dismutase (SOD; EC 1.15.1.1) declined when glucose was abundant, increased as it was depleated, and remained steady at a high level when it was absent. Transition to autotrophic growth produced only a small (20% over 5 d) increase in specific activity above the values obtained in dark-grown cells after glucose and starch-reserve depletion. This small, but consistent, increase did, however, parallel a similar increase in photosynthetic capacity.
Polyacrylamide
-gel electrophoresis showed the existence of nine isoenzymes of SOD. The three major and one of the minor isoenzymes were present in all extracts while three minor isoenzymes were found only in autotrophically grown cells and two only in heterotrophically grown cells. Characterization studies indicated that two of the major isoenzymes are dimeric FeSODs the other is a tetrameric
MnSOD
, and of the minor isoenzymes, two are dimeric FeSODs and four are dimeric MnSODs.
