The presence of peroxisomes and their enzymic content were investigated and compared in healthy and neoplastic human colon epithelial cells using cytochemical studies at the ultrastructural level as well as biochemical analyses.
Catalase
-positive organelles were found to be more numerous in normal than in colonic neoplastic cells. Biochemical assays revealed that no D-aminoacid oxidase or L-alpha-hydroxyacid oxidase activity was detected in normal or tumor tissues. The specific activities of catalase, fatty-acyl CoA oxidase and enoyl-CoA hydratase/3 hydroxyacyl-CoA dehydrogenase (the so-called
peroxisomal bifunctional enzyme
of the beta-oxidation system) were found to be diminished in carcinoma cells compared with the control tissue. The fall in catalase activity correlated well with tumor stage according to Dukes, suggesting that this peroxisomal enzyme could be used as a potential prognostic marker.

We have investigated the hepatic effect of ciprofibrate, a potent peroxisomal proliferator, in 9 strains of mice to ascertain whether all strains show similar peroxisome proliferation or if there are any that are resistant to the induction of peroxisome proliferation. Dietary feeding of ciprofibrate at 2 concentrations (0.0125% or 0.025% w/w) for 2 weeks resulted in a significant increase in liver weight (170 to 200%) and a 7- to 11-fold increase in volume density of peroxisomes.
Catalase
and peroxisomal beta-oxidation enzymes increased by 1.7- to 2.7- and 1.9- to 9.3-fold, respectively, over the controls. SDS-polyacrylamide slab gel electrophoresis of post-nuclear fractions of livers showed a marked increase in 80,000-mol. wt. polypeptide. Immunocytochemical studies, as expected, revealed higher levels of PBE. Ciprofibrate treatment also induced hepatic DNA synthesis in all strains as determined by [3H]thymidine incorporation and autoradiography. When the 9 strains were ranked for each parameter, CBA/Ca was the least responsive mouse strain and the B6C3F1 was the most responsive. However, the results of this study indicate that there is no significant interstrain difference in rankings across strains to ciprofibrate-induced hepatic pleiotropic response.

activity was enhanced in HepG2 cells treated with 3 mmol/l clofibric acid to 135% of control and the mRNA value to 2.6 fold, whereas in cetaben treated cells the enhancement (up to 119% of control) was less pronounced. In MH1C1 cells catalase activity was not changed by any of the drugs. The activity of the
peroxisomal bifunctional enzyme
was not affected in HepG2 cells by clofibric acid and cetaben, whereas the mRNA level was elevated to 2.3 fold by 10 micromol/l cetaben. At high concentrations of cetaben all enzyme activities were decreased in both cell lines due to its high cytotoxicity. Our data show that, due to the differences in the genomic organisation, the regulation of the enzyme activities is different in human and rat, but the results from the human and rat hepatoblastoma cells correlate with the findings in whole man and rat, so that a human in vitro system is more suitable for pharmacological tests. These results suggest that the human hepatoma cell line HepG2 may be a useful model system for studies of the influence of hypolipidaemics on the peroxisomal enzyme system.
