Previously we have reported in vitro evidence suggesting that that H2O2 may support wound healing by inducing
VEGF
expression in human keratinocytes (C. K. Sen et al., 2002, J. Biol. Chem.277, 33284-33290). Here, we test the significance of H2O2 in regulating wound healing in vivo. Using the Hunt-Schilling cylinder approach we present the first evidence that the wound site contains micromolar concentrations of H2O2. At the wound site, low concentrations of H2O2 supported the healing process, especially in p47(phox)- and MCP-1-deficient mice in which endogenous H2O2 generation is impaired. Higher doses of H2O2 adversely influenced healing. At low concentrations, H2O2 facilitated wound angiogenesis in vivo. H2O2 induced FAK phosphorylation both in wound-edge tissue in vivo and in human dermal microvascular endothelial cells. H2O2 induced site-specific (Tyr-925 and Tyr-861) phosphorylation of FAK. Other sites, including the Tyr-397 autophosphorylation site, were insensitive to H2O2. Adenoviral gene delivery of catalase impaired wound angiogenesis and closure.
Catalase
overexpression slowed tissue remodeling as evidenced by a more incomplete narrowing of the hyperproliferative epithelium region and incomplete eschar formation. Taken together, this work presents the first in vivo evidence indicating that strategies to influence the redox environment of the wound site may have a bearing on healing outcomes.

Diabetes the global epidemic is rapidly increasing at an alarming rate. Developing countries like India harbor the majority of diabetic people and by the year 2030 AD India will have the largest number of diabetic patients. Diabetic foot is one of the common diabetic complications found in India. Both aerobic and anaerobic pathogens form the etiology for diabetic foot infection. Members of the Enterobacteriaceae family were the most prominent among the aerobes while members of the Genus Peptostreptococcus and Clostridium were most prominent among the anaerobes. Ulcers infected with anaerobic pathogens showed a longer healing time than ulcers infected with aerobic pathogens. Oxidative stress is one of the major markers of inflammatory response and oxidative stress markers such as lipid peroxidation, thiobarbituric acid reactive substance (TBARS), Superoxide Dismutase (SOD),
Catalase
, G Peroxidase, G-S Peroxidase and plasma total antioxidant play a major role in the nonhealing of diabetic foot ulcers. Growth factors such as platelet-derived growth factor (PDGF), transforming growth factor (
VEGF
), and epidermal growth factor (EGF) are needed for normal wound repair, while proteases such as matrix metalloproteinase (MMP) and serine proteases found in chronic wounds delay the healing process.

Climatic change induced heat stress causes a formidable challenge for maintaining optimum productivity in goats. The SOD1 gene (superoxide dismutase 1) is one of the genes that regulates the heat tolerance capacity. This study aimed to in silico characterize this gene. Sequence based phylogenetic tree analysis showed the caprine SOD1 gene has close evolutional relationship with that of sheep. STRING database reveals its interaction with SOD2 (Superoxide dismutase [Mn], mitochondrial), ATOX1 (Copper transport protein), RAC1 (Ras-related C3 botulinum toxin substrate 1), GPX2 (Glutathione peroxidase 2), GPX4 (Glutathione peroxidase 4), GPX5, GPX6, CAT (
Catalase
). The KEGG pathway maps gave interaction with eNOS, iNOS,
VEGF
and MAPK in which gene transcription modulates response to heat stress. The three dimensional protein structure was predicted using Modeller, Phyre 2, and Swiss Model. Structure validation was done observing the Ramachandran Plot and Hydrophobicity Plot in Discovery Studio considering amino acid residues in favoured region.
