Increased generation of reactive oxygen species (ROS) leads to oxidative stress in diabetes. Catalase is a highly conserved heme-containing protein that reduces hydrogen peroxide to water and oxygen and is an important factor decreasing cellular injury owing to oxidative stress. Hyperglycemic conditions increase oxidative stress and angiotensinogen gene expression. Angiotensinogen conversion to angiotensin II leads to a furtherance in oxidative stress through increased generation of reactive oxygen species. In this study, we utilized mice transgenically overexpressing rat catalase in a kidney-specific manner to determine the impact on ROS, angiotensinogen and apoptotic gene expression in proximal tubule cells of diabetic animals. Proximal tubules isolated from wild-type and transgenic animals without or with streptozotocin-induced diabetes were incubated in low glucose media in the absence or presence of angiotensin II or in a high-glucose media. Our results show that the overexpression of catalase prevents the stimulation of ROS and angiotensinogen mRNA in tubules owing to elevated glucose or angiotensin II in vitro. Additionally, overexpression of catalase attenuated ROS generation, angiotensinogen and proapoptotic gene expression and apoptosis in the kidneys of diabetic mice in vivo. Our studies point to an important role of ROS in the pathophysiology of diabetic nephropathy.

Blue mussels (Mytilus edulis) were exposed to an extract made of natural cyanobacterial mixture containing toxic cyanobacterium Nodularia spumigena (70-110 microg nodularin l(-1), 24-h exposure followed by 144-h depuration period in clean water). Toxin concentration increased from initial 400 to 1100 mg kg(-1) after 24-h exposure, measured by liquid chromatography/mass spectrometry (LC/MS). Acetylcholinesterase activity (AChE), a biomarker of direct neurotoxic effects, showed inhibition after 12 and 24h exposure but returned to control level during the depuration period. Catalase (CAT) activity, an indicator of oxidative stress, showed significantly elevated levels in exposed mussels but only 72 h after the end of the exposure. No change in the activity of glutathione-S-transferase (GST) involved in conjugation reactions could be observed. A gradual yet incomplete elimination of nodularin (from 1100 to 600 mg kg(-1)) was observed during the depuration period, and the tissue levels were 30% lower in clean water after 24 h. The observed increase in oxidative stress indicated by elevated CAT activity is likely connected to detoxification reactions leading to the production of reactive oxygen species, including an apparent time lag in this specific enzymatic defence response. That no change in GST activity was observed suggests that this enzyme is not significantly involved in the detoxification process of nodularin-containing cyanobacterial extract in M. edulis.

Seasonal variation of antioxidant enzymes activities (superoxide dismutase, catalase, and glutathione peroxidases) and lipid peroxidation (LPO) were studied in the clam Ruditapes decussatus in relation to body burdens of polycyclic aromatic hydrocarbons (PAHs). Clams were sampled in eight sites from the Ria Formosa lagoon. PAH concentrations were seasonally rather than spatially dependent, being higher in summer (August). Antioxidant enzymes activities and LPO levels in the clam digestive gland were also seasonally dependent. Antioxidant enzymes presented distinct seasonal variations: Mit SOD (superoxide dismutase activity measured in the mitochondrial fraction) was induced in the summer and down-regulated in winter and spring, while Cyt SOD activity (measured in the cytosolic fraction) was highest in autumn and lower in the summer. Neither Mit nor Cyt SOD were related to the clam PAH body burden, suggesting that cells are using other antioxidant systems to eliminate oxyradicals. Catalase (CAT), however, was induced in spring and down-regulated in summer, the inverse of the PAH concentrations in clam tissues. CAT induction in spring appears to be related to the excess of oxyradicals arising from the metabolic activity associated with the reproductive cycle. Conversely, the decrease in CAT activity in the summer may be related to the high water temperatures reached in the Ria Formosa (up to 30 degrees C). Glutathione peroxidases (total fraction - T-GPx and dependent on selenium - Se-GPx) presented a similar seasonal pattern, and were negatively related to PAH concentrations, which may indicate a precarious state of the clams, associated with PAH toxicity. Similarly, LPO was also inversely correlated to the PAH concentrations indicating that increases in PAH concentrations were not causing membrane oxidative damage in R. decussatus digestive gland. The results suggest that antioxidant enzymes in R. decussatus digestive gland are strongly affected by seasonal factors stressing the need of other experiments to clarify the PAHs effect on this clam species.

Pomegranate juice (PJ) possesses a high antioxidant activity, which has been related to beneficial health properties. However, in vivo confirmation and characterization of these effects on biological systems are lacking and needed. This study was performed in order to investigate the effect of prolonged PJ ingestion on general oxidation status. For this purpose, mice ingested PJ (or water in control group) during four weeks, after which damage to lipids, proteins and DNA were evaluated as oxidative cell biomarkers. Levels of hepatic glutathione and the activities and expression of enzymes involved in its metabolism were determined. Catalase and SOD activities were quantified as these enzymes have a crucial role in antioxidant defence. Protection against protein and DNA oxidation was found in PJ group. There was also a significant decrease in GSH and GSSG, without change in the GSH/GSSG ratio. All studied enzymatic activities (GPx, GST, GR, SOD and catalase) were found to be decreased by PJ treatment. Additionally, RT-PCR results showed that GST and GS transcription were also decreased in this group. These results are compatible with a protective effect of PJ against systemic oxidative stress in mice.

The present study evaluated the inhibitory effects of zinc on colonic antioxidant defense system and histoarchitecture during 1,2 dimethylhydrazine (DMH) induced colon carcinogenesis in male Spraque Dawley rats. The rats were segregated into four groups viz., normal control, DMH treated, zinc treated, DMH + zinc treated. Colon carcinogenesis was induced through weekly subcutaneous injections of DMH (30 mg/kg body weight) for 8 weeks. Zinc (in the form of zinc sulphate) was supplemented to rats at a dose level of 227 mg/l in drinking water, ad libitum for the entire duration of the study. Increased lipid peroxidation was accompanied by a decrease in reduced glutathione (GSH), glutathione reductase (GR), glutathione-s-transferase (GST), superoxide dismutase (SOD), and catalase. Administration of zinc to DMH treated rats significantly decreased the lipid peroxidation levels with simultaneous enhancement of GSH, GR, GST, SOD, and Catalase. Histopathological studies from DMH treated rats revealed disorganization of colonic histoarchitecture. However, zinc treatment to DMH treated rats greatly restored normalcy in the colonic histoarchitecture, with no apparent signs of abnormality. Energy Dispersive X-Ray Fluorescence (EDXRF) studies revealed a significant decrease in tissue concentrations of zinc in the colon following DMH treatment, which upon zinc supplementation were recovered to near normal levels. In conclusion, the results of this study suggest that zinc has a beneficial effect during the initiation of key events leading to the development of experimentally induced carcinogenesis.

To investigate the effects of a synthetic soybean isoflavone (ISF) on growth performance, meat quality, and antioxidation in male broilers, 1,500 birds that were 42 d old were allotted to 5 treatments with 6 replicates per treatment (50 birds per replicate). Birds were fed diets supplemented with 0, 10, 20, 40, or 80 mg of ISF/kg, respectively, for a period of 3 wk ad libitum. The results showed that dietary supplementations with 10 or 20 mg of ISF/kg increased weight gain by 13.6 and 16.2% (P < 0.01) and elevated feed intake by 7.37% (P < 0.05) and 11.2% (P < 0.01), respectively. Addition of 10 mg of ISF/kg decreased feed:gain by 5.5% (P < 0.05). Supplementation with 40 mg of ISF/kg in the diet slightly increased water-holding capacity by 17.24% (P < 0.1), and the addition of 20 or 40 mg/kg of ISF significantly increased the pH value of meat (P < 0.01), although adding 40 or 80 mg of ISF/kg increased the lightness of meat color (P < 0.05). Malondialdehyde production was slightly reduced in plasma of 20 mg of ISF/kg supplemented chickens (P < 0.1) and significantly decreased in breast muscles of 20, 40, or 80 mg of ISF/kg supplemented chickens (P < 0.01). The addition of 40 or 80 mg of ISF/kg significantly increased total antioxidant capability (P < 0.01) and slightly elevated total superoxide dismutase activity (P < 0.1) in plasma of chickens. The dose of 80 mg of ISF/kg slightly improved catalase activity in plasma (P < 0.06). In breast muscle, treatment of birds with 40 or 80 mg of ISF/kg caused an increase of total superoxide dismutase activity by 25.36% (P < 0.05) or 63.93% (P < 0.01). Catalase activity significantly increased by 70.61% by the supplemental ISF at the 40-mg level (P < 0.05). Also, 10, 20, or 40 mg doses of ISF/kg decreased lactic acid production (P < 0.05). The results of this study indicate that dietary ISF could improve growth performance and meat quality by decreasing lipid peroxidation and improving antioxidative status in male broilers.

Catalase is one of the central enzymes involved in scavenging the high level of reactive oxygen species (ROS) by degradation of hydrogen peroxide to oxygen and water. The full-length catalase cDNA of Zhikong scallop Chlamys farreri (denoted as CfCAT) was identified from hemocytes by expressed sequence tag (EST) and rapid amplification of cDNA ends (RACE) approaches. The nucleotide sequence of CfCAT cDNA consisted of 3146bp with a 5' UTR of 103bp, an unusually long 3' UTR of 1519bp with a canonical polyadenylation signal sequence AATAAA and a polyA tail, and an open reading frame (ORF) of 1521bp encoding a polypeptide of 507 amino acids with predicted molecular weight of 57.5kDa. The deduced amino acid sequence of CfCAT has significant homology to catalases from animals, plants and bacteria. Several highly conserved motifs including the proximal heme-ligand signature sequence RLFSYNDTH, the proximal active site signature FNRERIPERVVHAKGGGA, and the three catalytic amino acid residues of His(72), Asn(145) and Tyr(355) were identified in the deduced amino acid sequence of CfCAT. The CfCAT was demonstrated to be a peroxisomal glycoprotein with two potential glycosylation sites and a peroxisome targeting signal of ANL that was consistent with human, mouse and rat catalases. The time-course expression of CfCAT in hemocytes was measured by quantitative real-time PCR. The expression of CfCAT increased gradually and reached the highest point at 12h post-Vibrio infection, then recovered to the original level at 24h. All these results indicate that CfCAT, a constitutive and inducible protein, is a member of the catalase family and is involved in the process against ROS in scallop.

Catalase, which is one of the key enzymes of the cellular antioxidant defense system, prevents free hydroxyl radical formation by breaking down hydrogen peroxide into oxygen and water. Here, we show the cloning and characterization of a catalase gene in a coleopteran insect. This gene was isolated by searching the white-spotted flower chafer Protaetia brevitarsis cDNA library, and the gene itself encodes a protein of 505 amino acids in length, named PbCat. PbCat shows high similarities to the insect catalase genes known to date. The recombinant PbCat, which is expressed as a 56-kDa polypeptide in baculovirus-infected insect Sf9 cells, shows the highest activity at 30 degrees C and pH 7.0. Northern and Western blot analyses revealed the presence of PbCat in all tissues examined, showing its ubiquitous expression. P. brevitarsis larvae in which H(2)O(2) was overloaded, showed a marked up-regulation in PbCat expression. Moreover, P. brevitarsis larvae showed an apparent increase in PbCat expression even after a wounding through injection. These results indicate that PbCat is up-regulated after wounding and oxidative pressure induced by H(2)O(2), reflecting an important role of PbCat in H(2)O(2) scavenging.

The toxicity of Roundup, a glyphosate-based herbicide widely used in agriculture, was determined for the Neotropical fish Prochilodus lineatus. The 96 h-LC(50) of Roundup was 13.69 mg L(-1), indicating that this fish is more sensitive to Roundup than rainbow trout (Oncorhynchus mykiss) and Atlantic salmon (Salmo salar). These differences should be considered when establishing criteria for water quality and animal well-being in the Neotropical region. Short-term (6, 24 and 96 h) toxicity tests were then performed to evaluate the effects of sub-lethal concentrations of the herbicide (7.5 and 10 mg L(-1)) to P. lineatus. Roundup did not interfere with the maintenance of the ionic balance and there was no significant alteration in plasma cortisol levels in Roundup-exposed fish. However an increase in plasma glucose was noted in fish exposed to 10 mg L(-1) of the herbicide, indicating a typical stress response. Catalase liver activity also showed an increase in fish exposed to 10 mg L(-1) of the herbicide, suggesting the activation of antioxidant defenses after Roundup exposure. In addition, Roundup induced several liver histological alterations that might impair normal organ functioning. Therefore, short-term exposure to Roundup at subletal concentrations induced biochemical, physiological and histological alterations in P. lineatus.

Catalase (CAT) dismutates the reactive oxygen species H2O2 into water and dioxygen and in plants; it is located in peroxisomes and glyoxysomes. In the present study, we investigated the effect of cadmium (a well-known oxidative stress inducer) on catalase in roots and cotyledons of developing sunflower seedlings, at 10 microM and 100 microM. Although germination was unaltered after 48 h of exposure to 100 microM Cd2+, root length was significantly reduced. CAT activity was also significantly reduced, but this activity was completely restored (10 microM treatment) or even enhanced (100 microM treatment) 24 h later. Although CAT protein abundance remained similar to control in roots and cotyledons of Cd-treated seedlings, cadmium produced CAT protein oxidation, indicating that the mechanism of CAT inactivation by Cd2+ involves oxidation of the protein structure. The transcripts of the four genes described for sunflower (CATA1 to CATA4) increased after cadmium treatment; CATA1 and CATA2 were the most overexpressed in cotyledon and root, respectively. The differential expression of catalase genes in sunflower seedlings under Cd stress might be related to the synthesis of CAT isoforms less sensitive to oxidation, which would prevent enzyme inactivation and H2O2 accumulation.
