In guinea-pigs given 400 micrograms per kg of tunicamycin, the toxin responsible for the disease known as annual ryegrass toxicity, alterations in blood-brain barrier permeability were assessed with vascular tracers of differing particle size. The toxin caused loss of integrity of the barrier to the smallest diameter tracer, horseradish peroxidase, but the cerebral endothelium was able to retain larger particles of ferritin and colloidal gold in the circulation. The effect of tunicamycin on cultured endothelial cells was characterized by dose-dependent lethal damage to these cells, marked dilatation of cisternae of RER, severe cytoplasmic vacuolation and a reduced incidence of mitosis in surviving endothelial cells. It was concluded that tunicamycin is able to damage directly endothelial cells, with a resultant increase in cerebral vascular permeability.

Three insect tissues have particular roles as filters to maintain the fluid composition of the hemolymph. Water and ions enter and leave through the midgut. The pericardial cells filter circulating hemolymph. Malpighian tubules, often with the rectum, allow resorption from a hemolymph filtrate that passes to the hindgut. All three tissues have plasma membrane infolds making a reticulum on their hemolymph surfaces, and all three have RER leading to SER extensions into their reticula. SER is a catch-all description for membranes lacking ribosomes in the pre-Golgi complex set of compartments of the vacuolar system. Some kinds of SER are well known for their role in housing enzymes for steroid metabolism and for detoxification. The SER ramifying within the plasma membrane reticular systems of tissues concerned with hemolymph filtration contains ferritin, suggesting that this SER has another, different function. In contrast to vertebrate cells, where ferritin is confined to the cytosol and lysosomes, we have found that in Calpodes and perhaps in most insects, ferritin occurs in the vacuolar system and not in the cytosol. Ferritin occurs naturally in the RER and SER of cells at the hind end of the midgut, in pericardial cells and in the yellow region of the Malpighian tubules. Additional ferritin is induced by loading the gut or hemolymph with iron. Overloading with iron causes ferritin secretion to the gut lumen. We propose that the SER in these cells functions in iron homeostasis by holding ferritin for loading and unloading as it moves to and from the reticulum at the cell surface where it can be maximally exposed to extracellular fluid flow.

The rat liver after extrahepatic biliary obstruction was studied by SEM and TEM in correlation with basic histochemical techniques. Cholestasis was verified by serological methods. The biochemical data (increase in serum bilirubin values, a gradual lowering of the albumin fraction), in agreement with the ultrastructural results of a sparse RER, suggested a gradual decrease of the protein synthetic activity of the hepatocyte. SEM and TEM revealed numerous fat-storing cells, closely associated with patches of connective fibrils in the subendothelial spaces. Further ultrastructural observations demonstrated: a) a proliferation of the intrahepatic biliary tree (ductular proliferation, including newly formed ducts with sacculation and diverticuli); b) an increased number of canaliculo-ductular junctions and, c) an increase in the length of the bile canalicular network due to its tortuous course, pocketing and side branching. The occurrence of an intact cytoplasmic barrier separating the bile canalicular lumen from the Disse's space together with the results obtained by retrograde infusion of ferritin into the biliary tree suggested that the regurgitation pathway by ductular reabsorption and by transhepatocytic transport is the best documented and most acceptable, at least in the rat.
