We have investigated the importance of several clinical and laboratory parameters on the development of acquired cystic kidney disease (ACKD) as detected by ultrasonography in 19 patients who had received dialysis therapy for at least three years. We were particularly interested on the possible effect of the serum levels of oxalate and silicon, which can produce tubular obstruction, and that of vanadium, which can affect cell proliferation. The severity of ACKD increased with the duration of dialysis and was greater in men than in women. Positive correlations were observed between the grades of ACKD and the levels of hemoglobin, hematocrit, and parathyroid hormone, while there was a negative correlation between ACKD and serum
ferritin
levels. The serum levels of oxalate, silicon, and vanadium, pre- and postdialysis, were markedly and significantly higher than those in normal controls, but there was no significant correlation between these levels and the duration of dialysis therapy or severity of ACKD. The pre- and postdialysis levels of vanadium were not significantly different, while the levels of oxalate and silicon were significantly lower in the postdialysis samples. No significant correlations were detected between ACKD and age of the patients, blood pressure, protein catabolic rate, efficiency of dialysis index, or the serum levels of iron,

In order to determine if there are biochemical changes in plasma-membrane oligosaccharides of regenerating retinal pigment epithelium, the binding of colloidal iron oxide at low pH and
ferritin
-conjugated wheat germ agglutinin--probes of sialic acid and N-acetylglucosamine on the cell surface--was examined electron-microscopically. An animal model of retinal pigment epithelium regeneration--rabbits with
sodium
iodate induced retinopathy--was used. In this model, large expanses of regenerating pigment epithelium are present for comparison with zones of spared pigment epithelium in the same animals. In thin sections examined by transmission electron microscopy,

-conjugated wheat germ agglutinin appeared to bind more intensely to the exposed plasma membrane of regenerating retinal pigment epithelium than to spared pigment epithelium, or that of normal rabbits. Morphometry verified this. Colloidal iron oxide intensely labelled the plasma membranes of regenerating, spared, and normal pigment epithelium, and was visibly reduced after exposure of tissue to neuraminidase. The observations indicate that the plasma membrane of regenerating retinal pigment epithelium bears sialic acid and N-acetylglucosamine residues as in normal retinal pigment epithelium. However, the amount of plasma membrane bearing exposed N-acetylglucosamine increases during regeneration.

resulted in the release of iron from

and the released iron-catalyzed lipid peroxidation. Hemoglobin, which binds NO, and superoxide anion, which reacts with NO, inhibited nitroprusside-dependent iron release from

, thereby providing evidence that NO can mobilize iron from

. These results suggest that NO generation in vivo could lead to the mobilization of iron from

disrupting intracellular iron homeostasis and increasing the level of reactive oxygen species.

Hydrolytic activities present in PR fluid were assayed with a number of peptide and carbohydrate substrates. Enzymes which hydrolyzed t-butyloxycarbonyl-Leu-Ser-Thr-Arg-methylcoumarylamide, t-butyloxycarbonyl-Phe-Ser-Arg-methylcoumarylamide, and t-butyloxycarbonyl-Val-Leu-Lys-methylcoumarylamide were found to be present in PR fluid at levels elevated by threefold or more over amounts found in a comparable volume of blood plasma.

The oxidation of acetaldehyde (generated from the metabolism of ethanol) by oxidases such as xanthine oxidase generates free radicals which can mobilize
ferritin
iron, alter hepatic glutathione and produce lipid peroxidation. The stomach, a site of ethanol metabolism and rich in xanthine oxidase, was studied with respect to the effects of ethanol on intrinsic factor (IF) binding of vitamin B-12 as well as gastric glutathione (GSH). Incubations of gastric homogenates with acetaldehyde-xanthine oxidase inhibited the B-12 binding ability by IF. A large acute dose of ethanol in vivo (5 g/kg, conc. greater than 40% w/v) decreased gastric IF binding of B-12 and depressed gastric GSH; these effects were markedly attenuated by the feeding of
sodium
tungstate which inhibited xanthine oxidase. Changes in B-12 binding paralleled changes in gastric GSH. Scatchard plots of IF binding of B-12 for homogenates suggested decreased number of binding sites rather than altered affinity. In conclusion, the gastric metabolism of ethanol generates free radicals which alter IF binding of B-12, depress gastric GSH and may play a role in alcohol-induced gastric injury.

The cell surface of Clostridium symbiosum HB25 is covered by a squarely arranged surface layer (S-layer) glycoprotein. Sodium dodecyl sulfate-polyacrylamide gel electrophoresis of the
sodium
dodecyl sulfate-soluble whole-cell extract showed the presence of several high-molecular-weight protein bands in a narrow range (approximate Mr, 140,000) which, upon periodic acid-Schiff staining, gave a positive reaction. After proteolytic degradation of the purified S-layer glycoprotein, a single glycopeptide fraction was obtained by gel permeation chromatography. Hydrolysis, treatment with aqueous hydrofluoric acid, and 1H and 13C nuclear magnetic resonance studies showed that the glycoprotein glycan is a high-molecular-weight polymer (approximate Mr, 15,000) of tetrasaccharide repeating units with the component sugars N-acetylgalactosamine (GalNAc), N-acetylmannosamine (ManNAc), and N-acetylbacillosamine (BacNAc; 2-N-acetyl-4-amino-2,4,6-trideoxy glucose) linked by monophosphate diesters. The following structure is proposed: [----6)-alpha-D-ManpNAc-(1----4)-beta-D-GalpNAc-(1----3)-alpha-D-+ ++BacpNAc- (1----4)-alpha-D-GalpNAc-(1----PO3)----]n. The nuclear magnetic resonance data provided evidence for a charge interaction between the free amino group of BacNAc and the phosphate group of adjacent glycan chains. Since polycationic

A comparative study of carrier-free 67Ga-citrate uptake by Ehrlich ascites tumor cells in the presence of lactoferrin, transferrin and
ferritin
has demonstrated that lactoferrin considerably increases the uptake of 67Ga, and that this increase seems to be determined by its iron-load. The other iron-binding proteins assayed have a null or negative effect. Their behavior in the presence of
sodium
citrate supports the concept of lactoferrin-binding by the cells as responsible for the uptake. The different behavior of 67Ga-citrate iron-binding protein complexes appears to support this hypothesis.

A 5-HT3 binding site, with high affinity for (S-)[3H]zacopride, was solubilized from rabbit small bowel muscularis membranes utilizing 0.5%
sodium
cholate and 400 mM (NH4)2SO4. Approximately 72% of the (S-)[3H]zacopride binding activity was recovered in a form that retained the high affinity (Kd = 0.7 nM) and specificity for this radioligand that is characteristic of the membrane-bound receptor. ICS 205-930 and other 5-HT3 compounds were effective inhibitors and exhibited the same rank order of potency in the solubilized and membrane-bound preparations. The receptor-detergent complex did not sediment after centrifugation for 1 h at 150,000 x g and eluted between thyroglobulin (MW = 669,000) and
apoferritin
(MW = 443,000) when fractionated by high-performance liquid chromatography gel filtration. This is the first report of the solubilization of a 5-HT3 binding site.

Mineral status of highly trained (HT) women runners (n = 14) and untrained (UT) women (n = 11), as determined from 3-day dietary records, blood, and urine samples, was compared. HT women had significantly higher energy (HT: 2331 +/- 121 vs UT: 1973 +/- 107 kcal/day; p less than 0.05), magnesium (Mg) (HT: 369 +/- 29 vs UT: 262 +/- 18 mg/day, p less than 0.05), and potassium (K) (HT: 3874 +/- 384 vs UT: 2646 +/- 244 mg/day; p less than 0.05) intakes than UT women. However, mean daily intakes of calcium (Ca), iron (Fe), copper (Cu), and
sodium
(Na) were not different for the two groups. Plasma concentrations and urinary excretion of Ca, Mg, and Cu of HT and UT women did not differ. Plasma Fe concentrations of the two groups were also similar (HT: 21 +/- 2 vs UT: 22 +/- 1 mumol/L), and, although serum
ferritin
concentrations of HT women were lower than UT women (HT: 18 +/- 4 vs UT: 30 +/- 6 micrograms/L), the difference was not significant. Urinary Na excretion of the two groups did not differ. HT women had a higher K intake than UT women but urinary K excretion of the two groups did not differ. Whether present mineral intakes by HT women are adequate during periods of intensive training remains to be determined.
